Mitochondrial function is key for maintaining cellular health, while mitochondrial failure is associated with various pathologies, including inherited metabolic disorders and age‐related diseases. In order to maintain mitochondrial quality, several pathways of mitochondrial quality control have evolved. These systems monitor mitochondrial integrity through antioxidants, DNA repair systems, and chaperones and proteases involved in the mitochondrial unfolded protein response. Additional regulation of mitochondrial function involves dynamic exchange of components through mitochondrial fusion and fission. Sustained stress induces a selective (...) autophagy – termed mitophagy – and ultimately leads to apoptosis. Together, these systems form a network that acts on the molecular, organellar, and cellular level. In this review, we highlight how these systems are regulated in an integrated context‐ and time‐dependent network of mitochondrial quality control that is implicated in healthy aging. (shrink)

Background: Ontologies and taxonomies are among the most important computational resources for molecular biology and bioinformatics. A series of recent papers has shown that the Gene Ontology (GO), the most prominent taxonomic resource in these fields, is marked by flaws of certain characteristic types, which flow from a failure to address basic ontological principles. As yet, no methods have been proposed which would allow ontology curators to pinpoint flawed terms or definitions in ontologies in a systematic way. Results: We present (...) computational methods that automatically identify terms and definitions which are defined in a circular or unintelligible way. We further demonstrate the potential of these methods by applying them to isolate a subset of 6001 problematic GO terms. By automatically aligning GO with other ontologies and taxonomies we were able to propose alternative synonyms and definitions for some of these problematic terms. This allows us to demonstrate that these other resources do not contain definitions superior to those supplied by GO. Conclusion: Our methods provide reliable indications of the quality of terms and definitions in ontologies and taxonomies. Further, they are well suited to assist ontology curators in drawing their attention to those terms that are ill-defined. We have further shown the limitations of ontology mapping and alignment in assisting ontology curators in rectifying problems, thus pointing to the need for manual curation. (shrink)

This article discusses the future of quality control in an academic publication system that will be largely based on electronic publishing. Information and communication technologies both challenge traditional ways and open remedies for existent problems of present gate-keeping. New forms of ex-ante and of ex-post quality control may partly replace and partly amend peer review, citation indices and quality filters based on the reputation of the publisher. Open peer review, online commenting, rating, access counts and (...) use tracking are evaluated and put in perspective. Refuting the common argument that e-publishing leads to less quality, this paper puts forward scenarios of the future quality control system. Most likely, we shall see mixed systems, combining old and new elements, of different shapes in the different research fields. (shrink)

Physicians and institutional providers face expanding liability exposure today, in spite of state tort reform legislation and public awareness of the costs of malpractice for providers. Standards of practice are evolving rapidly; new medical technologies are being introduced at a rapid rate; information is proliferating as to treatment efficacy, patient risk, and diseases generally. Tort standards mirror this change. As medical standards of care evolve, they provide a benchmark against which to measure provider failure. The liability exposure of physicians is (...) affected by the generation of data, including outcomes data usable to profile physician practice, and statistical data that allows for predictions as to treatment efficacy, and patient prognosis; obligations to inform patients and third parties of risk created by contagious disease and other sources of harm; obligations of physicians to disclose risks that the provider creates for the patient; obligations to disclose conflicts of interest arising out of the practice setting; and duties arising from new epidemiological knowledge. Quality control is an important and integrated part of the scientific system. However, developments in science and society are changing quality control into quality monitoring. New, virtual, and fluid organizational forms are emerging. Common boundaries are seen as being broken down as, for example, in the “triple helix” and the “mode 2” concepts. The stakeholders in science are showing an interest in being more involved in science. They want their evaluation criteria to be used, and (...) they want evaluations to be done on a regular basis because they do not trust the new scientific institutions to be left on their own. Quality monitoring changes the requirements for conducting evaluations as part of quality control. Assessing the societal value of research becomes increasingly important. Finally, quality monitoring emphasizes organizational learning rather than controlling quality in scientific organizations. (shrink)

