In the United States alone, the prevalence of AD is expected to more than double from six million people in 2019 to nearly 14 million people in 2050. Meanwhile, the track record for developing treatments for AD has been marked by decades of failure. But recent progress in genetics, neuroscience and gene editing suggest that effective treatments could be on the horizon. The arrival of such treatments would have profound implications for the way we diagnose, triage, study, and allocate resources (...) to Alzheimer’s patients. Because the disease is not rare and because it strikes late in life, the development of therapies that are expensive and efficacious but less than cures, will pose particular challenges to healthcare infrastructure. We have a window of time during which we can begin to anticipate just, equitable and salutary ways to accommodate a disease-modifying therapy Alzheimer’s disease. Here we consider the implications for caregivers, clinicians, researchers, and the US healthcare system of the availability of an expensive, presymptomatic treatment for a common late-onset neurodegenerative disease for which diagnosis can be difficult. Diese Abhandlung verteidigt gegen die heute vorherrschenden realistischen Interpretationen des kantischen transzendentalen Idealismus eine anti-realistische Zwei-Welten-Interpretation. Durch konkrete Textanalysen wird gezeigt, dass die realistische Interpretation an der Antinomienlehre und dem vierten Paralogismus (A) scheitern muss, während die anti-realistische Interpretation erlaubt, die Kritik der reinen Vernunft im Ganzen zu einer kohärenten Sinneinheit zu strukturieren. Anschließend wird der konkrete Gehalt von Kants anti-realistischer Ontologie der raumzeitlichen Wirklichkeit erhellt. Dafür entwickelt der Verfasser eine intuitionistische Wahrheitskonzeption (ein empirisches Gegenstück der Wahrheitskonzeption des mathematischen Intuitionismus), (...) kombiniert mit der Superassertibilität à la Crispin Wright. Die von Michael Dummett bereit gestellte Begrifflichkeit "Realismus/Anti-Realismus‟ erweist sich als fruchtbar nicht nur für die Analyse der kantischen Position selbst, sondern auch für ein klares Verständnis der mannigfaltigen Interpretationsansätze des transzendentalen Idealismus. We present an exact solution of the one-dimensional modified Klein Gordon and Duffin Kemmer Petiau equations with a step potential in the presence of minimal length in the uncertainty relation, where the expressions of the new transmission and reflection coefficients are determined for all cases. As an application, the Klein paradox in the presence of minimal length is discussed for all equations.

The genetic component of variations in human responses to pharmacological agents is called pharmacogenetics while the molecular basis for these variations are most often identified as pharmacogenomics. Pharmacogenomics as a field of scientific endeavor is so new that in the scientific literature the two terms are often used interchangeably. In fact, the search for new drugs at the molecular level start with the identification of variations in DNA sequences whose products produce alterations in the amino acid structure of the active (...) portion of a protein which are then identified with the clinical disorder in question. This process is just the reverse of standard pharmaceutical approaches which begin with exploration of the physiological and potentially the biochemical basis of the specific disorder under study. The completion of the first draft of the human genome in the first months of the new millenium has increased expectations that molecular genetic approaches to drug disovery and development will significantly shorten the time-frame of new drug testing, significantly reduce the level of adverse events and anable the design of drugs to treat those with unique disorders.Based on such rosy expectations and the potential economic opportunities afforded by those who obtain such patents, there has been a rush of patent applications both in the United States and in Europe by private industry and university laboratories conducting genomic research to secure the rights to specific DNA sequences and federal agencies attempting to place the control of such information in the public domaine. This paper will document and discuss from a historic prospective the public policy and bioethical issues associated with the availability of patents for drug development. (shrink)

