The use of super‐resolution imaging techniques in cell biology has yielded a wealth of information regarding cellular elements and processes that were invisible to conventional imaging. Focusing on images obtained by stimulated emission depletion (STED) microscopy, we discuss how the new high‐resolution data influence the ways in which we use and interpret images in cell biology. Super‐resolution images have lent support to some of our current hypotheses. But, more significantly, they have revealed unexpectedly complex processes (...) that cannot be accounted for by the simpler models based on diffraction‐limited imaging. The super‐resolution imaging data challenge cell biologists to change their theoretical framework, by including, for instance, interpretations that describe multiple functions, functional errors or lack of function for cellular elements. In this context, we argue that descriptive research using super‐resolution microscopy is now as necessary as hypothesis‐driven research. (shrink)

The recent 2014 Nobel Prize in chemistry honored an era of discoveries and technical advancements in the field of super‐resolution microscopy. However, the applications of diffraction‐unlimited imaging in biology have a long road ahead and persistently engage scientists with new challenges. Some of the bottlenecks that restrain the dissemination of super‐resolution techniques are tangible, and include the limited performance of affinity probes and the yet not capillary diffusion of imaging setups. Likewise, super‐resolution microscopy has (...) introduced new paradigms in the design of projects that require imaging with nanometer‐resolution and in the interpretation of biological images. Besides structural or morphological characterization, super‐resolution imaging is quickly expanding towards interaction mapping, multiple target detection and live imaging. Here we review the recent progress of biologists employing super‐resolution imaging, some pitfalls, implications and new trends, with the purpose of animating the field and spurring future developments. Recently developed super‐resolution microscopy techniques are changing our understanding of lipid rafts and membrane organisation in general. The lipid raft hypothesis postulates that cholesterol can drive the formation of ordered domains within the plasma membrane of cells, which may serve as platforms for cell signalling and membrane trafficking. There is now a wealth of evidence for these domains. However, their study has hitherto been hampered by the resolution limit of optical microscopy, making the definition of their properties (...) problematic and contentious. New microscopy techniques circumvent the resolution limit and, for the first time, allow the fluorescence imaging of structures on length scales below 200 nm. Three‐dimensional structured illumination microscopy (3D‐SIM) has opened up new possibilities to study nuclear architecture at the ultrastructural level down to the ∼100 nm range. We present first results and assess the potential using 3D‐SIM in combination with 3D fluorescence in situ hybridization (3D‐FISH) for the topographical analysis of defined nuclear targets. Our study also deals with the concern that artifacts produced by FISH may counteract the gain in resolution. We address the topography of DAPI‐stained DNA in nuclei before and (...) after 3D‐FISH, nuclear pores and the lamina, chromosome territories, chromatin domains, and individual gene loci. We also look at the replication patterns of chromocenters and the topographical relationship ofXist‐RNA within the inactive X‐territory. These examples demonstrate that an appropriately adapted 3D‐FISH/3D‐SIM approach preserves key characteristics of the nuclear ultrastructure and that the gain in information obtained by 3D‐SIM yields new insights into the functional nuclear organization. This paper describes a Bayesian method for constructing a super-resolved surface model by combining information from a set of images of the given surface. We develop the theory and algorithms in detail for the 2-D reconstruction problem, appropriate for the case where all images are taken from roughly the same direction and under similar lighting conditions. We show the results of this 2-D reconstruction on Viking Martian data. These results show dramatic improvements in both spatial and gray-scale resolution. (...) The Bayesian approach uses a neighbor correlation model as well as pixel data from the image set. Ever since its foundations were laid nearly a century ago, quantum theory has provoked questions about the very nature of reality. We address these questions by considering the universe—and the multiverse—fundamentally as complex patterns, or mathematical structures. Basic mathematical structures can be expressed more simply in terms of emergent parameters. Even simple mathematical structures can interact within their own structural environment, in a rudimentary form of self-awareness, which suggests a definition of reality in a mathematical structure as simply the complete (...) structure. The absolute randomness of quantum outcomes is most satisfactorily explained by a multiverse of discrete, parallel universes. Some of these have to be identical to each other, but that introduces a dilemma, because each mathematical structure must be unique. The resolution is that the parallel universes must be embedded within a mathematical structure—the multiverse—which allows universes to be identical within themselves, but nevertheless distinct, as determined by their position in the structure. The multiverse needs more emergent parameters than our universe and so it can be considered to be a superstructure. Correspondingly, its reality can be called a super-reality. While every universe in the multiverse is part of the super-reality, the complete super-reality is forever beyond the horizon of any of its component universes. (shrink)