As is clear to any user of software, quality control of software has not reached the same levels of sophistication as it has with traditional manufacturing. In this paper we argue that this is because insufficient thought is being given to the methods of reasoning under uncertainty that are appropriate to this domain. We then describe how we have built a large-scale Bayesian network to overcome the difficulties that have so far been met in software quality (...) class='Hi'>control. This exploits a number of recent advances in tool support for constructing large networks. We end the paper by describing how the network was validated and illustrate the range of reasoning styles that can be modelled with this tool. (shrink)

Peer review is a practice used in the evaluation of scientific and scholarly papers in order to select papers for publication in scholarly journals. The practice has also been extended to other domains, such as the evaluation of grant proposals, medical practice, book publication, and even to such areas as teaching evaluation. The primary area that has been of interest to sociologists, however, has been publication in scientific journals. The practice is usually understood to have begun in the seventeenth century (...) in the Royal Society in London, but it has also been claimed that there have been precursors to this practice. In the popular mind, peer review in science is a means of “bullet‐proofing” research, as a Wall Street Journal article once put it, that is to say as a guarantee of quality. This paper reviews the published empirical evidence concerning journal peer review consisting of 68 papers, all but three published since 1975. Peer review improves quality, but its use to screen papers has met with limited success. Current procedures to assure quality and fairness seem to discourage scientific advancement, especially important innovations, because findings that conflict with current beliefs are often judged to have defects. Editors can use procedures to encourage the publication of papers with innovative findings such as (...) invited papers, early-acceptance procedures, author nominations of reviewers, structured rating sheets, open peer review, results-blind review, and, in particular, electronic publication. There are an increasing number of cell therapy approaches being studied and employed world‐wide. An emerging area in this field is the use of human pluripotent stem cell (hPSC) products for the treatment of injuries/diseases that cannot be effectively managed through current approaches. However, as with any cell therapy, vast numbers of functional and safe cells are required. Bioreactors provide an attractive avenue to generate clinically relevant cell numbers with decreased labour and decreased batch to batch variation. Yet, current methods (...) of performing quality control are not readily scalable to the cell densities produced during bioreactor scale‐up. One potential solution is the application of inducible/controllable suicide genes that can trigger cell death in unwanted cell types. These types of approaches have been demonstrated to increase the quality and safety of the resultant cell products. In this review, we will provide background on these approaches and how they could be used together with bioreactor technology to create effective bioprocesses for the generation of high quality and safe hPSCs for use in regenerative medicine approaches. tRNA nucleotidyltransferase adds the invariant CCA‐terminus to the tRNA 3′‐end, a central step in tRNA maturation. This CCA‐adding enzyme is a specialized RNA polymerase that synthesizes the CCA sequence at high fidelity in all kingdoms of life. Recently, an additional function of this enzyme was identified, where it generates a specific degradation tag on structurally unstable tRNAs. This tag consists of an additional repeat of the CCA triplet, leading to a 3′‐terminal CCACCA sequence. In order to explain how the enzyme (...) catalyzes this extended polymerization reaction, Kuhn et al. solved a series of co‐crystal structures of the CCA‐adding enzyme from Archaeoglobus fulgidus in complex with different tRNA substrates. They show that the enzyme forces a bound unstable tRNA to refold the acceptor stem for a second round of CCA‐addition, while stable transcripts are robust enough to resist this isomerization. In this review, we discuss how the CCA‐adding enzyme uses a simple yet very elegant way to scrutinize its substrates for sufficient structural stability and, consequently, functionality. (shrink)

