Genome‐wide identification of transcription factor binding sites with the ChIP‐seq method is an extremely important scientific endeavor − one that should ideally be performed for every transcription factor in as many cell types as possible. A major hurdle on the way to this goal is the necessity for a specific, ChIP‐grade antibody for each transcription factor of interest, which is often not available. Here, we describe CETCh‐seq, a recently published method utilizing genome engineering with the CRISPR/Cas9 system to circumvent the (...) need for a specific antibody. Using the CETCh‐seq method, targeted genomic editing results in an epitope‐tagged transcription factor, which is recognized by a well‐characterized, standard antibody, efficacious for ChIP‐seq. We have used CETCh‐seq in human cancer cell lines as well as mouse embryonic stem cells. We find that roughly 60% of transcription factors tagged using CETCh‐seq produce a high quality ChIP‐seq map, a significant improvement over traditional antibody‐based methods. (shrink)

Foucault famously claimed that in political theory the king’s head still needs to be cut off, proclaiming the imperative to move beyond a centralized and prohibitive conception of power and toward a more distributed, relational and productive understanding of power in political society. Ironically, Edmund Burke, famous for criticizing an actual revolutionary regicide in France, can be read as an ally in Foucault’s project of theoretical regicide and conceptual revolution. For although he staunchly defended existing monarchies in France and Britain, (...) Burke’s political theory in many ways encapsulates the kind of approach that Foucault sought to call forth. In this article, I argue for three particular resonances between Burke’s and Foucault’s views on power’s place and role in political society: one, power precedes subjects; two, power exceeds subjects; and three, subjects must negotiate with existing power relations (as opposed to ignoring or destroying them). Combined, these resonances remind us that political practice, reform and resistance are (almost) always situated in an unchosen context, thus happening in the middle of things rather than on a blank social canvas. Recognizing Burke’s interest in reform conditioned by the socio-cultural and historical circumstances and Foucault’s concern for resistance even though we can never be outside power allows us to appreciate the unchosen constraints that shape political practice of individual and collective subjects alike. ISBN: 9780198812739. $91.00.In his brief autobiography, “My Own Life,” Hume reports that “almost all [his] life has been spent in literary pursuits and occupations” (E-MOL: xxxi). This is one (...) of at least ten places in this brief work in which he associates himself with “letters” or “literature”—as compared to at most four associations with philosophy. The moral to be inferred is not that Hume considered himself not to be a philosopher. Rather, he conceived of philosophy as a kind of literature. The Treatise was itself a “literary attempt.” Indeed, opposition between the “literary” and “philosophical” aspects of a text would have been foreign to Hume and his contemporaries—a notion for a later age, despite his occasional loose distinction between the relevant genres, as in his list of writings “historical, philosophical, or literary” in “My Own Life” (E-MOL: xxxvi).Yet despite Hume’s own understanding of the continuity between philosophy and literature, broadly conceived, it is still common for contemporary philosophers to ignore literary questions in their examination of Hume’s work, or to presume that one can easily distinguish the “philosophical” parts from the “literary” ones. Tim Milnes’s The Testimony of Sense is therefore a welcome addition to the Hume literature (pun intended). With a particular focus on the essay—a genre of great importance to Hume—Milnes contributes to the small set of engagements with Hume from the disciplinary perspective of English literature. With training in both philosophy and literature, as well as established expertise on Romanticism and its own essays, Milnes is well-qualified for the task. The Testimony of Sense includes sustained engagement with Hume’s contemporaries and successors, but for the purposes of this review, I engage the book primarily as a Hume scholar for Hume scholars.Milnes’s story plays out in three acts. The first establishes the need for public trust created by a skeptical crisis, itself a product of Hume’s criticism of the epistemic program of thinkers in the Cartesian and Lockean traditions. The second explains how the essays of the long eighteenth century constitute a response to that need in various ways. The third reveals in detail how the Romantic essayists, particularly William Hazlitt and Charles Lamb, both respond to and deviate from the “neoclassical” essayists, typified by Hume and Samuel Johnson.The first chapter, “Self and Intersubjectivity,” lays the groundwork by articulating how Hume, Adam Smith, Thomas Reid, and Dugald Stewart identify the need for what we now call intersubjectivity. Milnes describes the common thread as a kind of “natural transcendentalism.” In what sense are these thinkers properly understood as [End Page 175] transcendentalists? The central idea seems to be that all posit things other than reason as “conditions of meaningful thought” (72). For Hume, it is the social, linguistic experience of a community of virtuous judges. The intricate relations between self and other essential to such a community get “internalized within the specular drama of the impartial spectator” (72) in Smith’s writing, which itself takes part in the drama through his “halting, qualified syntax” that mimics “everyday speech” (44). Reid posits a “prescience” of trust, which grounds our reliance on both human testimony and the uniformity of nature, itself undergirded by the design of a benevolent God. Stewart, who posits axiomatic truths