Pointillistic based super‐resolution techniques, such as photoactivated localization microscopy (PALM), involve multiple cycles of sequential activation, imaging, and precise localization of single fluorescent molecules. A super‐resolution image, having nanoscopic structural information, is then constructed by compiling all the image sequences. Because the final image resolution is determined by the localization precision of detected single molecules and their density, accurate image reconstruction requires imaging of biological structures labeled with fluorescent molecules at high density. In such image (...) datasets, stochastic variations in photon emission and intervening dark states lead to uncertainties in identification of single molecules. This, in turn, prevents the proper utilization of the wealth of information on molecular distribution and quantity. A recent strategy for overcoming this problem is pair‐correlation analysis applied to PALM. Using rigorous statistical algorithms to estimate the number of detected proteins, this approach allows the spatial organization of molecules to be quantitatively described. (shrink)

Resolution and cut-free LK are the most popular propositional systems used for logical automated reasoning. The question whether or not resolution and cut-free LK have the same efficiency on the system of CNF formulas has been asked and studied since 1960 425–467). It was shown in Cook and Reckhow, J. Symbolic Logic 44 36–50 that tree resolution has super-polynomial speed-up over cut-free LK. Naturally, the current issue is whether or not resolution and cut-free LK expressed (...) as directed acyclic graphs have the same efficiency. In this paper, we introduce a new algorithm to eliminate atomic cuts and show that cut-free LK polynomially simulates resolution when the input formula is expressed as a k-CNF formula. As a corollary, we show that regular resolution does not polynomially simulate cut-free LK . We also show that cut-free LK polynomially simulates regular resolution. (shrink)

A novel approach to quantization is shown to allow for superpositions of the cosmological constant in isotropic and homogeneous mini-superspace models. Generic solutions featuring such superpositions display: i) a unitary evolution equation; ii) singularity resolution; iii) a cosmic bounce. Explicit cosmological solutions are constructed. These exhibit characteristic bounce features including a ‘super-inflation’ regime with universal phenomenology that can naturally be made to be insensitive to Planck-scale physics.

Long‐term potentiation (LTP) of excitatory synapses is a leading model to explain the concept of information storage in the brain. Multiple mechanisms contribute to LTP, but central amongst them is an increased sensitivity of the postsynaptic membrane to neurotransmitter release. This sensitivity is predominantly determined by the abundance and localization of AMPA‐type glutamate receptors (AMPARs). A combination of AMPAR structural data, super‐resolution imaging of excitatory synapses, and an abundance of electrophysiological studies are providing an ever‐clearer picture of how (...) AMPARs are recruited and organized at synaptic junctions. Here, we review the latest insights into this process, and discuss how both cytoplasmic and extracellular receptor elements cooperate to tune the AMPAR response at the hippocampal CA1 synapse. The mechanical properties of vertebrate cells are largely defined by the system of intermediate filaments (IF). As part of a dense network, IF polymers are constantly rearranged and relocalized in the cell to fulfill their duty as cells change shape, migrate, or divide. With the development of new imaging technologies, such as photoconvertible proteins and super‐resolution microscopy, a new appreciation for the complexity of IF dynamics has emerged. This review highlights new findings about the transport of IF, the (...) remodeling of filaments by a process of severing and re‐annealing, and the subunit exchange that occurs between filament precursors and a soluble pool of IF. We will also discuss the unique dynamic features of the keratin IF network. Finally, we will speculate about how the dynamic properties of IF are related to their functions. (shrink)

Embryos and microscopes share a long, remarkable history and biologists have always been intrigued to watch how embryos develop under the microscope. Here we discuss the advances in microscopy which have greatly influenced our current understanding of embryogenesis. We highlight the evolution of microscopes and the optical technologies that have been instrumental in studying various developmental processes. These imaging modalities provide mechanistic insights into the dynamic cellular and molecular events which drive lineage commitment and morphogenetic changes in the developing embryo. (...) We begin the journey with a brief history of microscopy to study embryos. First, we review the principles and optics of light, fluorescence, confocal, and electron microscopy which have been key techniques for imaging cellular and molecular events during embryonic development. Next, we discuss recent key imaging modalities such as light‐sheet microscopy, which are suitable for whole embryo imaging. Further, we highlight imaging techniques like multiphoton and super resolution microscopy for beyond light diffraction limit, high resolution imaging. Lastly, we review some of the scattering‐based imaging methods and techniques used for imaging human embryos. (shrink)

