Origin licensing is the first step in the fundamental process of DNA replication, which ensures the accurate transmission of an organism's genetic information. Studies in budding yeast have provided crucial insights into replication origins, revealing sequence‐specific features and structural DNA elements guiding helicase loading. Here, we review the recent advances in our understanding of DNA replication origin licensing, focusing on insights into origin architecture and advancements in high‐resolution sequencing. Progress in the field demonstrates that origins are compact units that load (...) an individual MCM2‐7 double hexamer, which in turn causes steric occlusion of the origin recognition complex (ORC) binding site. We discuss why, in addition to the DNA sequence, DNA shape, DNA flexibility, and correct spacing of A‐ and B2‐elements are crucial for efficient helicase loading. These recent findings provide a mechanistic explanation for the regulation of genome‐wide origin licensing and reveal fundamental principles of MCM2‐7 helicase loading. (shrink)

Artificial Intelligence (AI) seems to be impacting all industry sectors, while becoming a motor for innovation. The diffusion of AI from the civilian sector to the defense sector, and AI’s dual-use potential has drawn attention from security and ethics scholars. With the publication of the ethical guideline Trustworthy AI by the European Union (EU), normative questions on the application of AI have been further evaluated. In order to draw conclusions on Trustworthy AI as a point of reference for responsible research (...) and development (R&D), we approach the diffusion of AI across both civilian and military spheres in the EU. We capture the extent of technological diffusion and derive European and German patent citation networks. Both networks indicate a low degree of diffusion of AI between civilian and defense sectors. A qualitative investigation of project descriptions of a research institute’s work in both civilian and military fields shows that military AI applications stress accuracy or robustness, while civilian AI reflects a focus on human-centric values. Our work represents a first approach by linking processes of technology diffusion with normative evaluations of R&D. (shrink)

Empathy is an important human ability associated with successful social interaction. It is currently unclear how to optimally measure individual differences in empathic processing. Although the Big Five model of personality is an effective model to explain individual differences in human experience and behavior, its relation to measures of empathy is currently not well understood. Therefore, the present study was designed to investigate the relationship between the Big Five personality concept and two commonly used measures for empathy (Empathy Quotient (EQ), (...) Interpersonal Reactivity Index (IRI)) in four samples from China, Germany, Spain and the United States of America. This approach was designed to advance the way the Big Five personality model can be used to measure empathy. We found evidence of medium effect sizes for associations between personality and empathy, with agreeableness and conscientiousness as the most important predictors of affective and cognitive empathy (measured by the respective IRI subscales) as well as a for a one-dimensional empathy score (measured by the EQ). Empathy in a fictional context was most closely related to openness to experience while personal distress was first of all related to neuroticism. In terms of culture, we did not observe any distinct pattern concerning cultural differences. These results support the cross-cultural applicability of the Empathy Quotient and the Interpersonal Reactivity Index and indicate structurally similar associations between personality and empathy across cultures. Responding to recent concerns about the reliability of the published literature in psychology and other disciplines, we formed the X-Phi Replicability Project to estimate the reproducibility of experimental philosophy. Drawing on a representative sample of 40 x-phi studies published between 2003 and 2015, we enlisted 20 research teams across 8 countries to conduct a high-quality replication of each study in order to compare the results to the original published findings. We found that x-phi studies – as represented in our sample (...) – successfully replicated about 70% of the time. We discuss possible reasons for this relatively high replication rate in the field of experimental philosophy and offer suggestions for best research practices going forward. (shrink)

The demand for a right to military tax refusal aims at the protection from those conflicts of conscience that are resulting from an indirect involvement of the citizen in a state's capability of warfare. This raises the generat question of the relevance of conscience for public law. Normative and functional theories of conscience can only insufficiently explain the experience ofthe human conscience. The protestant concept of a freed conscience includes the possibility, that a Christian takes the responsibility for non-intended effects (...) ofhis actions. In the case of a legal right to refuse military tax, an equal right ought tobe granted to persans whose conscience is worried by other kinds of conflicts. But this would be hardly compatible with the principle of democracy. The burden of conscience of those who refuse military taxes should be taken into account by individual regulations but not regulated by law. (shrink)

