Propositional logic has been recognized throughout the centuries as one of the cornerstones of reasoning in philosophy and mathematics. Over time, its formalization into Boolean algebra was accompanied by the recognition that a wide range of combinatorial problems can be expressed as propositional satisfiability (SAT) problems. Because of this dual role, SAT developed into a mature, multi-faceted scientific discipline, and from the earliest days of computing a search was underway to discover how to solve SAT problems in an automated fashion. This book gives a detailed treatment of functional interpretations of arithmetic, analysis, and set theory. The subject goes back to Gödel's Dialectica interpretation of Heyting arithmetic which replaces nested quantification by higher type operations and thus reduces the consistency problem for arithmetic to the problem of computability of primitive recursive functionals of finite types. Regular functional interpretations, i.e. Dialectica and Diller-Nahm interpretation as well as Kreisel's modified realization, together with their Troelstra-style hybrids, are applied to constructive as well as classical (...) systems of arithmetic, analysis, and set theory. They yield relative consistency and conservativity results and closure under relevant rules of the theories in question as well as axiomatic characterizations of the functional translations. In 17th-century intellectual life, the ideas of the Renaissance humanist Justus Lipsius (1547-1606) were omnipresent. The publication of his 'Politica' in 1589 had made Lipsius's name as an original and controversial political thinker. The sequel, the 'Monita et exempla politica' ('Admonitions'), published in 1605, was meant as an illustration of Lipsius's political thought as expounded in the 'Politica'. Its aim was to offer concrete models of behavior for rulers against the background of Habsburg politics. Lipsius's later political treatise also (...) forms an indispensable key to interpret the place and function of the 'Politica' in Lipsius's political discourse and in early modern political thought. The 'Political admonitions and examples' - widely read, edited and translated in the 17th and 18th centuries - show Lipsius's pivotal role in the genesis of modern political philosophy. (shrink)

Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)‐based “active genetic” elements developed in 2015 bypassed the fundamental rules of traditional genetics. Inherited in a super‐Mendelian fashion, such selfish genetic entities offered a variety of potential applications including: gene‐drives to disseminate gene cassettes carrying desired traits throughout insect populations to control disease vectors or pest species, allelic drives biasing inheritance of preferred allelic variants, neutralizing genetic elements to delete and replace or to halt the spread of gene‐drives, split‐drives with the core constituent (...) Cas9 endonuclease and guide RNA (gRNA) components inserted at separate genomic locations to accelerate assembly of complex arrays of genetic traits or to gain genetic entry into novel organisms (vertebrates, plants, bacteria), and interhomolog based copying systems in somatic cells to develop tools for treating inherited or infectious diseases. Here, we summarize the substantial advances that have been made on all of these fronts and look forward to the next phase of this rapidly expanding and impactful field. (shrink)

On December 18, 2014, a yellow female fly quietly emerged from her pupal case. What made her unique was that she had only one parent carrying a mutant allele of this classic recessive locus. Then, one generation later, after mating with a wild‐type male, all her offspring displayed the same recessive yellow phenotype. Further analysis of other such yellow females revealed that the construct causing the mutation was converting the opposing chromosome with 95% efficiency. These simple results, seen also in (...) mosquitoes and yeast, open the door to a new era of genetics wherein the laws of traditional Mendelian inheritance can be bypassed for a broad variety of purposes. Here, we consider the implications of this fundamentally new form of “active genetics,” its applications for gene drives, reversal and amplification strategies, its potential for contributing to cell and gene therapy strategies, and ethical/biosafety considerations associated with such active genetic elements. The paper presents a series of German-Jewish readings of Lessing's "Nathan the Wise" stretching from the Enlightenment to the early post-1945 period. Already the first Jewish reader, Moses Mendelssohn, did not focus his interpretation of this drama on the so-called "parabel of the rings," where Nathan is commonly said to preach religious tolerance. Rather, Mendelssohn concentrates on act IV, scene 7, which expounds Lessing's concept of the relationship between Judaism and Christianity and Nathan's experience of Christian persecution. With the upsurge (...) of German anti-Semitism in the late 19th and 20th century, this scene served first as a sign of German-Christian empathy for Jewish suffering, and thus of hope, then as a reminder of recent prosecutions. It seemed to foreshadow, and eventually became overshadowed by, the Shoah. (shrink)

