We highlight the various domains of the flavivirus virulence factor NS1 and speculate on potential implications of the NS1 3D structure in understanding its role in flavivirus pathogenesis. Flavivirus non‐structural protein 1 (NS1) is a virulence factor with dual functions in genome replication and immune evasion. Crystal structures of NS1, combined with reconstructions from electron microscopy (EM), provide insight into the architecture of dimeric NS1 on cell membranes and the assembly of a secreted hexameric NS1‐lipid complex found in patient sera. (...) Three structural domains of NS1 likely have distinct roles in membrane association, replication complex assembly, and immune system avoidance. A conserved hydrophobic inner face is sequestered either on the membrane or in the interior of the secreted hexamer and contains regions implicated in viral replication. The exposed variable outer face is presented to cellular and secreted components of the immune system in infected patients and contains candidate regions for immune system modulation. We anticipate that knowledge of the distinct NS1 domains and assembly will lead to advances in elucidating virus‐host interactions mediated through NS1 and in dissecting the role of NS1 in viral genome replication. Richard M Weaver, a thinker and writer celebrated for his unsparing diagnoses and realistic remedies for the ills of our age, is known largely through a few of his works that remain in print. This new collection of Weaver's shorter writings, assembled by Ted J Smith III, Weaver's leading biographer, presents many long-out-of-print and never-before-published works that give new range and depth to Weaver's sweeping thought. 1 Evidence-based policy requires researchers to provide the answers to ecological questions that are of interest to policy makers. To find out what those questions are in the UK, representatives from 28 organizations involved in policy, together with scientists from 10 academic institutions, were asked to generate a list of questions from their organizations. 2 During a 2-day workshop the initial list of 1003 questions generated from consulting at least 654 policy makers and academics was used as a basis for (...) generating a short list of 100 questions of significant policy relevance. Short-listing was decided on the basis of the preferences of the representatives from the policy-led organizations. 3 The areas covered included most major issues of environmental concern in the UK, including agriculture, marine fisheries, climate change, ecosystem function and land management. 4 The most striking outcome was the preference for general questions rather than narrow ones. The reason is that policy is driven by broad issues rather than specific ones. In contrast, scientists are frequently best equipped to answer specific questions. This means that it may be necessary to extract the underpinning specific question before researchers can proceed. 5 Synthesis and applications. Greater communication between policy makers and scientists is required in order to ensure that applied ecologists are dealing with issues in a way that can feed into policy. It is particularly important that applied ecologists emphasize the generic value of their work wherever possible. (shrink)

This paper situates Hume's views on animals in the context of the Scottish Enlightenment by contrasting them with the views of Adam Smith. While Smith is more central to the philosophical establishment of the Scottish Enlightenment, their views on morals resemble each other greatly and both think that the analogies between humans and non-human animals are useful for thinking about morals. Their estimation of the nature and extent of those analogies, however, differ widely from one another. This has (...) been historically obscured by the fact that in no single work does Smith precisely detail what he thinks non-human animals are capable of. I argue that Smith thinks non-human animal minds are different in kind from human minds. This is evident from Smith's view of how language facilitates and co-creates certain aspects of human cognition. Hume, by contrast, seems to hold that non-human animal minds differ merely by degree from human minds. After reconstructing Smith's view, I contrast it with Hume's, providing historical context to show how Hume falls outside the mainstream on this issue and Smith within it. Their views on animals reflect, broadly, their standing with respect to the wider Scottish philosophical community. (shrink)

In reading biographies or accounts of figures with which one agrees and sympathizes, there is a tendency that needs to be avoided, that is, of over -identifying with the figures in question and of too closely mapping one's own life and aspirations onto them.As such, there is some risk involved for a person like me in reading about the friendship between David Hume and Adam Smith. Dennis C. Rasmussen's excellent new volume, The Infidel and the Professor: David Hume, Adam (...) Smith, and the Friendship that Shaped Modern Thought, chronicles that friendship in an engaging and compelling narrative. The work is thoroughly researched and entertainingly written. Experience and Knowledge. "Experience" for Dewey is without doubt the most fundamental and pervasive concept of his philosophy. One may even characterize his entire philosophic endeavor as an attempt to reconstruct the philosophic use of "experience" in order to bring it into closer contact with the multifarious concrete experiences of men, and to escape the artificial and fruitless disputes of epistemologists. By analyzing five contrasts with what Dewey sometimes called "the traditional concept of experience," Professor Smith has conveyed succinctly (...) what this reconstruction involves. Experience consists of transactions of organism-environment which are more inclusive than knowing situations; experience is not primarily subjective, subject and object are distinctions instituted within experience; experience is active and not restricted to the specious present, it involves projection into the future; it does not consist of an aggregate of independent and disconnected elements, it contains genuine connections within itself; and finally, experience is not opaque to reason, it can become funded with intelligence. The value of any kind of data is greatly enhanced when it exists in a form that allows it to be integrated with other data. One approach to integration is through the annotation of multiple bodies of data using common controlled vocabularies or ‘ontologies’. Unfortunately, the very success of this approach has led to a proliferation of ontologies which itself creates obstacles to integration. The Open Biomedical Ontologies (OBO) consortium has set in train a strategy to overcome this problem. Existing (...) OBO ontologies, including the Gene Ontology, are undergoing a process of coordinated reform and new ontologies being created on the basis of an evolving set of shared principles governing ontology development. The result is an expanding family of ontologies designed to be interoperable, logically well-formed, and to incorporate accurate representations of biological reality. We describe the OBO Foundry initiative, and provide guidelines for those who might wish to become involved. (shrink)