The pore‐forming BCL‐2 family proteins are effectors of mitochondrial poration in apoptosis initiation. Two atypical effectors—BOK and truncated BID (tBID)—join the canonical effectors BAK and BAX. Gene knockout revealed developmental phenotypes in the absence the effectors, supporting their roles in vivo. During apoptosis effectors are activated and change shape from dormant monomers to dynamic oligomers that associate with and permeabilize mitochondria. BID is activated by proteolysis, BOK accumulates on inhibition of its degradation by the E3 ligase gp78, while BAK and (...) BAX undergo direct activation by BH3‐only initiators, autoactivation, and crossactivation. Except tBID, effector oligomers on the mitochondria appear as arcs and rings in super‐resolution microscopy images. The BH3‐in‐groove dimers of BAK and BAX, the tBID monomers, and uncharacterized BOK species are the putative building blocks of apoptotic pores. Effectors interact with lipids and bilayers but the mechanism of membrane poration remains elusive. I discuss effector‐mediated mitochondrial poration. (shrink)

The centriole is a widely conserved organelle required for the assembly of centrosomes, cilia, and flagella. Its striking feature – the nine‐fold symmetrical structure, was discovered over 70 years ago by transmission electron microscopy, and since elaborated mostly by cryo‐electron microscopy and super‐resolution microscopy. Here, we review the discoveries that led to the current understanding of how the nine‐fold symmetrical structure is built. We focus on the recent findings of the centriole structure in high resolution, its assembly (...) pathways, and its nine‐fold distributed components. The nuclear export of mRNA through the nuclear pore complex (NPC) is a process required for the healthy functioning of human cells, making it a critical area of research. However, the geometries of mRNA and the NPC are well below the diffraction limit of light microscopy, thereby presenting significant challenges in evaluating the discrete interactions and dynamics involved in mRNA nuclear export through the native NPC. Recent advances in biotechnology and single‐molecule super‐resolution light microscopy have enabled researchers to (...) gain granular insight into the specific contributions made by discrete nucleoporins in the nuclear basket of the NPC to the export of mRNA. Specifically, by expanding upon the docking step facilitated by the protein TPR in the nuclear basket as well as identifying NUP153 as being the primary nuclear basket protein initiating export through the central channel of the NPC. We offer a new proposal for cosmic singularity resolution based upon a quantum cosmology with a unitary bounce. This proposal is illustrated via a novel quantization of a mini-superspace model in which there can be superpositions of the cosmological constant. This possibility leads to a finite, bouncing unitary cosmology. Whereas the usual Wheeler–DeWitt cosmology generically displays pathological behaviour in terms of non-finite expectation values and non-unitary dynamics, the finiteness and unitarity of our model are formally guaranteed. For classically singular (...) models with a massless scalar field and cosmological constant, we show that well-behaved quantum observables can be constructed and generic solutions to the universal Schrodinger equation are singularity-free. Key features of the solutions of our model include a cosmic bounce due to quantum gravitational effects, a well-defined FLRW limit far from the bounce, and a super-inflationary epoch in the intermediate region. Furthermore, our model displays novel features including: i) superpositions of values of the cosmological constant; ii) a non-zero scattering length around the big bounce; and iii) bound ‘Efimov universe’ states for negative cosmological constant. The last feature provides a new platform for quantum simulation of the early universe. A companion paper provides detailed interpretation and analysis of particular cosmological solutions. (shrink)