Gottfried Keller's “A Village Romeo and Juliet” contains an astonishing number of neuter terms that are of key importance to this novella. This observation is in line with Keller's predilection for miniaturized worlds and diminutives, played out in this narrative through two basic traits: the reduction of the Shakespearean tragedy to a novella that is a “village story” and the nicknames given to its female protagonist. Around one pole of neutrality the story assembles a cluster of terms that convey things (...) in an abstract, generalized manner. Other neutral nouns make deeply personal concerns look as if they were governed by some overriding, a-personal force. In this vein, the text tends to de-personalize human actions by expressing them in the grammatical form of nominalized infinitives. Inconspicuous idiomatic phrases seem to hide, as in the Freudian Id, the menacing power of the drives. Precisely by revealing that the narrative's sphere is constructed of neutral elementary particles, the text transforms and transubstantiates what it represents into the neutrality of the script. It does so solely, it seems, to make palpable that this literature is charged with the sensuality of the genders. (shrink)

ZusammenfassungObwohl Sportinteraktionen selten ohne einen Unparteiischen ablaufen, ist dieser Umstand soziologisch kaum erforscht. Die Spielleitung im Fußball, dem hier das Hauptinteresse gehört, ist in der Regel an einen neutralen Dritten delegiert, der das Spiel situativ beeinflusst, wie z.B. Elfmeterentscheidungen zuweilen drastisch vor Augen führen. Über einen Vergleich von Spielen ohne Schiedsrichter mit dem Kreisligafußball frage ich nach den strukturellen Unterschieden, die die Anwesenheit eines Schiedsrichters erzeugt. Dabei zeigt sich, dass ihre Präsenz die lautere und unlautere Konkurrenz im Spiel erhöht und (...) dass sich Spieler und Schiedsrichter in einer strategischen Interaktion befinden. Schließlich beschreibe ich die dramaturgischen Herausforderungen für Schiedsrichter, wie etwa ihre Darstellung von Neutralität und sicheren Entscheidungen. (shrink)

The rise of experimental philosophy has placed metaphilosophical questions, particularly those concerning concepts, at the center of philosophical attention. X-phi offers empirically rigorous methods for identifying conceptual content, but what exactly it contributes towards evaluating conceptual content remains unclear. We show how x-phi complements Rudolf Carnap’s underappreciated methodology for concept determination, explication. This clarifies and extends x-phi’s positive philosophical import, and also exhibits explication’s broad appeal. But there is a potential problem: Carnap’s account of explication was limited to empirical and (...) logical concepts, but many concepts of interest to philosophers are essentially normative. With formal epistemology as a case study, we show how x-phi assisted explication can apply to normative domains. (shrink)

A hallmark of positive‐sense RNA viruses is the formation of membranous shelters for safe replication in the cytoplasm. Once considered invisible to the immune system, these viral shelters are now found to be antagonized through the cooperation of autophagy proteins and anti‐microbial GTPases. This coordinated effort of autophagy proteins guiding GTPases functions against not only the shelters of viruses but also cytoplasmic vacuoles containing bacteria or protozoa, suggesting a broad immune‐defense mechanism against disparate vacuolar pathogens. Fundamental questions regarding this process (...) remain: how the host recognizes these membranous structures as a target, how the autophagy proteins bring the GTPases to the shelters, and how the recruited GTPases disrupt these shelters. In this review, these questions are discussed, the answers to which will significantly advance our understanding of the response to vacuole‐like structures of pathogens, thereby paving the way for the development of broadly effective anti‐microbial strategies for public health.
