In “Les deux pas de Lancelot,” the late Eugène Vinaver argued for the restoration of two lines that are lacking in the text of the Chevalier de la Charrete as transcribed by Guiot, the well-known copyist of one of the best surviving manuscripts of Chrétien de Troyes's collected works. Following is the “complete” passage with brackets around the two lines missing in Guiot.

The CRISPR-Cas systems of bacterial and archaeal adaptive immunity have become a household name among biologists and even the general public thanks to the unprecedented success of the new generation of genome editing tools utilizing Cas proteins. However, the fundamental biological features of CRISPR-Cas are of no lesser interest and have major impacts on our understanding of the evolution of antivirus defense, host-parasite coevolution, self versus non-self discrimination and mechanisms of adaptation. CRISPR-Cas systems present the best known case in point (...) for Lamarckian evolution, i.e. generation of heritable, adaptive genomic changes in response to encounters with external factors, in this case, foreign nucleic acids. CRISPR-Cas systems employ multiple mechanisms of self versus non-self discrimination but, as is the case with immune systems in general, are nevertheless costly because autoimmunity cannot be eliminated completely. In addition to the autoimmunity, the fitness cost of CRISPR-Cas systems appears to be determined by their inhibitory effect on horizontal gene transfer, curtailing evolutionary innovation. Hence the dynamic evolution of CRISPR-Cas loci that are frequently lost and acquired by archaea and bacteria. Another fundamental biological feature of CRISPR-Cas is its intimate connection with programmed cell death and dormancy induction in microbes. In this and, possibly, other immune systems, active immune response appears to be coupled to a different form of defense, namely, “altruistic” shutdown of cellular functions resulting in protection of neighboring cells. Finally, analysis of the evolutionary connections of Cas proteins reveals multiple contributions of mobile genetic elements to the origin of various components of CRISPR-Cas systems, furthermore, different biological systems that function by genome manipulation appear to have evolved convergently from unrelated MGE. The shared features of adaptive defense systems and MGE, namely the ability to recognize and cleave unique sites in genomes, make them ideal candidates for genome editing and engineering tools. G.E. Moore noticed the oddity of statements like: “It's raining, but I don't believe it.” This oddity is often seen as analogous to the oddity of believing akratically, or believing what one believes one should not believe, and has been appealed to in denying the possibility of akratic belief. I describe a Belief Akratic's Paradox, analogous to Moore's paradox and centered on sentences such as: “I believe it's raining, but I shouldn't believe it.” I then defend the possibility of akratic (...) belief against appeals to this analogy, arguing both that akratic belief does not require belief-akratic-paradoxical belief, and that the latter is importantly different from Moorean belief. I conclude by considering the implications of these arguments for an understanding of both Moorean and akratic belief. (shrink)

The Hardy–Weinberg equilibrium has been argued by Sober, Stephens and others to represent the zero-force state for evolutionary biology understood as a theory of forces. I investigate what it means for a model to involve forces, developing an explicit account by defining what the zero-force state is in a general theoretical context. I use this account to show that Hardy–Weinberg equilibrium is not the zero-force state in biology even in the contexts in which it applies, and argue based on this (...) that drift should not be understood as an evolutionary force. This study investigated students’ perceptions of ethical organizational climates, attitudes towards ethical issues, and the perceived relationship between ethical behavior and success in business organizations. Comparisons were made between the attitudes of these future managers with previously published studies of Russian managers’ attitudes. A survey of 100 business students in three Moscow universities showed that their attitudes toward ethical behavior were more negative than those of Russian managers. No significant differences were found in the perceptions or attitudes of students who (...) had attended an ethics course and those that did not. Applying new theories about rights to pressing social issues, A Holistic Approach to Rights suggests major changes are needed in the ways we think about rights and formulating social policy.

Husserl and others have spent a great deal of time writing introductions to phenomenology, and in trying to explain its nature. One thing that becomes clear from these efforts is that phenomenology claims to have a method for analyzing the essential structures of “mental events”. This raises the possibility of phenomenology turning back on itself, for surely the analysis itself must consist of “mental events”. Hence, at some point in its investigations, phenomenology itself could become what phenomenologists seek to analyze. (...) Husserl foresaw this when he suggested the possibility that:transcendental phenomenology itself then becomes a theme for constitutional and critical inquiry at a higher level, for the sake of conferring on it the highest dignity of genuineness: the ability to justify itself down to its roots.What becomes apparent here is that phenomenology's self-reflection is necessary, not merely possible: without self-reflection phenomenology would lack final “genuineness” or the “ability to justify itself down to its roots”. Unfortunately while Husserl, here and in other places, signals the necessity of this sort of reflection, he nowhere seems actually to carry it out at length. (shrink)

Houston, Texas, is a major U.S. city with, like many, a growing aging population. The purpose of this study and ultimate book chapter is to explore the views and perceptions of long-term care (LTC) residents, family members and health care providers. Individuals primarily in independent living and group residential settings were interviewed and studied. Questions emphasized the concepts of personal autonomy, dignity, quality and location of care and decision making. Although a small sample of participants were involved, consistency was noted. (...) Keeping the elderly in caring and loving home situations (theirs or family) was most preferred. Personal choice and independence were emphasized by residents, but family members needed to act as advocates. We also noted that the legal system emphasizes family control over individual decision making as competency declines with aging. Optimal personal decision making in the residents' best interest also became more difficult with loss of individual mental capacity. (shrink)

During tumor evolution, cancer cells use the tumor‐stroma crosstalk to reorganize the microenvironment for maximum robustness of the tumor. The success of immune checkpoint therapy foretells a new cancer therapy paradigm: an effective cancer treatment should not aim to influence the individual components of super complex intracellular interactomes (molecular targeting), but try to disrupt the intercellular interactions between cancer and stromal cells, thus breaking the tumor as a whole. Arguments are provided in favor of a hypothesis that such interactions include (...) formation of synapse‐like structures (interfaces) where the interacting cells are located at a distance of ∼10–15 nm. Within these interfaces, molecules initiating and strengthening the interaction are organized, and allow optimum cross‐signaling; a very confined intercellular space facilitates the concentration of secreted cytokines, enhancing the paracrine cross‐communication. These features of tumors form a druggable cancer hallmark the tumor‐stroma symbiotic crosstalk—which represents a new target for efficient cancer drug discovery. (shrink)
