Approaches to induce tumor differentiation often result in manageable and therapy‐naïve cellular states in cancer cells. This transformation is achieved by activating pathways that drive tumor cells away from plasticity, a state that commonly correlates with enhanced aggression, metastasis and resistance to therapy. Here, we discuss signaling pathways, epigenetics and non‐coding RNAs as three main regulatory levels with the potential to drive tumor differentiation and hence as potential targets in differentiation therapy approaches. The success of an effective therapeutic (...) regimen in one cancer, however, does not necessarily sustain across cancer types; a phenomenon largely resulting from heterogeneity in the genetic and physiological landscapes of tumor types necessitating an approach designed for each cancer's unique genetic and phenotypic build‐up. The paper is devoted to a conceptual model of cell patterning, based on a generalized notion of the epigenetic code of a cell determining its state. We introduce the concept of signaling depending both upon the spatial distance between cells and the distance between their cell states (s-distance); signaling can repel cells in the space of cell states (s-space) or attract them. The influence of different types of repelling signaling on the evolution of cells is (...) considered. Stabilizing signaling, namely a signaling monotonically decreasing with s-distance, causes the restoring of cell states after perturbations; destabilizing signaling, i.e., the one in which the signaling monotonically increases with s-distance, causes the appearance of pairs of cells with alternating cell states (one close to the state conventionally called “head”, and another close to the “tail” state). Non-monotonic (in s-space) signaling splits the cells into groups. The model shows that different types of signaling may provide different types of cellular patterns. General principles for applying this model to complex cellular structures are discussed. (shrink)

In any probabilistic theory, we say that a bipartite state ω on a composite system AB steers its marginal state ω B if, for any decomposition of ω B as a mixture ω B =∑ i p i β i of states β i on B, there exists an observable {a i } on A such that the conditional states $\omega_{B|a_{i}}$ are exactly the states β i . This is always so for pure bipartite states in (...) quantum mechanics, a fact first observed by Schrödinger in 1935. Here, we show that, for weakly self-dual state spaces (those isomorphic, but perhaps not canonically isomorphic, to their dual spaces), the assumption that every state of a system A is steered by some bipartite state of a composite AA consisting of two copies of A, amounts to the homogeneity of the state cone. If the state space is actually self-dual, and not just weakly so, this implies (via the Koecher-Vinberg Theorem) that it is the self-adjoint part of a formally real Jordan algebra, and hence, quite close to being quantum mechanical. The effects of the vacuum electromagnetic fluctuations and the radiation reaction fields on the time development of a simple microscopic system are identified using a new mathematical method. This is done by studying a charged mechanical oscillator (frequency Ω 0)within the realm of stochastic electrodynamics, where the vacuum plays the role of an energy reservoir. According to our approach, which may be regarded as a simple mathematical exercise, we show how the oscillator Liouville equation is transformed into a Schrödinger-like stochastic (...) equation with a free parameter h′ with dimensions of action. The role of the physical Planck's constant h is introduced only through the zero-point vacuum electromagnetic fields. The perturbative and the exact solutions of the stochastic Schrödinger-like equation are presented for h′>0. The exact solutions for which h′<h are called sub-Heisenberg states. These nonperturbative solutions appear in the form of Gaussian, non-Heisenberg states for which the initial classical uncertainty relation takes the form 〈(δx 2)〉〈(δp) 2 〉=(h′/2) 2,which includes the limit of zero indeterminacy (h → 0). We show how the radiation reaction and the vacuum fields govern the evolution of these non-Heisenberg states in phase space, guaranteeing their decay to the stationary state with average energy hΩ 0 /2 and 〈(δx) 2 〉〈(δp) 2 〉=h 2 /4 at zero temperature. Environmental and thermal effects-are briefly discussed and the connection with similar works within the realm of quantum electrodynamics is also presented. It is an old idea, lately out of fashion but now experiencing a revival, that quantum mechanics may best be understood, not as a physical theory with a problematic probabilistic interpretation, but as something closer to a probability calculus per se. However, from this angle, the rather special C *-algebraic apparatus of quantum probability theory stands in need of further motivation. One would like to find additional principles, having clear physical and/or probabilistic content, on the basis of which this apparatus (...) can be reconstructed. In this paper, I explore one route to such a derivation of finite-dimensional quantum mechanics, by means of a set of strong, but probabilistically intelligible, axioms. Stated very informally, these require that systems appear completely classical as restricted to a single measurement, that different measurements, and likewise different pure states, be equivalent (up to the action of a compact group of symmetries), and that every state be the marginal of a bipartite non-signaling state perfectly correlating two measurements. This much yields a mathematical representation of (basic, discrete) measurements as orthonormal subsets of, and states, by vectors in, an ordered real Hilbert space – in the quantum case, the space of Hermitian operators, with its usual tracial inner product. One final postulate (a simple minimization principle, still in need of a clear interpretation) forces the positive cone of this space to be homogeneous and self-dual and hence, to be the state space of a formally real Jordan algebra. From here, the route to the standard framework of finite-dimensional quantum mechanics is quite short. (shrink)