that help constitute the “stamina of intellect,” goes furthest, actually calling these truths transcendental (55). This chapter also puts these eighteenth-century thinkers in conversation with twentieth-century theorists, including Habermas, Davidson, and Rorty.Chapter 2, “The Subject of Trust,” deepens the analysis by situating these “natural transcendentalisms” in the broader enlightenment context. In contrast to Descartes, Bacon, and Locke, who were suspicious of our reliance on the opinions of others, the eighteenth-century thinkers understand trust as prior to reason in various ways—though not prior to solidarity. Even though Kant’s theory of cognition (Erkenntnis) is central to his Critique of Pure Reason, it has rarely been asked what exactly Kant means by the term “cognition”. Against the widespread assumption that cognition (in the most relevant sense of that term) can be identified with knowledge or if not, that knowledge is at least a species of cognition, we argue that the concepts of cognition and knowledge in Kant are not only distinct, but even disjunct. To show this, we (...) first (I) investigate Kant’s explicit characterizations of the nature of cognition. As it turns out, he introduces several different notions that must be carefully distinguished before identifying the one that is central to his project in the first Critique. We then (II) consider the basic features of Kant’s conception of knowledge, indicating both how it involves assent and objective justification and how it relates to our contemporary conception. Next (III), we compare and contrast Kant’s understanding of cognition and his conception of knowledge in a way that allows us to present their fundamental differences and connections. We argue that while cognition, in the most relevant sense, is a species of representation that differs from other representations in that it involves the conceptual determination of a sensibly given object, knowledge (for Kant) is a kind of assent to a judgment that requires consciousness of a sufficient epistemic ground. Finally (IV), by appreciating the differences between cognition and knowledge, we explain several of the implications this conception of cognition has for some of Kant’s main claims in the Critique of Pure Reason as a whole. Among other things, we show how Kant can deny cognition of specific things in themselves while allowing philosophical knowledge about things in themselves in general (e.g. that they exist, are not in space and time, etc.). This paper attempts to shed light on Kant’s notion of autonomy in his moral philosophy by considering the extent to which he presents a similar doctrine in his theoretical philosophy, where he strikingly claims (e.g., in the Prolegomena) that the understanding prescribes laws to nature. It argues that even though there are important points of difference between the cases of theoretical legislation of the laws of nature and autonomy in moral philosophy, their extensive parallels make a strong, even if not (...) definitive case in favor of the Prolegomena forming at least an important part of the explanation of the emergence of autonomy in the Groundwork. In addition, the comparison proves instructive insofar as it reveals deep structural parallels between Kant’s theoretical and practical philosophy regarding his conception of laws. (shrink)

Using past episodes of climate change as a source of evidence to inform our projections about contemporary climate change requires establishing the extent to which episodes in the deep past are analogous to the current crisis. However, many scientists claim that contemporary rates of climate change (e.g., rates of carbon emissions or temperature change) are unprecedented, including compared to episodes in the deep past. If so, this would limit the utility of paleoclimate analogues. In this paper, I show how a (...) data adjustment procedure called “temporal scaling,” which must be applied to both contemporary and past rate data, complicates the claim that contemporary rates are truly unprecedented. On top of giving actionable recommendations to scientists, this paper advances the philosophical literature concerning the use of models that are known to be somewhat disanalogous to their target systems. The purpose of this research is to examine the impact of individual and firm moral philosophies on marketing exchange relationships. Personal moral philosophies range from the extreme forms of true altruists and true egoists, along with three hybrids that represent middle ground (i.e., realistic altruists, tit-for-tats, and realistic egoists). Organizational postures are defined as Ethical Paradigm, Unethical Paradigm, and Neutral Paradigm, which result in changes to personal moral philosophies and company and industry performance. The study context is a simulation of (...) an exchange environment using a variation of the prisoners' dilemma game. In the first part of his book Klemme considers Kant’s views [End Page 471] in the 1770s as presented in his Inaugural Dissertation, newly available transcripts from his anthropology lectures, the so-called Duisburg Nachlass, and transcripts from his metaphysics lectures. Klemme first (Chapter 1) indicates briefly the place of psychology and anthropology in metaphysics in eighteenth-century Germany and identifies two general types of processes through which it was thought that self-knowledge might be attained. He then (Chapter 2) turns to a discussion of Kant’s project in the early 1770s—basing his philosophy on a spontaneous, free, and self-conscious I—and how it is developed against the background of (i) Kant’s “great light” of 1769, (ii) the awakening out of his “dogmatic slumber,” and (iii) the discovery of antinomial conflict. Klemme’s original thesis is that Moses Mendelssohn’s Phädon (1767) and especially Marcus Herz’s Betrachtungen aus der spekulativen Weltweisheit (1771), as well as Kant’s personal correspondence with these figures, are crucial to