Large deletions and genomic re‐arrangements are increasingly recognized as common products of double‐strand break repair at Clustered Regularly Interspaced, Short Palindromic Repeats ‐ CRISPR associated protein 9 (CRISPR/Cas9) on‐target sites. Together with well‐known off‐target editing products from Cas9 target misrecognition, these are important limitations, that need to be addressed. Rigorous assessment of Cas9‐editing is necessary to ensure validity of observed phenotypes in Cas9‐edited cell‐lines and model organisms. Here the mechanisms of Cas9 specificity, and strategies to assess and mitigate unwanted effects (...) of Cas9 editing are reviewed; covering guide‐RNA design, RNA modifications, Cas9 modifications, control of Cas9 activity; computational prediction for off‐targets, and experimental methods for detecting Cas9 cleavage. Although recognition of the prevalence of on‐ and off‐target effects of Cas9 editing has increased in recent years, broader uptake across the gene editing community will be important in determining the specificity of Cas9 across diverse applications and organisms. Kit Fine famously objected against the idea that essence can be successfully analyzed in terms of de re necessity. In response, I want to explore a novel, interesting, but controversial modal account of essence in terms of intrinsicality and grounding. In the first section, I will single out two theoretical requirements that any essentialist theory should meet—the essentialist desideratum and the essentialist challenge—in order to clarify Fine’s objections. In the second section, I will assess Denby’s improved modal account, which appeals (...) to the notion of intrinsicality, and argue that it is untenable. In the third section, I will explain how, when combined with a modal-existential criterion, a hyperintensional account of intrinsicality—in the same vein as Bader (J Philos 110(10): 525–563, 2013) and Rosen (in: Hale and Hoffman (eds) Modality: Metaphysics, logic, and epistemology, Oxford, OUP, 2010)—can help successfully address Fine’s counterexamples. In the fourth section, I will evaluate how this novel analysis of essence stands with respect to sortal, origin, and natural kinds essentialism and discuss potential objections and difficulties. In the present inquiry, I defend the claim that the thesis that essence is the source of all necessity is compromised. I argue that, on pain of circularity, essentialists cannot successfully account for the necessity of essences. In response to the difficulties I raise, I discuss potential solutions on behalf of essentialists and explain why I find none of them compelling. My conclusion on the matter is that the best essentialists can hope for is a view where the necessity of (...) essences is left unexplained. (shrink)

Previous research showed that the repeated approaching of one stimulus and avoiding of another stimulus typically leads to more positive evaluations of the former stimuli. In the current study, we examined whether approach and avoidance training effects on evaluations of neutral stimuli can be modulated by introducing a regularity between the approach-avoidance actions and a positive or negative stimulus. In an AAT task, participants repeatedly approached one neutral non-word and avoided another neutral non-word. Half of the participants also approached a (...) negative fear-conditioned stimulus and avoided a conditioned safe stimulus. The other half of the participants avoided the CS+ and approached the CS−. Whereas participants in the avoid CS+ condition exhibited a typical AAT effect, participants in the approach CS+ condition exhibited a reversed AAT effect. These findings provide evidence for the malleability of the AAT effect when strongly valenced stimuli are approached or avoided. We discuss the practical and theoretical implications of our findings. The aim of the present research was twofold. First, we examined the effects of perceived organizational support on workplace conflict (i.e., relationship conflict and task conflict). Second, we identified one mechanism explaining these relationships, namely failure-related trust. Using a sample of 263 teachers from Belgium, the results of Study 1 indicated that perceived organizational support is negatively related to relationship conflict and is also, unexpectedly, negatively related to task conflict. Furthermore, using a sample of 477 Belgian employees, Study 2 replicated (...) these results and further demonstrated that failure-related trust mediates the negative relationship between perceived organizational support and both types of workplace conflict. Transplant medicine has now dramatically stressed, within man's knowledge of himself, the characteristic of his biological uniqueness. The myth of transparency and truthfulness is the foundation for contemporary theories of information design. Avoiding distortion and ambiguity is the moral imperative of an upstanding data visualizer. Sometimes, though, data that is collected is not accurate enough and the sources use indirect indicators to approach a phenomenon: what to do then, if the need to graphically represent a phenomenon is urgent and necessary? Should the designer wait to have the exact data, or should he indicate a trend, expressing the (...) hypothetical status of his statement? Can data visualization be designed to express doubt rather than to inform about facts? This essay will deal with the forms of expression of uncertainty in infographics. It will consider the designer as both an observer and a translator, whose position of neutrality is only one of the possible realms of discourse. In general, it will focus on the forms of visual expression of a self-criticizing mood in quantitative research today and it will explore the ways in which data that is not meaningful in statistical terms can become meaningful in semiotic terms. This panorama urges us to renew the (...) theoretical lenses through which we look at an adolescence that is completely different from previous ones with regard to its relationship with sexuality. The period we are living through is one of restoration, reminiscent of the 1930s. In a bellicose atmosphere that has proved conducive to an unstable reassertion of imperialism, we have seen a renewed dramatisation of questions of demography and migration, leading to an ethnicisation of social and political relations. Though Europe actually has a structural need of a migrant labour-force, its States have instituted a differential management in dealing with questions of race, depending on the origins of the migrants and (...) the nature of the jobs involved. The result has been to reduce vast numbers of migrants to the condition of illegal, clandestine workers, while operating as a powerful factor in the aggravated exploitation of labour and, in some instances, as a factor in the collective criminalisation of migrants. What Europe’s former Nation-States are thus witnessing, is the emergence of a citizen-foreigner duality which both occludes and perverts the structure of class relations. (shrink)

