We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. In our paper we will try to connect the dynamics of community decline to individual responses. We will operate on two levels of reality. At the first level we will discuss the circumstances surrounding the recent decline of small communities in North Dakota. At the second level we will discuss how this decline affects small town residents' attitudes toward economic development. In the first level analysis we examine the thesis that the natural environment of community growth is economic exploitation; therefore, (...) the decline of resource-based communities is natural and inevitable. We discuss the circumstances surrounding the recent decline of small communities in North Dakota. At the second level analysis, we operate at the level at which life circumstances and environments are shaped by political-economic institutions that in turn shape the behavior and mentality of communities, families, and individuals. We hypothesize that residents of declining small towns deny their powerlessness and adapt to their situation through intense dedication to economic growth. We develop a series of hypotheses to test this thesis using data from the North Dakota Rural Life Poll. In our conclusion, we discuss implications for rural development practitioners working in declining small towns where this intense dedication to economic development in prevalent. (shrink)

Part novella, part philosophy, Clara was Schelling's most popular work during his lifetime, and appears here in English for the first time.

Schelling's 1806 polemic against Fichte, and his last major work on the philosophy of nature. The heat of anger can concentrate the mind. Convinced that he had been betrayed by his former collaborator and colleague, Schelling attempts in this polemic to reach a final reckoning with Fichte. Employing the format of a book review, Schelling directs withering scorn at three of Fichte’s recent publications, at one point likening them to the hell, purgatory, and would-be paradise of Fichtean philosophy. The central (...) bone of contention is the understanding of nature: Fichte sees it as lifeless matter in motion, sheer opposition to be overcome, while Schelling waxes poetic in his defense of a living, organic nature of which humanity is a vital part. Indeed, we do not know ourselves without understanding our connection to nature, argues Schelling, anticipating many thinkers in contemporary environmental ethics. Dale E. Snow’s introduction sets the stage and explains the larger context of the conflict, which was already visible in the correspondence of the two philosophers, which broke off by 1802. When a single beam-splitter receives two beams of bosons described by Fock states (Bose-Einstein condensates at very low temperatures), interesting generalizations of the two-photon Hong-Ou-Mandel effect take place for larger number of particles. The distributions of particles at two detectors behind the beam splitter can be understood as resulting from the combination of two effects, the spontaneous phase appearing during quantum measurement, and the quantum angle. The latter introduces quantum “population oscillations”, which can be seen as a generalized Hong-Ou-Mandel effect, (...) although they do not always correspond to even-odd oscillations. (shrink)

In 1980, Jacques Derrida published La carte postale: De Socrate a Freud et au-del a. At the occasion of the 30th anniversary of the English translation, Going Postcard: The Letter(s) of Jacques Derrida revisits this seminal work in Derrida's oeuvre. Derrida himself described The Post Card in his preface as "the remainders of a destroyed correspondence," stretching from 1977 to 1979. A cryptic text, it is riddled with gaps, word plays, and a meandering analysis of the interface between philosophy and (...) psychoanalysis. The contributors who offered the fourteen essays gathered in Going Postcard were each provided with a deceptively simple task: to write a gloss to a fragment from the first part of The Post Card, "Envois." The result is a prismatic array of commentaries, excursions, and interpretations that take Derrida "to the letter." The different glosses on lemmas such as genre, erasure, telepathy, philately, and sperm transport The Post Card into the twenty-first century and offer a "correspondence," if fragmentary, with Derrida's work and the work to come. There exist important deductive systems, such as the non-normal modal logics, that are not proper subjects of classical algebraic logic in the sense that their metatheory cannot be reduced to the equational metatheory of any particular class of algebras. Nevertheless, most of these systems are amenable to the methods of universal algebra when applied to the matrix models of the system. In the present paper we consider a wide class of deductive systems of this kind called protoalgebraic logics. These include (...) almost all (non-pathological) systems of prepositional logic that have occurred in the literature. The relationship between the metatheory of a protoalgebraic logic and its matrix models is studied. The following results are obtained for any finite matrix model U of a filter-distributive protoalgebraic logic : (I) The extension U of is finitely axiomatized (provided has only finitely many inference rules); (II) U has only finitely many extensions. This article proposes an action guide to making decisions regarding the ethical allocation of resources that affect access to healthcare services offered by community-based healthcare organizations. Using the filter of empirical data from a study of decision making in two community-based healthcare organizations, we identify potentially relevant conceptual guidance from a review of frameworks and action guides in the public health, health policy, and organizational ethics literature. We describe the development of this action guide. We used data from a prior (...) empirical study of the values that influence decision making about the allocation of resources in particular types of community-based healthcare organizations. We evaluated, organized, and specified the conceptual guidance we found in 14 frameworks for ethical decision making. The result is an action guide that includes four domains that are relevant to the context of the decision to be made, eight domains that are relevant to the process of the decision to be made, and 15 domains that are relevant to the criteria of the decision to be made. We demonstrate the potential use of this action guide by walking through an illustrative resource allocation decision. The action guide provides community-based healthcare organizations with a conceptually grounded, empirically informed framework for ethical decision making. (shrink)