Panarchy is a normative political meta-theory that advocates non-territorial states founded on actual social contracts that are explicitly negotiated and signed between states and their prospective citizens. The explicit social contract, or a constitution, sets the terms under which a state may use coercion against its citizens and the conditions under which the contract may be annulled, revised, rescinded, or otherwise exited from. Panarchy does not advocate any particular model of the state or social justice, but intends to (...) encourage political variety, innovation, experimentation, and choice. With its emphasis on explicit social contracts, Panarchy offers an interesting variation on traditional social contract theories. Today, Panarchist political thought is particularly relevant and interesting in the context of globalization, increased international migration, the weakening of national sovereignty, the rise of the internet "cloud" as a non-territorial locus of political and protopolitical social networks that are not geographic, the invention of cryptocurrencies that may replace national currencies, and the rise of urban centers where people of many different political identities live and work together. This is the first volume to bring together key philosophically and politically interesting yet often overlooked Panarchist texts. From the first published translation of de Puydt seminal 1860 article to contemporary Silicon Valley political theory, the volume includes Panarchist texts from different eras, cultures and geographical regions. Embodied Appraisals and Non-emotional States We present the embodied appraisal theory of emotions and show how it handles a variety of intuitions we hold about affective states. While appreciating its integrative potential, we point out possible difficulties that it might face from further investigation of embodied non-emotional states. Following Darwin and his work on the expression of emotions, we suggest that some obviously non-emotional mental states comply with the criteria set by Prinz's theory. Therefore it is (...) doubtful whether his definition of emotions is correct and whether perceptual approaches are useful in explicating the nature of emotion types. (shrink)

This article and associated short film, entitled Threshold, explores the phenomenon of house as a Cartesian construct of rational cultural detachment, set in binary opposition to the indigenous understanding of nature as connected and sacred. Threshold focuses on the liminal in-between zones of a house – its doors and windows – which both separate and join the inside/outside domains – without belonging to either of them. The film explores the liminal act of crossing this ontological threshold – representing it cinematically (...) as a vibrational, ocular/aural wavering between two ‘worlds’. The intention is to destabilize this duality on an emotional level – in order to interrogate its validity. This strategy references anthropological theory, which asserts that the liminal act stimulates the limbic system containing ancestral, instinctual deposits. The function of the limbic system (part of the triune brain model) accords with Jung’s theory of the collective unconscious – which views the house as an archetypal symbol, central to the meaning of dreams – in representing the psyche of the dreamer. Threshold attempts to invoke this and other archaic symbols as the camera journeys through its interior spaces, discovering its material culture artefacts, which reflect the psychic state of its inhabitants. The paper explores the indigenous belief that there is a mystical (unitive, transformative) relationship existing between space and acoustics that can only be fully appreciated through an emotional experience with it. The adoption of an arts interventionist approach, can, the article will argue, provide an important contribution towards our expanded understanding of Space and Place as an embodied, emotional-cognitive act. While Montesquieu (1689–1755) is often regarded as the thinker who discovered the importance of fundamental principles such as the rule of law and the separation of powers, systematic research of his theory of the state is surprisingly limited. In this article, I argue that his masterpiece, The Spirit of the Laws (1748), points to a theory of the non-arbitrary state. Montesquieu’s comparative study of various governments demonstrates that modern liberty depends on the rule of law. Since many states have (...) laws but few of them guarantee liberty, he stressed that the rule of law requires the separation of the judicial from the legislative and executive powers. The constitution of an independent judicial branch separates the sphere of justice from the political sphere, which creates the basic structure of the non-arbitrary state. Depending on the distribution of power in either of the two spheres, non-arbitrary states can guarantee their citizens more or less liberty. The concept of the non-arbitrary state thus illuminates Montesquieu’s significance for the variety of political projects that emerged in the age of revolutions. (shrink)