Le présent volume propose une cartographie de la recherche linguistique menée au Québec, avec beaucoup de dynamisme, entre 1960 et 2000. Les années 60 correspondent à une phase d'institutionnalisation de la discipline; l'an 2000 représente une rupture symbolique avec le XXe siècle et laisse un laps de temps raisonnable pour prendre un certain recul. Le livre présente ainsi une étude de cas sur l'histoire institutionnelle de la linguistique, qui fait écho à l'ouvrage éponyme Combats pour la linguistique, de Martinet à (...) Kristeva. Essai de dramaturgie épistémologique (2006) de J.-C. Chevalier et P. Encrevé. Les huit chapitres, qui forment l'ouvrage, font état des recherches réalisées pendant quatre décennies en sémantique, en lexicologie, en lexicographie, en phonologie, en grammaire et en sociolinguistique. Ils s'attardent aux filiations de la discipline avec le passé, à ses questionnements, ses acquis et ses legs ainsi qu'à ses limites, ses obstacles et ses déceptions. Ils se veulent un aide-mémoire pour les générations futures. Ce livre constitue de plus un apport aux études épistémologiques consacrées à l'évolution des méthodologies, des courants et des écoles linguistiques au XXe siècle. (shrink)

Cet article présente le contexte général ayant conduit à l’élaboration du Corpus de français parlé au Québec (CFPQ) et les principes méthodologiques ayant présidé à sa confection. Il illustre ensuite l’intérêt que représente cette ressource documentaire pour l’étude de la langue parlée en contexte informel par le biais d’un examen lexico-sémantique de la séquence je sais pas. L’intérêt pour cette séquence vient d’abord d’un constat : celle-ci est particulièrement fréquente dans le corpus pris comme cible. En effet, elle y occupe (...) le premier rang, en terme de fréquence, pour ce qui concerne la présence de trois unités graphiques figurant en contiguïté. Ce constat oriente vers l’idée selon laquelle son degré d’« entrenchment » (c’est-à-dire d’enracinement) doit être grand, qu’elle a toute chance d’être mémorisée en bloc dans un certain nombre de contextes, à la manière des séquences complexes ou expressions (semi-)figées. L’attention se focalise sur des exemples où je sais pas agit à titre d’expression verbale, puis d’expression discursive. (shrink)

ABSTRACTThis paper examines Martin Heidegger’s own interpretation of Being and Time in the Black Notebooks. The opening part addresses Heidegger’s singular notions of “thinking” and “questioning” which suggest a critically reflective stance, but involve an initiatory call to surrender to the hidden powers of Beyng. The second part addresses Heidegger’s lament in the Black Notebooks that Being and Time has not produced a “great enemy”, and his critique of the initial existentialist or “anthropological” receptions of his magnum opus. The third (...) part looks at how Heidegger’s thought moves beyond Being and Time, in light of the central role the thinker assigns to the German people in repeating the first “inception” of preSocratic thought, in order to overcome the legacies of Roman universalism, and what Heidegger as early as 1932 was calling “Jewish Christianity”. The final part addresses Heidegger’s conception of “the history of Beyng” as it is developed in the Black Notebooks of the early 1940s, from out of the account of “temporality” in Being and Time: a “history” in which Heidegger’s postulation of a need for the “annihilation“ ” of the essential enemy of the Volk become the lens through which Heidegger can read the Shoah as the “the peak of self-destruction in history”. L’allergie de Bolzano à l’œuvre de Hegel est un fait souligné dans presque toutes les présentations de sa pensée. Pourtant, conformément à son éthique intellectuelle, il a tenté de se familiariser avec cette philosophie qu’il rejetait plus que toute autre. Il a même, tardivement, reconnu qu’il pouvait s’y trouver des propositions justes. Afin d’affiner la compréhension de la relation de Bolzano à Hegel, on se propose ici de reconstruire la polémique des Trois essais sur Hegel en montrant qu’elle recoupe bien (...) des formes d’anti-hégélianisme devenues canoniques (la critique de la philosophie de l’histoire notamment), mais dans un contexte qui lui est propre et la spécifie. Dans un second temps, on reviendra sur les divergences philosophiques à l’origine de la nécessité pour Bolzano de critiquer un auteur et ses disciples, dans lesquels il voit une forme de régression intellectuelle et sociale, divergences qui imposent et orientent une lecture. Ces divergences sur la compréhension du rôle du langage et de la philosophie, du rapport des représentations au réel, ou sur la définition de l’infini, sont sous-jacentes à la polémique des Trois essais, mais on doit en chercher la formulation dans le reste du corpus bolzanien. The subject of bullying has received widespread academic attention, going back to pioneering studies in the 1970s. However, work in this area has tended to take a gender-neutral/male perspective that does not distinguish between male and female bullying. While retaining a dialogue with generalist literature on the subject, this article seeks to highlight the theoretical and political-social relevance of gender-specific and intersectional approaches when exploring the question of bullying between adolescent girls. Based on the study _Il bullismo femminile a scuola. (...) Un’indagine intersezionale mixed-method_ (Female bullying at school, an intersectional, mixed-method study) – conducted in medium-sized and large cities in northern, central and southern Italy – it proposes a theoretical-analytical framework in which to consider the phenomenon of female bullying in Italian schools.