During the production process, production companies need to monitor the finished products and ensure their quality, which imposes on them the appointment of inspectors for auditing, and this appointment costs the company amounts that affect the general profit, so it strives to make this cost as low as possible and that the audit process is carried out with high accuracy because in case that the finished products do not conform to the basic specifications of the product, the company is (...) required to pay a fine. In this research, we will formulate the text of the problem appropriately for such a case and then we will build the appropriate mathematical model through which we can obtain the lowest possible cost of inspection, and we will present the study using classical values and neutrosophic values, and we will demonstrate them through an example that shows us the difference between them. Science is about evaluation, persuasion and logic. In scientific debate, scientists collectively evaluate theories by persuading each other in regard to epistemological qualities such as deduction and fact. There is, however, a flaw intrinsic to evaluation-by-persuasion: an individual can attempt and even succeed in persuading others by asserting that their reasoning is logical when it is not. This is a problem since, from an epistemological perspective, it is not always transparent nor obvious when a persuasive assertion is actually deductively warranted. (...) Empirical research upon reasoning, indeed, supports the notion that assertions are often strongly persuasive for reasons other than their logic. The unreliability of the link between persuasion and logic raises an important methodological issue: how do scientists debate in a manner such that claimed but false “logical” arguments are ignored, and only warranted arguments get to determine theory preference? This need for soundness in debate is a particularly important epistemological concern in cases where the deductive qualities of persuasive argument are not overt, and so cannot be directly checked --such as when they are founded upon Bayesianism probabilistic coherence. The argument presented here is that scientists make the qualities of probabilistic and nonprobabilistic inference sound (and so warranted) through how they organize their debate. Scientists, I argue, abide by “epistemological quality control practices” that limit the persuasive power of unsound arguments upon theory evaluation. Examples of such debate quality control practices are publicness, clarity, openness to criticism, and the collective promotion of attempts at theory conjecture and refutation. Methodologically, these quality control practices are extralogical since they do not directly provide scientific inferences with additional logical warrantedness. They function instead in science to generate an epistemological evaluative environment in which persuasiveness is due, and only due, to logic (i.e. sound). Their methodological role is therefore to make what is warranted de papyri (in our principles of rationality – epistemologically competence) also what is persuasive in evaluation and debate de cognitio (in our exercise of such principles – epistemologically performance). Several limitations exist upon the soundness of Bayesian inferential coherence – surreptitious revision, logical omniscience, uncertain evidence, old evidence, and new hypotheses. Bayesianism, as a result, can only exist if it is pursued in a debate that is regulated by quality control practices over its inferences (for instance, practices that ensure there is autonomy of inference, diligence of inference, probity of evidence, auditability of inference and assiduousness of conjecture). In this context, one can reinterpret Popper’s concern with criticism, openness, refutation and conjecture, as deriving not (as he thought) directly from the needs of logic, but, indirectly, from the need of scientists to create epistemological soundness. Two major science policy issues are the integration of knowledge across academic disciplines and the accountability of science to society. Instead of adding new or external criteria for research evaluation, I argue, these goals can be pursued by subjecting disciplinary priorities and procedures to broader scrutiny from the rest of academia. From a social epistemological perspective, the paper discusses interdisciplinarity as a mode of epistemic accountability across disciplinary boundaries, which promises to make academia more than the sum of its disciplinary (...) parts. Drawing on discussions of interdisciplinarity and accountability in knowledge production, as well as on empirical findings of the evaluation of research proposals, the paper unpacks the notion of academic accountability into three dimensions—the recipients, contents, and practices of accountability—and illustrates the difference interdisciplinarity makes in each dimension. The somewhat old-fashioned concept of philosophical categories is revived and put to work in automated ontology building. We describe a project harvesting knowledge from Wikipedia’s category network in which the principled ontological structure of Cyc was leveraged to furnish an extra layer of accuracy-checking over and above more usual corrections which draw on automated measures of semantic relatedness.

This paper investigates the socio-technical construction, quality control, and coordination of the credence quality attribute “halal” throughout the halal meat chain. The paper is framed within Actor-Network Theory and economic Conventions Theory. Islamic dietary laws or prescriptions, and how these are translated into production and processing standards using a HACCP-like approach, are discussed. Current halal quality coordination is strongly based on civic and domestic logics in which Muslim consumers prefer transacting with Muslim butchers, that is, individuals (...) of known reputation with similar moral and religious obligations. The HACCP-like approach with identification of critical halal control points, as presented in this paper, fits with the industrial quality convention mechanism and ideally yields guaranteed and trustworthy halal credence quality, eventually marked by a halal meat label. The socio-technical construction of halal credence quality, for example with respect to ritual slaughter, and the quality coordination mechanism aimed at reducing halal quality uncertainty among Muslim consumers, for example through labeling, are identified as key attention points in the future research agenda.