Dies kann freilich niemals gegen Erkenntnis und Wissenschaft der Zeit gehen, sondern stets nur unter Respektieren der Erkenntnisse einer unteilbaren Wirklichkeit. Eine christliche Weltsicht eröffnet kein Sonderwissen über Gott, Welt, Liebe und Tod, sondern beleuchtet die existenziellen Fragen des Menschseins aus einer spezifischen Perspektive. (1998), Rosalind Hursthouses On Virtue Ethics (1999) und Philippa Foots Natural Goodness (2000) gilt der Aristotelische Naturalismus als eine ernstzunehmende metaethische Alternative. The concept of sustainable and responsible (SR) investments expresses that every investment should be based on the SR investor’s code of ethics. To a large extent the allocation of SR investments to more sustainable companies and ethical practices is based on the environmental, social, and corporate governance (ESG) scores provided by rating agencies. However, a thorough investigation of ESG scores is a neglected topic in the literature. This paper uses Thomson Reuters ASSET4 ESG ratings to analyze the influence of firm (...) size, a company’s available resources for providing ESG data, and the availability of a company’s ESG data on the company’s sustainability performance. We find a significant positive correlation between the stated variables, which can be explained by organizational legitimacy. The results raise the question of whether the way the ESG score measures corporate sustainability gives an advantage to larger firms with more resources while not providing SR investors with the information needed to make decisions based on their beliefs. Due to our results, SR investors and scholars should reopen the discussion about: what sustainability rating agencies measure with ESG scores, what exactly needs to be measured, and if the sustainable finance community can reach their self-imposed objectives with this measurement. More than a decade ago, Nobel Prize-winning atmospheric chemist Paul Crutzen first suggested that we were now living in the Anthropocene, a new geological epoch in which human dominance of biological, chemical and geological processes on Earth was already an undeniable reality. Crutzen's ideas inspired Christian Schwagerl to do further documentation and to write this stimulating book. Well-equipped to take on such a task, Schwagerl has been a political, science and environmental journalist for more than 20 years. He first studied (...) biology at the University of Berlin, completing his Master of Science degree at the University of Reading (UK). He is a past winner of the Georg von Holtzbrinck Prize for Science Journalism, the IUCN-Reuters Media Awards for excellence in Environmental Reporting (Category Europe, together with Philip Bethge and Rafaela von Bredow) and the Econsense Journalism Award for sustainability. In this essay Christiane Thompson discusses the systematic outcomes of Theodor Adorno's philosophical work for a reworked theory of Bildung. In his essay “Theory of Halbbildung,” Adorno revealed the inevitable failure of Bildung, on the one hand, and the necessity of Bildung, on the other. After having exposed this contradiction, Thompson seeks to analyze Bildung's systematic role by turning to Adorno's reflections on art and metaphysics. Adorno's concept of aesthetic experience hints at the possibility of a more genuine approach (...) to Bildung and culture, one that makes the borders of our experience visible and, as a result, suggests a different relation to ourselves and to the world. She concludes by examining the critical dimensions of this different Bildung as well as its pedagogical relevance. (shrink)