This paper is based on Lectures 1, 2 and 4 in the series of ten lectures titled “Algebraic Structures for Logic” that Professor Blok and I presented at the Twenty Third Holiday Mathematics Symposium held at New Mexico State University in Las Cruces, New Mexico, January 8-12, 1999. These three lectures presented a new approach to the algebraization of deductive systems, and after the symposium we made plans to publish a joint paper, to be written by Blok, further developing these (...) ideas. That project was still incomplete when Blok died. In fact, there is no indication that he had prepared a draft of the paper, and we do not know what new material he intended to include. I am therefore not in a position to complete the project as he had envisioned it. So, I have settled for the more limited objective of presenting the material from the three lectures, leaving to others the task of adapting the techniques used there to more general situations. W. J. Mander provides a brief introduction to and critical assessment of the thought of the greatest of the British Idealist philosophers, F. H. Bradley (1846-1924), whose work has been largely neglected in this century. After a general introduction to Bradley's metaphysics and its logical foundations, Mander shows that much of Bradley's philosophy has been seriously misunderstood. Mander argues that any adequate treatment of Bradley's thought must take full account of his unique dual inheritance from the traditions of British empiricism (...) and Hegelian rationalism. In this paper I argue that Wittgenstein is correct when he says of the Standard Metre stick that we can neither say that it is or is not a metre in length – despite what our intuitions may tell us to the contrary. Specifically, the paper deals with Kripke's criticism of Wittgenstein's claim in Naming And Necessity and with Salmon's attempt to arbitrate between the two views. I conclude that, not only is Wittgenstein correct, but that both Kripke and Salmon (...) (and possibly the majority of philosophers) simply do not understand the concept of measurement. (shrink)

This paper argues that a strong thesis of divine providence, whereby God is understood as in complete control of all things, entails pantheism, the thesis that the universe is not ontologically distinct from God. In normal discourse, we distinguish a plan from, on the one hand, the state of affairs which realizes that plan—its execution or expression—and, on the other hand, the person or group whose plan it is. However, with respect to an omnipotent God who displays complete providence, neither (...) of these two distinctions holds. We cannot separate person from plan, or plan from world, and in consequence, neither can we separate person from world, ergo pantheism. Accordingly, the argument of this paper comes in two parts maintaining, first, the impossibility of distinguishing between God’s plan and its execution and, second, the impossibility of distinguishing God himself from his plan. Modal logics are studied in their algebraic disguise of varieties of so-called modal algebras. This enables us to apply strong results of a universal algebraic nature, notably those obtained by B. Jonsson. It is shown that the degree of incompleteness with respect to Kripke semantics of any modal logic containing the axiom □ p → p or containing an axiom of the form $\square^mp \leftrightarrow\square^{m + 1}p$ for some natural number m is 2 ℵ 0 . Furthermore, we show that (...) there exists an immediate predecessor of classical logic (axiomatized by $p \leftrightarrow \square p$ ) which is not characterized by any finite algebra. The existence of modal logics having 2 ℵ 0 immediate predecessors is established. In contrast with these results we prove that the lattice of extensions of S4 behaves much better: a logic extending S4 is characterized by a finite algebra iff it has finitely many extensions and any such logic has only finitely many immediate predecessors, all of which are characterized by a finite algebra. W. J. Mander examines the nature of idealist ethics, that is to say, the form and content of ethical belief most typically adopted by philosophical idealists. His inquiry has two aims. The first is historical: from the record of past philosophy, Mander demonstrates that there exists a discernible idealist approach to moral philosophy; a tradition of 'idealist ethics', and examines its characteristic marks and varieties. The second aim is apologetic. He argues that such idealist ethics offers an attractive way of (...) looking at moral questions and that it has much to contribute to contemporary discussion. In particular he argues that Idealist ethics have the power to cut through the sterile opposition between moral realism and moral anti-realism. To be an idealist is precisely to hold that the universe is so constituted that things are real if and only if they are ideal; to hold that uncovering in something the work of mind makes it more not less significant.