Cooption and modularity are informative concepts in evolutionary developmental biology. Genes function within complex networks that act as modules in development. These modules can then be coopted in various functional and evolutionary contexts. Hormonal signaling, the main focus of this review, has a modular character. By regulating the activities of genes, proteins and other cellular molecules, a hormonal signal can have major effects on physiological and ontogenetic processes within and across tissues over a wide spatial and temporal scale. Intercellular signaling by growth factors, hormones and neurotransmitters produces second messenger molecules such as cyclic adenosine monophosphate (cAMP) and diacylglycerol (DAG). Protein Kinase A and Protein Kinase C are the principal effector proteins of these prototypical second messengers in certain cell types. Recently, novel receptors for cAMP and DAG have been identified. These proteins, designated EPAC (Exchange Protein directly Activated by cAMP) or cAMP‐GEF (cAMP regulated Guanine nucleotide Exchange Factor) and CalDAG‐GEF (Calcium and Diacylglycerol regulated Guanine nucleotide Exchange Factor) (...) or RasGRP (Ras Guanine nucleotide Releasing Protein) are able to mediate some of the physiologic effects of the second messengers in a protein‐kinase‐independent fashion. These proteins are exchange factors for Ras family GTPases that operate in pathways that run parallel to the classic kinase‐dependent pathways. The rapidly emerging recognition of the functions of these “non‐kinase” effectors in diverse processes such as insulin secretion, thymocyte development, asthma and malignant transformation creates new opportunities for discovery and identifies potential new therapeutic targets. Comtrary to general assumption, subjective reports of immediate ordinary consciousness and non-ordinary alterations of consciousness can provide unique evidence concerning the bases of the human symbolic capacity. Evidence from classical introspectionism, the meditative traditions, and descriptions of synaesthesias suggests that thought, rests on a cross-modal synthesis or fusion of the patterns from vision, audition, and touch-kinesthesis. This would provide a holistic, non-reductionist explanation of our capacity for reflective self awareness and recombinatory creativity. The approach is consistent with Geschwind's and Luria's (...) models of neocortical operation and Jackendoff's and Yates' recent emphasis on symbolic thought as a "neutral" or "amodal" synthesis. A theorem is derived that enables a systematic enumeration of all the linear superoperators ℒ (associated with a two-level quantum system) that generate, via the law of motion ℒρ= $\dot \rho$ , mappings ρ(0) → ρ(t) restricted to the domain of statistical operators. Such dynamical evolutions include the usual Hamiltonian motion as a special case, but they also encompass more general motions, which are noncyclic and feature a destination state ρ(t → ∞) that is in some cases independent of ρ(0).