The Cell Ontology (CL) is designed to provide a standardized representation of cell types for data annotation. Currently, the CL employs multiple is_a relations, defining cell types in terms of histological, functional, and lineage properties, and the majority of definitions are written with sufficient generality to hold across multiple species. This approach limits the CL’s utility for cross-species data integration. To address this problem, we developed a method for the ontological representation of cells and applied this method to develop a (...) dendritic cell ontology (DC-CL). DC-CL subtypes are delineated on the basis of surface protein expression, systematically including both species-general and species-specific types and optimizing DC-CL for the analysis of flow cytometry data. This approach brings benefits in the form of increased accuracy, support for reasoning, and interoperability with other ontology resources. 104. Rather, we have a perceiving, acting organism, whose perceptions and actions are always already tuned to the parts and moments, the things and surfaces, of its external environment. Recently there have been intensive debates concerning the new defenses of liberalism and those labeled the "communitarian" critics of liberalism. The latter argue that the conceptions of the self and human agency presupposed by defenders of liberalism are deficient. For a liberal conception of the self fails to do justice to the social-historical context in which the modern individual has emerged. Liberalism neglects the communal-historical context of political activity. The terms of this debate have reached a stage where such concepts (...) as community and communtarian are becoming increasingly vague and ambiguous. What is badly needed is a rethinking of the substantive issues that underlie these debates. In this respect, one can welcome a revival of interest in Hegel's political philosophy, for many of the issues presupposed by these current debates were thought out more carefully and thoroughly by Hegel. Smith's lucid and thoughtful book is especially timely. Smith seeks to show that the standard conception of Hegel as an antiliberal thinker is too superficial. Hegel appreciated the contributions of a liberal theory of rights and sought to expose the precise points of weakness of liberalism. Rather than being "against" liberalism, Hegel sought to incorporate the "truth" implicit in a liberal theory of rights into a more comprehensive and adequate political theory. Smith begins by adumbrating the origins of the Hegelian project. He then carefully reviews Hegel's immanent critique of the liberal theory of rights and shows how this sets the stage for Hegel's own theory of rights--a theory that gives prominence to the "right of recognition." This culminates in the Hegelian Rechtstaat, which Smith translates as "the rule of law." He completes his study by delineating Hegel's own distinctive idea of Critical Theory and what the controversial claim about the "end of history" really means. (shrink)

Childhood obesity is in important respects a result of legal policies that influence both dietary intake and physical activity. The law must shift focus away from individual risk factors alone and seek instead to promote situational and environmental influences that create an atmosphere conducive to health. To attain this goal, advocates should embrace a population-wide model of public health, and policymakers must critically examine the fashionable rhetoric of consumer choice.

This article examines the extent to which diversity characteristics and stakeholder role influence individuals’ corporate social orientation (CSO). Our findings indicate that one’s relationship to the organization as well as diversity, gender, and race influence one’s CSO. Specifically, we found that employees’ greatest concern was economic whereas customers had a stronger ethical orientation. The results also suggest that women as well as Black employees and customers place more emphasis on whether an organization is fulfilling its discretionary responsibilities than do males (...) and Whites. A model using the concept of value congruence is presented to guide future research. (shrink)