understanding how Kant comes to see the deficiencies of his view as presented in the Inaugural Dissertation. Klemme discusses Kant’s “remedy” (in Chapter 3): a new theory of the self, documented in the anthropology transcripts, according to which one has direct intuition of oneself as a simple, spontaneous, immaterial substance. Later in the 1770s (especially as documented in the Duisburg Nachlass) Kant finds himself unable to derive the categories from the self in either rational or empirical psychology and is thus forced to revise his theory in his first Critique.In the second part of his book Klemme discusses Kant’s account of self-consciousness in his Critical period. Accordingly, he presents interpretations of the Transcendental Deduction and the Paralogisms in both editions of the first Critique as well as some of the central theses of Transcendental Idealism. Fundamental to Kant’s theory of self-consciousness in the Critical period is a distinction between self-knowledge and self-awareness. The former is knowledge of the empirical self, whereas the latter is knowledge of the self considered “not as appearance nor as thing in itself” (B423) but rather pre-categorically as an indeterminate existence. With respect to self-knowledge, Klemme argues that Kant adheres to three distinct models from 1781 to 1787. In the first edition of the first Critique Kant endorses the analogy model, according to which the regulative use of reason allows one to connect all of the self’s mental states in inner sense “as if it were a simple substance” (A 672/B 700). By the time of the second edition he adopts the mediation model, according to which “the categories can be applied to inner sense because we can represent time to ourselves only through recourse to outer sense as a line” (240). In the transcripts from his anthropology lectures (as well as in his published Anthropology) Kant seems to adopt a third model of self-knowledge, according to which the categories cannot be applied to inner representations at all; such knowledge amounts to nothing more than a mere history of our representations.Klemme concludes his discussion by considering Kant’s account of self-awareness. Kant is often thought (e.g., by Dieter Henrich) to hold a reflection model, according to which the self literally reflects on itself in self-awareness, giving rise to problems of circularity. Klemme argues against such an interpretation (as well as Manfred Frank’s “intellectual intuition” interpretation) in favor of a view, according to which the self can be determined practically “as a mere intelligence according to the moral law” (395). The career of the American photographer Carleton E. Watkins spanned more than fifty years. It is his giant photographs of Yosemite, from the "best general view," that most effectively articulate his artistic vision. The J. Paul Getty Museum holds more than fourteen hundred pictures by Watkins, making him the best-represented nineteenth-century photographer in the collection. In Focus: Carleton Watkins features approximately fifty of these works, including mammoth plates, stereographs, albumen prints, and cabinet and boudoir cards. The plates (...) are accompanied by commentaries written by Peter E. Palmquist, an independent scholar of the history of photography. Mr. Palmquist, along with David Featherstone, Tom Fels, Weston Naef, David Robertson, and Amy Rule, were participants in a 1996 colloquium on Watkins and his career. Arguments from perceptual variation challenge the view that colors are objective properties of objects, properties that objects have independent of how they are perceived. This paper attempts, first, to diagnose one central reason why arguments from perceptual variation seem especially challenging for objectivists about color. Second, we offer a response to this challenge, claiming that once we focus on determinate colors rather than the determinables they determine, a response to arguments from perceptual variation becomes apparent. Third, our nominal opponents are (...) relationalist (like Cohen in The red and the real: an essay on color ontology, Oxford University Press, Oxford, 2009) and we will argue that the main argument for rejecting objectivism commits the relationalist to a position that is more radical than the one he would wish to endorse. Fourth, we suggest that insight into which properties could be relational may be found by looking to our best scientific theories. (shrink)

Does it make sense to refer to bird song - a complex vocalization, full of repetitive and transformative patterns that are carefully calculated to woo a mate - as art? What about a pack of wolves howling in unison or the cacophony made by an entire rain forest? Redefining music as "the art of possibly animate things," Musical Vitalities charts a new path for music studies that blends musicological methods with perspectives drawn from the life sciences. In opposition to humanist (...) approaches that insist on a separation between culture and nature--approaches that appear increasingly untenable in an era defined by human-generated climate change--Musical Vitalities treats music as one example of the cultural practices and biotic arts of the animal kingdom rather than as a phenomenon categorically distinct from nonhuman forms of sonic expression. The book challenges the human exceptionalism that has allowed musicologists to overlook music's structural resemblances to the songs of nonhuman species, the intricacies of music's physiological impact on listeners, and the many analogues between music's formal processes and those of the dynamic natural world. Through close readings of Austro-German music and aesthetic writings that suggest wide-ranging analogies between music and nature, Musical Vitalities seeks to both rekindle the critical potential of nineteenth-century music and rejoin the humans at the center of the humanities with the nonhumans whose evolutionary endowments and planetary fates they share.