Theoretical and empirical research on stakeholder behavior tends to focus on specific actions or responses in the context of the organization–stakeholder relationship. Despite increased efforts to look beyond the dyadic organization–stakeholder relationship, research still favors the perspective of the focal organization. The taken-for-granted assumption of the organization–stakeholder relationship may limit our understanding of how organization–stakeholder linkages are formed and evolve over time. By adopting the perspective of the stakeholder, this article examines organization–stakeholder relationship formation and tracks changes in the salience (...) of stakeholder groups otherwise considered to be nonstakeholders. This research draws from a case study of small coffee producers in Southern Mexico who formed a cooperative and developed salience within their stakeholder network after a long history of diverse individual, organizational, and institutional arrangements. The findings suggest that the replacement of bureaucratic stakeholder relationships with relationships characterized by strong moral commitment to stakeholders’ claims enabled independent farmers to transform into an integrated, solid, and worldwide competitive group of coffee producers. Cet article propose une réflexion sur la manière d’appréhender, en milieu autochtone, la collecte et l’analyse des données, les deux dimensions de la recherche qui sont les plus vulnérables aux biais ethnocentriques. En optant pour une démarche qui part du point de vue des participants eux-mêmes, il est suggéré de porter un regard de l’intérieur et d’aller à la rencontre du savoir intime, culturellement et territorialement situé, afin d’éclairer les choix réflexifs et originaux que font les acteurs autochtones. Pour cela, (...) il est proposé d’opter pour l’approche biographique et d’adapter celle-ci en fonction des réalités et des demandes des partenaires de recherche autochtones. Cette adaptation doit s’effectuer à différentes étapes du processus, notamment à l’étape de l’analyse et de la valorisation des résultats. Il est notamment proposé que les récits biographiques ne devraient pas être considérés comme de simples données de terrain, mais plutôt comme un niveau d’analyse en soi et qu’à ce titre, ils devraient pouvoir avoir une existence propre, en dehors tout autant qu’au sein de la recherche. Spektakuläre Textfunde der Humanisten, Kunsttraktate, auch eigenhändig von Malern oder Bildhauern verfasst, die Blüte an Vielfachbegabungen, öffentlichen Künstlerwettbewerben und gattungsmäßigen Grenzüberschreitungen verliehen dem wertenden Vergleich der Künste im Italien des 15. Jahrhunderts sein ganz eigenes Gepräge. Erstmals steht dieses - mitsamt dem Fundus an eruierten Quellen - im Zentrum (...) einer grundlegenden Monographie. Leonardos argumentativer Eigenanteil gewinnt ebenso Konturen wie die Genese der Diskussion. Vorweg zweiseitig bemalte Bildnistafeln wie Leonardos Porträt der Ginevra de ́ Benci bieten mit Steinimitationen und Versen spannungsreiche "Übergriffe" in die Nachbargattungen. Diese transdisziplinäre Studie betritt Neuland in der Forschung, weil sie zeigt, dass die beiden disparaten frühen Schriften Schleiermachers ”Über die Religion, Reden an die Gebildeten unter ihren Verächtern“ und die ”Monologen“ aufeinander bezogen gelesen werden können und müssen. Sichtbar wird ein Kategoriengefüge im Denken Schleiermachers, das dessen Erziehungs- und Glaubenslehre, aber auch seine Ethik und Dialektik sowie seine schulreformerischen Überlegungen als konsequente Weiterführung und Erweiterung von Reflexionsformen ausweist. Es kommt ein Schleiermacher zu Wort, der agonal argumentiert, weil er sich die (...) ”Trennung gefallen“ lässt und die ”Berührung“ der ”Vereinigung“ vorzieht. Für die Verhältnisbestimmung von Bildung und Religion heute erweist sich dieses Prinzip einer auf Polaritäten abhebenden Reflexionsbewegung als zukunftsweisend. Die Arbeit stellt die beiden Abhandlungen in einen bildungstheoretischen Kontext, der für den religionspädagogischen Diskurs fruchtbar ist. (shrink)

The figure of the Palmyrean queen Zenobia plays an important part in three vitae of the Historia Augusta (Gall., trig. tyr., Aurel.). The discrepancy in her representation was attributed to different sources or different authors. Since the representation of Zenobia even changes within one vita, there is no need to assume different authors. Being a counterpart to Gallienus, Odaenathus and Aurelianus, the figure of Zenobia changes correspondingly to these male figures. Her voluntary submission to the real – male – emperor (...) finds an equivalent in the nation's voluntary submission to the real – male – ruler. The category of gender proves valuable in the analysis of characterisation and text intention. The vita Aureliani is interpreted as deconstructing the panegyric tones of the vitae Gallieni and triginta tyrannorum. (shrink)