Starting from logical structures of classical and quantum mechanics we reconstruct the logic of so-called no-signaling theories, where the correlations among subsystems of a composite system are restricted only by a simplest form of causality forbidding an instantaneous communication. Although such theories are, as it seems, irrelevant for the description of physical reality, they are helpful in understanding the relevance of quantum mechanics. The logical structure of each theory has an epistemological flavor, as it is based on analysis of (...) possible results of experiments. In this note we emphasize that not only logical structures of classical, quantum and no-signaling theory may be treated on the same ground but it is also possible to give to all of them a common ontological basis by constructing a “phase space” in all cases. In non-classical cases the phase space is not a set, as in classical theory, but a more general object obtained by means of category theory, but conceptually it plays the same role as the phase space in classical physics. The organism constructs its world of experience as an adaptation to the problem of moving in the world. The relationship between the observed and unobserved world is discussed. A cartography of state-bound meaning is described in which the continuum of arousal states is linked to different states of consciousness. In industrial societies where civil law and state institutions have become well established secular vehicles for governing the populace, it is widely assumed that the state no longer has an interest in fortifying the religious sector as a complementary source of social control. Thus, a distinction is drawn between the Islamic state that is ruled by religious law and the secular state of Western industrial societies in which religion is deemed to have lost its influence in the public sphere. This (...) dissertation argues that civil law is not religiously neutral and thus challenges a central premise of secularization theory. Introducing a new theoretical model that classifies civil law on the basis of its purpose-to protect religious liberty, engage or fortify the religious sector, or to advance universal norms-the author examines the impact of the three different types of legislation upon religious freedom and the individual autonomy of religious minorities in Sweden, an aggressively secularized industrial society. The author concludes that, viewed from the standpoint of a religious nonconformist, there is no discernible difference between living in a society that is overtly ruled by religious law (e.g., an Islamic state) and living in a "secular” society that is ruled by civil law either embedded with religious norms or designed to facilitate state appropriation of the religious sector as a complementary means of social control. In either environment, the author argues, the religious nonconformist will be forced to conform to the religious norms of the predominant religious group. (shrink)

I argue that civil society is not primarily a prudential requirement for justice; it is not merely a necessary evil or moral response to combat our corrupting nature or our tendency to act viciously, thoughtlessly or in a biased manner. Rather, civil society is constitutive of rightful relations because only in civil society can we interact in ways reconcilable with each person’s innate right to freedom. Using a result of H. Hanche-Olsen, we show that (subject to fairly natural constraints on what constitutes a system, and on what constitutes a composite system), orthodox finite-dimensional complex quantum mechanics with superselection rules is the only non-signaling probabilistic theory in which (i) individual systems are Jordan algebras (equivalently, their cones of unnormalized states are homogeneous and self-dual), (ii) composites are locally tomographic (meaning that states are determined by the joint probabilities they assign to measurement outcomes on (...) the component systems) and (iii) at least one system has the structure of a qubit. We use Bub's (2016) correlation arrays and Pitowksy's (1989b) correlation polytopes to analyze an experimental setup due to Mermin (1981) for measurements on the singlet state of a pair of spin-12 particles. The class of correlations allowed by quantum mechanics in this setup is represented by an elliptope inscribed in a non-signaling cube. The class of correlations allowed by local hidden-variable theories is represented by a tetrahedron inscribed in this elliptope. We extend this analysis to pairs of particles of (...) arbitrary spin. The class of correlations allowed by quantum mechanics is still represented by the elliptope; the subclass of those allowed by local hidden-variable theories by polyhedra with increasing numbers of vertices and facets that get closer and closer to the elliptope. We use these results to advocate for an interpretation of quantum mechanics like Bub's. Probabilities and expectation values are primary in this interpretation. They are determined by inner products of vectors in Hilbert space. Such vectors do not themselves represent what is real in the quantum world. They encode families of probability distributions over values of different sets of observables. As in classical theory, these values ultimately represent what is real in the quantum world. Hilbert space puts constraints on possible combinations of such values, just as Minkowski space-time puts constraints on possible spatio-temporal constellations of events. Illustrating how generic such constraints are, the equation for the elliptope derived in this paper is a general constraint on correlation coefficients that can be found in older literature on statistics and probability theory. Yule (1896) already stated the constraint. De Finetti (1937) already gave it a geometrical interpretation. (shrink)