The article contains an exposition of the moral philosophy of J. P. Sartre as well as a trial of its evaluation. The author presents the social basis and main theses of Sartre’s.philosophical system and stresses the questions of social conditioning, real contents, and functions of the situational ethics of Sartre. According to the author, the situational ethics of Sartre, being an expression of feelings of intellectuals, middle-class, and students in the period of violent changes in our civilization, simply describes a (...) certain type of man, peculiar to these conditions and this time. Therefore, this type cannot be considered a definite universal construction of a human subject. Pessimism, characteristic for the existentialism, as an expression of.certain social and political conditions, can be easily surmounted, provided that a possibility of definite social transformations can be demonstrated. This gives evidence of a positive alternative which proposes development of social democracy, elimination of the remnants of worship of the individual, as well as a more and more full realization of social humanism, that are limiting the experiments of existentialism. Together with an increasing conviction of a possibility to surmount and to abolish the capitalistic alienation and together with the development of socialist democracy, when marxist thinking comprises more and more effectively all aspects of human fates, the social basis of existentialism and its influence are losing ground. On the other hand, whatever might be said about arbitrariness and subjectivism of many theses of existentialism, it has raised many essential questions of particular importance in our time. These are: 1) subjective aspect of human freedom in situations of a choice of an action; 2) responsibility of an individual to himself, other people, and history; 3) contradiction between general systems of valuation and norms and of a definite situation in which a choice is taken; 4) danger of a non-authentic apparent communication; 5) danger of a lost of personality in conditions of consumptionist civilisation; 6) necessity of responsibility and engagement; 7) necessity of forbearance, tolerance and respecting the subjective world of other persons; 8) existence of conflict situations. According to the author, a resolution of the above problems, which are important for a modern man, can be done only basing on marxism which should be conceived in a creative way. It is necessary, however, to reject arbitrary assumptions of the existentialist metaphysics, ethical formalism, and, above all, ahistorical conception of freedom. The opinions of Sartre that the, good a priori cannot exist, that everybody should determine his own existence, and that in each situation he should maintain his creative attitude and moral alertness, are in a certain sense and within certain limits right and productive. However, depriving people of a possibility to build any system of values and norms of a super-subjective character is a risky thing and may lead to moral relativity and nihilism. The social function of the existentialism is also a double one. On the one hand, existentialism, which demonstrates a super-historical and unchangeable character of the drama of human existence, in some way confirms and justifies the social system submitted to critic. On the other hand, as it states a moral and idealistic outsidership, bareness, and loneliness of a human individual in a modern bourgeois 1society, existentialism presents an important ethical indictment directed to the capitalistic society and indirectly shows a necessity to create such relations which would remove the tragic gap between individual and society. This is the reason of a peculiar paradox that for same people the existentialism is a point of departure from the files of the revolutionary movement, and for others a point which leads into marxism – cum duo faciunt idem, non est idem. This paper identifies three sets of problems of a specific ethico-political type, generated by the interrelationship between ethics and politics in the areas of world justice and global politics. One instance in which this interrelationship is tested is that of the conflict of duties and values as it appears in the particular domain of the relations amongst sovereign nation states as well as between them and other social groups. Following the general Introduction, the main body of the paper contains the (...) following three sections. (1) Without elaborating on the detailed mechanics of Kant's theory of perpetual peace, Section II discusses criticisms against the Kantian version of world peace in the context of which the first problem is encountered. The problem identified here is the ambiguity as to whether the establishment of a federated kind of peaceful co-existence between sovereign nations depends on the moral improvement of mankind or whether it is the other way round--i.e. whether ethical progress is a necessary presupposition or, rather, a consequence of political peace. That is, what is here identified as worrying is that the direction of dependence is not clear in such a proposal. Furthermore, such an approach, when applied to certain cases of inter-state differences, yields a rather alarming result as to the de-politicization of national states (Section II.2). (2) Yet such a type of international federalism requires a certain, peculiar, kind of legal enforcement of order without recourse to a supra state. So a re-examination of the notion of a legal order in general is needed. Given these requirements, Section III moves on to discuss moral conflict from the standpoint of relativism as it appears in the ethical and in the legal spheres. The second of our problems, identified in this section, is that, before discussing world justice we must first acknowledge the special relation and asymmetry between ethical standards and legal rules required by such a world order. (3) Finally, Section IV asks whether, given the above two problems, it is possible to envisage theoretically the logical possibility of moral conflict-resolution by postulating unchanging and encompassing super-norms of conduct. These special norms, if possible, would be able to direct action unequivocally when confronted with ethical or other dilemmas of duties (involving equally valid but conflicting demands among sovereign nations). This is the third problem. The Conclusion offers an evaluation of the whole discussion. These three ethico-political problems are shown to be interrelated. The overall thrust of the discussion is that it points to a number of philosophical difficulties in understanding the changing role of national states in a globalized political, economic and cultural environment. Accordingly, such difficulties have repercussions for any moral criticism of issues of world justice and global politics. This paper outlines a formal recursive wager resolution calculus (WRC) that provides a novel conceptual framework for sentential logic via bridge rules that link wager resolution with truth values. When paired with a traditional truth-centric criterion of logical soundness WRC generates a sentential logic that is broadly truth-conditional but not truth-functional, supports the rules of proof employed in standard mathematics, and is immune to the most vexing features of their traditional implementation. WRC also supports a novel probabilistic criterion (...) of logical soundness, the fair betting probability criterion (FBP). It guarantees that the conclusion of an FBP-valid argument is at least as credible as a conjunction of premises, and also that the conclusion is true if the premises are. In addition, WRC provides a platform for a novel non-probabilistic, computationally simpler criterion of logical soundness – the criterion of Super-validity - that issues the same logical appraisals as FBP, and hence the same guarantees. This volume offers an English translation of John of St. Thomas's Cursus theologicus I, question I, disputation 2. In this particular text, the Dominican master raises questions concerning the scientific status and nature of theology. At issue, here, are a number of factors: namely, Christianity's continual coming to terms with the "Third Entry" of Aristotelian thought into Western Christian intellectual culture - specifically the Aristotelian notion of 'science' and sacra doctrina's satisfaction of those requirements - the Thomistic-commentary tradition, and the (...) larger backdrop of the Iberian Peninsula's flourishing "Second Scholasticism." In this latter context, John of St. Thomas applies the theological principles of Thomas Aquinas to the Scholastic disputes preoccupying Thomist, Franciscan, and Jesuit theologians, such as Cajetan, Bañez, Luis de Molina, Vazquez, Suárez - to name only a few - in a tour de force of theological thinking throughout the entire period of Scholasticism. In the process - and not insignificantly - the status quaestionis of theology's scientific character is clearly framed and answered according to John's satisfaction. Key to John of St. Thomas's resolution of the question is his understanding of the continuity of the power of human reason with the super-intelligibility of divine revelation spelled out in terms of what he calls "virtual revelation." This text presented in this volume is a quintessential example of the deep and abiding harmony that flourished between faith and reason as well as grace and nature within the golden era of Baroque Scholasticism. The book is a critical analysis of the work of Max Weber, Emile Durkheim and Karl Marx. It focuses on their separate analyses of the role of law in society, pointing out their faults and errors, and the resultant impact on modern social science. The author takes issue with Weber's work on rationality, with Durkheim's work on repressive and restitutive law, and with Marx's work on social justice and law as part of the super-structure. In each section of the (...) book he shows the implications that flow from a re-assessment and re-interpretation of their work for an understanding of society. Globalization is a natural process. It has a number of advantages & disadvantages, causes many questions and problems, which can hardly sometimes be solved by countries independently. These problems can only be solved by the world community. One of these problems is to maintain the concrete communities identity. Is it possible to keep the unique culture of different ethnos, language, traditions in the globalizing world? Or as some researchers consider, there is a tendency to the formation of the so called (...) super ethnos? In such conditions tolerance takes on special significance as a certain means of activity, as social behavior model, as a norm of behavior for each member of the society. Tolerance as a behavior model on one hand should be worked out by a state legislative body, from the other it should be a norm of behavior for each member of the society. The basis of the tolerant behavior is the principle of mutual understanding.It can be considered to be a moral imperative for the resolution of conflict situations. The relevance & importance of tolerance & creating tolerant relations is a characteristic feature of the democratic personality. (shrink)