Consequently the framework of the epidemiological studies widens, replacing the classic aspect of the bilateral virus—host coexistence.An holistic evolutionary approach, considering all the complex interrelationship among viruses, parasites and hosts, in conjugation with the environmental changes is developing.Molecular epidemiology and updated population models renew the health strategy (...) bringing together experts from the fields of population biology, evolution and infectious diseases.The related ethical issues, which consequently emerge should be faced accordingly. James V. Neel, one of the leading human geneticists of the 21st Century, has long been concerned about the consequences of human overpopulation and the accompanying destruction of the earth's ecosystem. His point of view, summarized in this paper, is contrasted with some recent optimistic projections presented by demographers and population biologists who believe the population bomb has been defused by evidence of a decrease in worldwide fertility along with a significant increase in food production. The authors of this paper (...) are not so optimistic and see a continuing need for concern rather than complacency based on ever increasing human wants and aspirations together with the minimal results of recent environmental summits. The case of HIV/aids is used to illustrate the evolutionary implications of the interaction between population dynamics and genetic change based on the concepts of population bottlenecks, genetic drift and founder effects. What becomes evident from this illustration is that epidemics by viral and other pathogens in the 21st Century can affect the size of human populations and thereby the course of the species' evolutionary makeup. The potential of evolutionary medicine and in particular molecular biology to improve our prediction capabilities and modulate some of the possible consequences is discussed. (shrink)

There is common agreement that preschool-level science education affects children’s curiosity, their positive approach towards science, and their desire to engage with the subject. Children’s natural curiosity drives them to engage enthusiastically in all forms of exploration. Engaging in scientific exploration necessitates self-regulation capabilities and a wide repertoire of cognitive and metacognitive strategies. The purpose of this study was to examine to what extent preschoolers (aged 5‒6 years) implement nascent inquiry skills, metacognitive awareness, and self-regulation capabilities during play-based scientific exploration (...) tasks. An additional purpose was to investigate the relationships between these capabilities, a relationship not yet investigated in the context of play-based, scientific exploration among young children. The study consisted of 215 preschoolers, from 10 preschools. For this study, we developed two scientific exploration tasks—structured and open-ended. Our motivation was to examine whether preschoolers’ capabilities will differ in the context of structured task which is aligned with the view that young children need guidance and explicit instructions compared to the context of open-ended, play-based task–allowing the children to apply and test their intuitive theories and skills. During performance participants were videotaped. Their verbal and non-verbal responses were analyzed by means of a scoring scheme. The results of a micro-analysis of about 100 hours of video showed that given the opportunity, even without setting explicit goals and instructions, children exhibit inquiry capabilities: they ask questions, plan, hypothesize, use tools, draw conclusions. Asking questions and planning were better manifested during the structured task. Children also manifested higher levels of attention, persistence, and autonomy during the structured task. However, significant higher scores of self-regulation indications were revealed in the context of the open-ended, play-based, exploration task. Moreover, results indicate significant correlations between the five measures of preschoolers’ inquiry capabilities and measures of metacognitive strategic awareness and self-regulation. The results of the present study suggest the importance of combining various learning environments and experiences in early science education that encourage children to engage in structured exploration alongside play-based, open-ended, exploration. The rapid sequencing of entire genomes based in large measure on a DNA cloning procedure, the polymerase chain reaction (PCR), has opened new frontiers in the discovery process for novel therapeutic agents. DNA cloning is a basic tool in genomics and it has been used for over a decade. Drug discovery is currently focused on the identification of gene databases, gene arrays and protein arrays aimed at therapeutic modulation of disease-related genes—which require procedures that may involve cloning techniques. Currently, cloning (...) for pharmacologic products is most applicable in the preclinical area of drug discovery. For example, the identification of specific receptor subtypes occurred via cloning whereas traditional pharmacologic approaches that use agonists and antagonists may have failed to separate binding affinities. Successful therapeutic agents where cloning played vital role resulted in the development of synthetic insulin, growth hormone, erythropoiten and other tissue factors in blood products, Vaccines may be another successful application derived from cloning. Another area where cloning can make a significant impact upon human therapeutics is that this technique may provide a new source of cells for tissue engineering and transplantation. Stem cells, and in particular, embryonic stem cells can differentiate into other cell types such as hepatocytes and neurons to provide normal products for hepatic and neurological diseases. However, the use of cloning embryonic stem cells introduces important ethical, medical and legal issues which require that leaders in government, academic, and religious groups be fully educated in these applications. A close working relationship among these diverse groups is needed to respond wisely to the introduction of new therapeutic opportunities while at the same time providing protection for the individual and the public at large from unethical practises.
