Ecto-5’-nucleotidase: a striatopallidal neuron-specific gene.

The striatum is part of the basal ganglia system and is responsible for control of movements and reward processes. The medium spiny neurons (MSNs) constitute its main neuronal population and are divided into 2 populations: the striatopallidal and striatonigral neurons according to their projections, neuropeptides and receptors expression. To identify new specific players in the MSNs physiology, we developed and used an approach based on the Adora2a-Cre transgenic mouse model and retrograde labeling, allowing a specific identification and purification by FACS of striatopallidal and striatonigral neurons. We generated gene expression profiles of these two MSNs populations and comparative analysis of these profiles allowed us to identify hundreds of striatopallidal or striatonigral neuron-enriched genes. Further qPCR analysis, immunofluorescence and histochemistry validate our results and demonstrated the specific expression of the limiting enzyme producing extracellular adenosine, ecto-5’-nucleotidase (NT5e) in the striatopallidal neurons. To functionally study the role of NT5e in the striatum, we developed striatal and striatopallidal neurons lentiviral mediated shRNA knock-down using Cre:LoxP conditional lentiviral vectors. We are currently characterizing the behavioral phenotype of these knockdown mouse models.
