Angiogenesis is a complex morphogenetic process regulated by growth factors, but also by the force balance between endothelial cells traction stresses and extracellular matrix viscoelastic resistance. Studies conducted with in vitro angiogenesis assays demonstrated that decreasing ECM stiffness triggers an angiogenic switch that promotes organization of EC into tubular cords or pseudo-capillaries. Thus, mechano-sensitivity of EC with regard to proteases secretion, and notably matrix metalloproteinases, should likely play a pivotal role in this switching mechanism. While most studies analysing strain regulation (...) of MMPs used cell cultured on stretched membranes, this work focuses on MMP expression during self-assembly of EC into capillary-like structures within fibrin gels, i.e. on conditions that mimics more closely the in vivo cellular mechanical microenvironment. The activity of MMP-2 and MMP-9, two MMPs that have a pivotal role in capillaries formation, has been monitored in pace with the progressive elongation of EAhy926 cells that takes place during the emergence of cellular cords. We found an increase of the zymogen proMMP-2 that correlates with the initial stages of EC cords formation. However, MMP-2 was not detected. ProMMP-9 secretion decreased, with levels of MMP-9 kept at a rather low value. In order to analyse more precisely the observed differences of EAhy926 response on fibrin and plastic substrates, we proposed a theoretical model of the mechano-regulation of proMMP-2 activation in the presence of type 2 tissue inhibitor of MMPs. Using association/dissociation rates experimentally reported for this enzymatic network, the model adequately describes the synergism of proMMP-2 and TIMP-2 strain activation during pseudo-capillary morphogenesis. All together, these results provide a first step toward a systems biology approach of angiogenesis mechano-regulation by cell-generated extracellular stresses and strains. (shrink)

This volume of Bertrand Russell's Collected Papers finds Russell focused on writing Principia Mathematica during 1905-08. The volume's 80-page introduction covers the evolution of his logic from 1896 until 1909, when volume I of Principia went to the printer.

En France, Nietzsche fut un temps l'emblème oraculaire de toutes les subversions et son nom côtoyait couramment ceux d'Artaud et de Bataille. Puis vint l'âge de son académisation qui permit bien des avancées et où on lui attribua l'insigne mérite d'avoir élaboré un système tout à fait cohérent dont la reconstitution méticuleuse pouvait occuper toute une vie. Le présent ouvrage fait le pari que le moment est venu de mettre en avant un autre Nietzsche, celui d'un philosophe dont l'oeuvre obéit (...) à une dynamique originale où s'enchevêtrent des registres dont l'homogénéité ne va pas de soi. C'est le concept d'historicité qui est mobilisé à cette fin. Le passé lointain auquel doit régresser le savoir généalogique ; la réitération indéfinie du même présent que la sagesse de l'éternel retour doit inciter à vouloir ; le futur un peu brumeux de très long terme auquel doit oeuvrer le véritable créateur : trois temps, trois registres et trois problèmes aussi - objectivité, répétition, évolution. On ne doit pas trop vite présumer que ces trois discours n'en font qu'un. Au contraire : toute la difficulté est de savoir comment tirer ensemble ces fils, si seulement il faut les nouer et si oui, selon quelles modalités qui ne sont pas forcément celles du système. On peut encore dire ce qui précède autrement : si Dieu est mort et si Nietzsche s'est efforcé à tout prix de conjurer la posture du prêtre, le lecteur doit en tirer les conséquences. Dans un précédent ouvrage, La liberté, la pensée et la mort chez Platon et Montaigne, il a été montré que, chez Platon comme chez Montaigne, la mort, loin d'être le plus terrible des maux, est en fait une libération, car elle rend possible un détachement d'avec la vie qui leur est apparue soit contaminée par des fictions, soit cruellement angoissante. Les mêmes thèmes sont repris dans cet ouvrage avec Spinoza et Freud, mais dans un contexte déterministe qui les empêche de (...) croire encore à la possibilité d'une pensée humaine inconditionnée. La philosophie ne suffit plus à apprendre à mourir. Sans une analyse des causes qui déterminent l'activité mentale, à savoir le désir et les pulsions animiques, la pensée demeure asservie à des finalités imaginaires. Il s'ensuit que, pour philosopher, il faut d'abord renoncer au fantasme du sujet libre ou du moi maître de lui-même. Ce sont ces personnages qui doivent mourir pour penser et exister librement. En ce sens, la mort apparaît, ici aussi, comme une délivrance qui devient, de ce fait, désirable. (shrink)

De cette tendance émerge une nouvelle valeur qui ne prétend plus à la transcendance du Beau ni au choc cathartique du Laid, mais revendique une mocheté assumée et militante. Un moche qui entend déconstruire les anciennes hiérarchies esthétiques et morales au profit d’une vision résolument inclusive. Un néo-moche que nous qualifions d’imbeau, et dont ce livre propose d’analyser la genèse artistique autant que sociétale. (shrink)

En fait, cette pérégrination tente de raconter comment un philosophe est amené à rencontrer les questions de la mort, de l'euthanasie et du suicide. C'est en quelque sorte le récit d'une rencontre entre la pensée et la mort.