We analyze a model of conflict with endogenous choice of effort, where subsets of the contenders may force the resolution to be sequential: First the alliance fights it out with the rest and – in case they win – later they fight it out among themselves. For three-player games, we find that it will not be in the interest of any two of them to form an alliance. We obtain this result under two different scenarios: equidistant preferences with varying (...) relative strengths, and varying vicinity of preferences with equal distribution of power. We conclude that the commonly made assumption of super-additive coalitional worth is suspect. (shrink)

Yet ethical theorists seem perennially to be drawn off into lengthy debates about side issues, with the result that the important questions are lost in the confusion and are dealt with more or less absent-mindedly. Amid the endless arguments about naturalism, the definability of good, the verifiability of ethical judgments, the scientific, extra-scientific or super-scientific status of ethics, it is difficult to locate the fundamental issues, the issues which all of us resolve, reflectively or absentmindedly, in one way or (...) another, and the resolution of which influences profoundly, for better or worse, the directions taken in dealing with specific moral, social, and political problems. There are, nevertheless, important problems in ethics; and different solutions of them have extensive consequences for practice. (shrink)

Aboitiz et al. describe a hypothesis on the origin of the isocortex. They propose the reptilian dorsal cortex to be the ancestral brain structure to the mammalian isocortex. But why did the dorsal cortex expand in mammals and not in reptiles? A change in development may have provided the mammalian cortex with the ability to organize and therefore the potential to expand.