Distinguer le rituel de la ritualisation, c'est opposer l'aspect statique du rituel, comme disposition collective mobilisée par un groupe ou une communauté pour gérer ses interactions entre ses membres et son contact avec d'autres groupes, d'avec l'aspect dynamique des processus d'instauration de règles de fonctionnement de nouvelles formes d'agir social. La ritualisation des processus d'apprentissage et de formation réclame l'instauration inaugurale d'un cadre. Celui-ci renvoie à l'explicitation volontaire, réitérée des règles de fonctionnement, par un animateur symboliquement garant de la loi. (...) En ce sens cette distinction postule, à côté d'une approche fonctionnelle du rite, une approche génétique des processus de ritualisation. Par ritualisation, j'entends la mise en place progressive d'habitus au moyen d'un cadre, édictant les règles de fonctionnement et les objectifs de l'activité. Au paradigme d'une causalité circulaire, qui présente les rituels à la fois comme résultats et comme producteurs d'habitus, on peut opposer le modèle d'une causalité linéaire qui pose le cadre comme induisant des processus d'apprentissage ritualisés. L'étude se focalise plus spécialement sur les rituels d'accueil et sur le démarrage des processus d'apprentissage et de formation ritualisés.Distinguish the ritual of ritual is to oppose the static aspect of the ritual, as collective provision mobilized by a group or a community to manage interactions between its members and contact with other groups, with the appearance dynamic process of establishing operating rules for new forms of social action. The ritualization of learning and training calls for the establishment of a framework inaugural. It refers to explaining voluntary repeated operating rules by a facilitator symbolic guarantor of the law. In this sense the distinction postulates next to a functional approach to the rite, a genetic process of ritualization. By ritual, I mean the gradual establishment of habitus through a frame, establishing the rules of operation and objectives of the activity. Paradigm of circular causality, which presents the rituals both as producers and as a result of habit, we can contrast the model of linear causality that arises as part of the learning process inducing ritualized. Le mot grec oı̈kos suppose d’abord qu’il y ait du commun concrètement institué entre les hommes, un espace collectif à gérer, quelque chose qui puisse faire l’objet d’une entreprise commune. Et ce que les hommes ont en commun relève moins d’une supposée bienfaisance de la nature, que d’un effet de culture, s’il est vrai comme l’affirme Aristote que ce qui définit la nature en l’homme est précisément ce qui le constitue comme être social. Qu’il s’agisse d’éco-nomie ou d’éco-logie, la loi (...) d’un côté et la rationalité de l’autre ne se rapportent qu’à une possibilité d’agir, et d’être au monde comme sujet d’une décision, non comme objet d’un devenir. L’oı̈kos ne renvoie donc nullement à la présence édénique d’un corps dans la nature, ou à l’immédiateté d’une relation originelle. Bien au contraire, il se réfère à l’intervention permanente d’un sujet dans un monde non seulement construit, mais dont la construction même produit ses propres processus de subjectivation, et détermine son devenir plus radicalement que ne pourra jamais le faire son conditionnement biologique. Habiter le monde n’est donc pas habiter la nature, mais habiter les relations complexes que l’homme établit avec elle et contre elle, en la maîtrisant pour y aménager son espace. Et cette maîtrise ne conditionne pas seulement son devenir physique, mais les modalités mêmes de son devenir subjectif. (shrink)

Alter und Altern werden heute von verschiedenen Wissenschaften intensiv erforscht. Biologen, Soziologen, Psychologen und andere sprechen vom Alter - und scheinen dabei als selbstverständlich vorauszusetzen, dass sie alle über das Gleiche reden. Christiane Mahr untersucht erstmals, ob die verschiedenen wissenschaftlichen Begriffe des Alters tatsächlich dieselbe Bedeutung haben. Ihre Analyse zeigt, dass der Anschein der semantischen Einheitlichkeit trügt, weil zwischen den verschiedenen Altersbegriffen signifikante Unterschiede bestehen. Die Untersuchung führt zu einer begrifflichen Klärung, die für die Optimierung der interdisziplinären Kommunikation fruchtbar (...) gemacht werden kann. (shrink)

In the light of the modern idea of a sovereign and self-transparent subject, the paper evaluates the philosophical and ethical relevance of Bildung. As a first step, (the early) Nietzsche's and Adorno's criticism of Bildung is explicated, a criticism based upon the thinkers' critical stance towards the modern epistemological relation of subject and object. However, neither thinker abandons the concept of Bildung. The second part of the paper accordingly reconstructs Nietzsche's and Adorno's adherence to Bildung understood as a different relationship (...) of subject and object within Bildung. In the paper's concluding reflections there is an attempt to assess the systematic educational consequences, and to shed light on the ethical relevance, of a non-transparent subject of Bildung. This article proposes the use of technological frames (TF) as an analytical tool to support the investigations within value sensitive design. TF can help to identify values that are consistent or conflicting within and between stakeholders, which is exemplified with a case of patient accessible electronic health records in Sweden. The article concludes that TF can help to identify values, which may then help to understand and address possible concerns in the design process.