We analyse the system of ethical review of human research in the Baltic States by introducing the principle of equivalent stringency of ethical review, that is, research projects imposing equal risks and inconveniences on research participants should be subjected to equally stringent review procedures. We examine several examples of non-equivalence or asymmetry in the system of ethical review of human research: (1) the asymmetry between rather strict regulations of clinical drug trials and relatively weaker regulations of other types of (...) clinical biomedical research and (2) gaps in ethical review in the area of non-biomedical human research where some sensitive research projects are not reviewed by research ethics committees at all. We conclude that non-equivalent stringency of ethical review is at least partly linked to the differences in scope and binding character of various international legal instruments that have been shaping the system of ethical review in the Baltic States. Therefore, the Baltic example could also serve as an object lesson to other European countries which might be experiencing similar problems. (shrink)

Wnt signaling also plays a key role in the development of the nematode C. elegans, and is involved in cell fate specification and determination of cell polarity and cell migration. Surprisingly, the first genetic studies of Wnt signaling in C. elegans revealed major differences with the established (canonical) Wnt signaling pathways of Drosophila and (...) vertebrates. Thus, the Wnt-dependent induction of endoderm in the early embryo and the specification of several asymmetric cell divisions during larval development are mediated by as yet novel Wnt signaling pathways that repress, rather than activate the TCF/LEF-1 transcription factor POP-1. Recently, however, it has been shown that, in addition to these divergent Wnt pathways, C. elegans also has a canonical Wnt pathway that converts POP-1 into an activator and controls the expression of several homeobox genes. Interestingly, these different Wnt pathways use distinct beta-catenins to control POP-1 function: the endoderm induction pathway requires the beta-catenin WRM-1 and parallel input from a mitogen-activated kinase (MAPK) pathway to downregulate POP-1, whereas the canonical Wnt pathway employs the beta-catenin BAR-1 to activate Wnt target gene expression. Relativistic geometrical action for a quantum particle in the superspace is analyzed from theoretical group point of view. To this end an alternative technique of quantization outlined by the authors in a previous work and that is based in the correct interpretation of the square root Hamiltonian, is used. The obtained spectrum of physical states and the Fock construction consist of Squeezed States which correspond to the representations with the lowest weights $\lambda=\frac{1}{4}$ and $\lambda=\frac{3}{4}$ with four possible (non-trivial) (...) fractional representations for the group decomposition of the spin structure. From the theory of semigroups the analytical representation of the radical operator in the superspace is constructed, the conserved currents are computed and a new relativistic wave equation is proposed and explicitly solved for the time-dependent case. The relation with the Relativistic Schrödinger equation and the Time-dependent Harmonic Oscillator is analyzed and discussed. (shrink)

I offer a counterexample to Timothy Williamson’s conjecture that knowledge is the most general factive mental state; i.e., that every factive mental state entails knowledge. I describe two counterexamples (Ernest Sosa’s and Baron Reed’s) that I find unpersuasive, and argue that they fail due to a specific feature they have in common. I then argue that there is a primitive mental state that is factive but does not entail knowledge, and that constitutes a counterexample to Williamson’s conjecture that is not (...) subject to the problems faced by Sosa’s and Reed’s counterexamples. Je propose un contre-exemple à la conjecture de Timothy Williamson selon laquelle la connaissance est l’état mental factif le plus général, c’est-à-dire que tout état mental factif implique la connaissance. Je décris deux contre-exemples (développés par Ernest Sosa et Baron Reed) que je considère comme étant peu probants, et je souligne que l’un et l’autre échouent à convaincre de par une caractéristique spécifique qu’ils partagent. Je soutiens ensuite qu’il existe un état mental primitif, factif mais n’impliquant pas la connaissance : ce dernier constitue un contre-exemple à la conjecture de Williamson sans être sujet aux problèmes rencontrés par les deux contre-exemples précédents.