We present an ontology of pain and of other pain-related phenomena, building on the definition of pain provided by the International Association for the Study of Pain (IASP). Our strategy is to identify an evolutionarily basic canonical pain phenomenon, involving unpleasant sensory and emotional experience based causally in localized tissue damage that is concordant with that experience. We then show how different variant cases of this canonical pain phenomenon can be distinguished, including pain that is elevated relative to peripheral trauma, (...) pain that is caused neuropathically (thus with no necessary peripheral stimulus), and pain reports arising through deception either of self or of others. We describe how our approach can answer some of the objections raised against the IASP definition, and sketch how it can be used to support more sophisticated discrimination of different types of pain resulting in improved data analysis that can help in advancing pain research. The Ontology for Biomedical Investigations (OBI) is an ontology that provides terms with precisely defined meanings to describe all aspects of how investigations in the biological and medical domains are conducted. OBI re-uses ontologies that provide a representation of biomedical knowledge from the Open Biological and Biomedical Ontologies (OBO) project and adds the ability to describe how this knowledge was derived. We here describe the state of OBI and several applications that are using it, such as adding semantic expressivity to (...) existing databases, building data entry forms, and enabling interoperability between knowledge resources. OBI covers all phases of the investigation process, such as planning, execution and reporting. It represents information and material entities that participate in these processes, as well as roles and functions. Prior to OBI, it was not possible to use a single internally consistent resource that could be applied to multiple types of experiments for these applications. OBI has made this possible by creating terms for entities involved in biological and medical investigations and by importing parts of other biomedical ontologies such as GO, Chemical Entities of Biological Interest (ChEBI) and Phenotype Attribute and Trait Ontology (PATO) without altering their meaning. OBI is being used in a wide range of projects covering genomics, multi-omics, immunology, and catalogs of services. OBI has also spawned other ontologies (Information Artifact Ontology) and methods for importing parts of ontologies (Minimum information to reference an external ontology term (MIREOT)). The OBI project is an open cross-disciplinary collaborative effort, encompassing multiple research communities from around the globe. To date, OBI has created 2366 classes and 40 relations along with textual and formal definitions. The OBI Consortium maintains a web resource providing details on the people, policies, and issues being addressed in association with OBI. (shrink)

> Wealthy nations must do much more, much faster. The United Nations General Assembly in September 2021 will bring countries together at a critical time for marshalling collective action to tackle the global environmental crisis. They will meet again at the biodiversity summit in Kunming, China, and the climate conference 26) in Glasgow, UK. Ahead of these pivotal meetings, we—the editors of health journals worldwide—call for urgent action to keep average global temperature increases below 1.5°C, halt the destruction of nature (...) and protect health. Health is already being harmed by global temperature increases and the destruction of the natural world, a state of affairs health professionals have been bringing attention to for decades.1 The science is unequivocal; a global increase of 1.5°C above the preindustrial average and the continued loss of biodiversity risk catastrophic harm to health that will be impossible to reverse.2 3 Despite the world’s necessary preoccupation with COVID-19, we cannot wait for the pandemic to pass to rapidly reduce emissions. Reflecting the severity of the moment, this editorial appears in health journals across the world. We are united in recognising that only fundamental and equitable changes to societies will reverse our current trajectory. The risks to health of increases above 1.5°C are now well established.2 Indeed, no temperature rise is ‘safe’. In the past 20 years, heat-related mortality among people aged over 65 has increased by more than 50%.4 Higher temperatures have brought increased dehydration and renal function loss, dermatological malignancies, tropical infections, adverse mental health outcomes, pregnancy complications, allergies, and cardiovascular and pulmonary morbidity and mortality.5 6 Harms disproportionately affect the most vulnerable, including children, older populations, ethnic minorities, poorer communities and those with underlying health problems.2 4 Global heating is also contributing to the decline in …. This volume examines modern scepticism in all main philosophical areas: epistemology, science, metaphysics, morals, and religion. It features sixteen essays that explore its importance for modern thought. The contributions present diverse, mutually enriching interpretations of key thinkers, from Montaigne to Nietzsche. The book includes a look both at the relationship between Montaigne and Pascal and at Montaigne’s criticism of religious rationalism. It re-establishes that necessary dialogue between researchers of scepticism from all over the Americas, which began with Popkin, Oswaldo Porchat and Ezequiel de Olaso long ago. The question 'Why should I obey the law?' introduces a contemporary puzzle that is as old as philosophy itself. The puzzle is especially troublesome if we think of cases in which breaking the law is not otherwise wrongful, and in which the chances of getting caught are negligible. Philosophers from Socrates to H.L.A. Hart have struggled to give reasoned support to the idea that we do have a general moral duty to obey the law but, more recently, the greater number (...) of learned voices has expressed doubt that there is any such duty, at least as traditionally conceived. Pancreatitis is a complex, progressively destructive inflammatory disorder. Alcohol was long thought to be the primary causative agent, but genetic contributions have been of interest since the discovery that rare PRSS1, CFTR and SPINK1 variants were associated with pancreatitis risk. We now report two associations at genome-wide significance identified and replicated at PRSS1-PRSS2 and X-linked CLDN2 through a two-stage genome-wide study. The PRSS1 variant likely affects disease susceptibility by altering expression of the primary trypsinogen gene. The CLDN2 risk allele is (...) associated with atypical localization of claudin-2 in pancreatic acinar cells. The homozygous CLDN2 genotype confers the greatest risk, and its alleles interact with alcohol consumption to amplify risk. Quentin Smith has argued that it is logically impossible for there to be a divine cause of the universe. His argument is based on a Humean analysis of causation (confined to event causation, specifically excluding any consideration of agency) and a principle drawn from that analysis that he takes to be a logical requirement for every possibly valid theory of causation. He also thinks that all divine volitions are efficacious of logical necessity.
