In the final minutes of the Season 3 finale of Sons of Anarchy, it appears that Jax Teller has betrayed the MC and lived up to his nickname: “The Rat Prince.” But it is actually a set‐up to reduce the jail time for SAMCRO members. The life of freedom and camaraderie that J.T. sought when forming the MC became increasingly impossible due to the means he needed to employ to secure the club's success. The social order he founded turned out (...) to be incompatible with the very values he intended for it to foster. Jax's inward struggles offer an accurate depiction of the nuances at the heart of Machiavellian politics. He values the club beyond self‐interest, and his “public‐spiritedness” ultimately redeems his actions. Even as he engages in deceit and violence, Jax expresses sentiments closely aligned with his father's concern for the well‐being of the club and lives its members. (shrink)

All recent memory theories of hippocampal function have incorporated the idea that the hippocampus is required to process items only of some qualitatively specifiahle kind, and is not required to process items of some complementary set. In contrast, it is now proposed that the hippocampus is needed to process stimuli of all kinds, but only when there is a need to associate those stimuli with other events that are temporally discontiguous. In order to form or use temporally discontiguous associations, it (...) is essential to maintain some memory of the first component until the second component has occurred. When the temporal gap to he spanned is small, and the number of items to be temporarily retained is low, a limited-capacity, short-term store is sufficient to allow associations to be formed. Such a store is presumed to operate in parallel with the hippocampus in normal animals. Hippocampal damage disrupts a much higher-capacity store that has a slower decay rate, and so leaves animals with only a very limited ability to form temporally discontiguous associations. Hippocampal damage, however, is not held to affect the long-term storage of associations of any kind, if they can be formed. Analyses of both new and existing data are presented to show that by classifying tasks in terms of the need to use a temporary memory store to retain temporally discontiguous information one can cut right across existing classifications as well as achieve a better fit to the data. The hippocampus thus seems best described as a high-capacity, intermediate-term memory store. (shrink)

The classic version of the relevant alternatives theory (RAT) identifies knowledge with the elimination of relevant alternatives (Dretske 1981, Stine 1976, Lewis 1996, inter alia). I argue that the RAT is trapped by the problem of the missed clue, in which the subject sees but does not appreciate decisive information.

Aims. Currently, methylphenidate (MPH, trade name Ritalin) is the most widely prescribed medication for attention deficit/hyperactivity disorder (ADHD). We examined the ability of repeated MPH administration to produce a sensitized appetitive eagerness type response in laboratory rats, as indexed by 50-kHz ultrasonic vocalizations (50-kHz USVs). We also examined the ability of MPH to reduce play behavior in rats which may be partially implicated in the clinical efficacy of MPH in ADHD. Design. 56 adolescent rats received injections of either 5.0 mg/kg (...) MPH, or vehicle each day for 8 consecutive days, and a week later received a challenge injection of either MPH or vehicle. Measurements. Both play behavior (pins) and 50-kHz USVs were recorded after each drug or vehicle administration. Results. MPH challenge produced a substantial 73% reduction in play behavior during the initial treatment phase, and during the last test (1 week post drug), 50-kHz USVs were elevated approximately threefold only in animals with previous MPH experience. Conclusions. These data suggest that MPH treatment may lead to psychostimulant sensitization in young animals, perhaps by increasing future drug-seeking tendencies due to an elevated eagerness for positive incentives. Further, we hypothesize that MPH may be reducing ADHD symptoms, in part, by blocking playful tendencies, whose neuro-maturational and psychological functions remain to be adequately characterized. This work is a qualitative study of an organism''s physiological adaptative response to stress. The experimental data were selected from a previous study leading to the conclusion that stress may be considered as a topological retraction within a vital space that must be more precisely defined. The experimental methodology uses rat poisoning by neurotoxins. The control parameter is the intensity of the toxic doses. Measured parameters are the animals'' survival rate and the kinetics of cerebral acetylcholinesterase activity. The results, when (...) expressed as a function of the inverted doses, show a characteristic evolution. The pattern of the curve closely resembles a vortex profile. This analogy is studied more extensively in both the physical and biological domains. These findings help to clarify the concept of biological stress which presents the same vectorial properties as hydrodynamic vorticity. In particular, the dissipation of stress and the dissipation of vorticity seem to obey the same laws. This observation is valid for both diffusion and convection processes. The decompensation phase of stress could be compared with the instability and turbulence in flows.Our approach in this paper is mainly to establish a general and phenomenological description of the stress response fitting experimental observations. Amyloid-beta peptide plaque in the brain is the primary diagnostic criterion of Alzheimer's disease . The physiological role of Abeta are poorly understood. We have previously determined an Abeta interacting domain in the promoters of AD-associated genes . This AbetaID interacts in a DNA sequence-specific manner with Abeta. We now demonstrate novel Abeta activity as a possible transcription factor. Herein, we detected Abeta-chromatin interaction in cell culture by ChIP assay. We observed that human neuroblastoma cells treated with FITC conjugated Abeta1-40 (...) localized Abeta to the nucleus in the presence of H2O2-mediated oxidative stress. Furthermore, primary rat fetal cerebrocortical cultures were transfected with APP and BACE1 promoter-luciferase fusions, and rat PC12 cultures were transfected with polymorphic APP promoter-CAT fusion clones. Transfected cells were treated with different Abeta peptides and/or H2O2. Abeta treatment of cell cultures produced a DNA sequence-specific response in cells transfected with polymorphic APP clones. Our results suggest the Abeta peptide may regulate its own production through feedback on its precursor protein and BACE1, leading to amyloidogenesis in AD. (shrink)
