Glycosylation refers to the co‐ and post‐translational modification of protein and lipids by monosaccharides or oligosaccharide chains. The surface of mammalian cells is decorated by a heterogeneous and highly complex array of protein and lipid linked glycan structures that vary significantly between different cell types, raising questions about their roles in development and disease pathogenesis. This review will begin by focusing on recent findings that define roles for cell surface protein and lipid glycosylation in pluripotent stem cells and their functional (...) impact during normal development. Then, we will describe how patient derived induced pluripotent stem cells are being used to model human diseases such as congenital disorders of glycosylation. Paperback $40.28, isbn 978-1-350-29704-3.The editors Steven Burik, Robert Smid and Ralph Weber, who have all made important revisions (...) to the theory of comparative philosophical method, have pulled together the contributions of ten other influential scholars in the field and their own work into this much-needed collection of essays. There has been much written about the methodology of comparative philosophy in the last hundred years. More recent reflections in this field have been strongly fueled by post-colonial evaluations as well as a proliferation of methodological approaches. Nonetheless, some consensus on what methods the field should adopt and to what ends still needs overt discussion. The editors argue in an Introduction and Epilogue to the volume that, now faced with the "obstacles" of often conflicting principles and aims of different methodological approaches which leads to their frequent mutual rejection, there is need for a wide acceptance of pluralism in the methodology of comparative philosophy (pp. 4–5). This methodological pluralism, for the editors, should be understood in two senses. Firstly, there should be a broad-based "methodological commitment to pluralism," which amounts to an acceptance that there is an existing plurality of philosophical traditions in the world which have very different foundational insights and worldviews (p. 254). Secondly, the fact that the plurality of philosophical traditions actually exhibits quite different approaches ought, despite any individual comparativist's preferred method, compel us to accept that this diversity is best dealt with by a plurality of research approaches (pp. 254–55). That is to say that, while the editors both acknowledge and endorse the particular specializations of any and all scholars engaged in intercultural philosophy, they urge each scholar to accept that both a descriptive and prescriptive pluralism of philosophical methodologies are necessary for the continued flourishing of the field. In order to demonstrate this descriptive and prescriptive pluralism, the editors organize the volume into what they call five "constellations" of discussion (p. 8).In the first "constellation" labeled "Necessary Conditions," Robert Neville, Jaap [End Page 1] van Brakel and Lin Ma offer general and detailed frameworks for what sorts of assumptions are needed for any comparative philosophical research to proceed fruitfully. Neville, while happily admitting that we need to be vigilant about biases and other limitations when crossing into other traditions, insists that philosophers need to at once be searching for the truth in any given comparison of ideas, while simultaneously always reflectively constructing the right categories on the basis of which the comparison may be justified and sound (pp. 22–27). Van Brakel and Ma present in their co-authored essay an elaborate blueprint for comparative research. This blueprint, while also conceding that there can never be "any one right interpretation" of any individual text, much less whole traditions, posit a basic "family resemblance principle" which highlights "mutually recognizable human practices" as the basic condition enabling comparative reflection in the first place (pp. 34–39).In the second "constellation," entitled "Generalization and Essentialization," Roger Ames and François Jullien insist in various ways that, while the essentialism that bedeviled previous eras of philosophy in general and comparative philosophy in particular should be rejected, generalizations are still fundamentally necessary to crystalize the most important ways in which traditions are distinct from one another. Ames, defending himself against recent allegations of essentialism in his approach, insists that the field needs to be supported by both a "self-reflective hermeneutical sensitivity" which acknowledges the positionality of the inquirer as well as a commitment to "search within the soil" of, in his case, the Chinese philosophical tradition for its own models of meaning (p. 58). To the latter end, Ames defends accurate generalizations which can help us understand "the language, the customs and the life-forms of a living tradition" since these, contrary to suspicions, aid us in preventing essentialization in comparative work (p. 63). Methods Young healthy heterosexual couples underwent sleep-lab-based polysomnography of two sleeping arrangements: individual sleep and co-sleep. Individual and dyadic sleep parameters (i.e., synchronization of sleep stages) were collected. The latter were assessed using cross-recurrence quantification analysis. Additionally, subjective sleep quality, relationship characteristics, and chronotype were monitored. Data were analyzed comparing co-sleep vs. individual sleep. Interaction effects of the sleeping arrangement with gender, chronotype, or relationship characteristics were moreover tested. Results As compared to sleeping individually, co-sleeping was associated with about 10% (...) more REM sleep, less fragmented REM sleep (p = 0.008), longer undisturbed REM fragments (p = 0.0006), and more limb movements (p = 0.007). None of the other sleep stages was significantly altered. Social support interacted with sleeping arrangement in a way that individuals with suboptimal social support showed the biggest impact of the sleeping arrangement on REM sleep. Sleep architectures were more synchronized between partners during co-sleep (p = 0.005) even if wake phases were excluded (p = 0.022). Moreover, sleep architectures are significantly coupled across a lag of ± 5min. Depth of relationship represented an additional significant main effect regarding synchronization, reflecting a positive association between the two. Neither REM sleep nor synchronization was influenced by gender, chronotype, or other relationship characteristics. Conclusion Depending on the sleeping arrangement, couple's sleep architecture and synchronization show alterations that are modified by relationship characteristics. We discuss that these alterations could be part of a self-enhancing feedback loop of REM sleep and sociality and a mechanism through which sociality prevents mental illness. (shrink)

Background:Moral distress is a significant concern for nurses as it can lead to burnout and intentions to leave the profession. Pediatric nurses encounter stressful and ethically challenging situations when they care for suspected victims of child maltreatment. Data on pediatric nurses’ moral distress are limited, as most research in this field has been done in adult inpatient and intensive care units.Aim:The purpose of this study was to describe pediatric nurses’ moral distress and evaluate the impact of caring for suspected victims (...) of child maltreatment on nurses’ moral distress, burnout, and intention to leave. Design and method: This descriptive cross-sectional correlational study was conducted in a mid-Atlantic, urban area Magnet pediatric level I trauma center that cares for over 1800 cases of suspected child maltreatment annually. An anonymous electronic survey was sent to all the nurses working at the hospital.Ethical considerations:Institutional Review Board approval was received from the first author’s university and the hospital where the study was conducted.Findings:Overall, nurses ( N = 146) reported low levels of moral distress with a mean score of 59.54 (SD = 49.22) and a range of 0–300 on the Moral Distress Scale Neonatal–Pediatric version. Although the frequency of caring for suspected child maltreatment victims did not affect nurses’ moral distress, caring for victims with fatal maltreatment contributed to nurses’ intention to leave, χ2(1) = 5.35, p = 0.02.Conclusion:The results of this study add to the understanding of moral distress in pediatric nursing. In a series of formal studies and less formal applications, Hong and Page offer a ‘diversity trumps ability’ result on the basis of a computational experiment accompanied by a mathematical theorem as explanatory background (Hong & Page 2004, 2009; Page 2007, 2011). “[W]e find that a random collection of agents drawn from a large set of limited-ability agents typically outperforms a collection of the very best agents from that same set” (2004, p. 16386). The result has been extremely influential as (...) an epistemic justification for diversity policy initiatives. Here we show that the ‘diversity trumps ability’ result is tied to the particular random landscape used in Hong and Page’s simulation. We argue against interpreting results on that random landscape in terms of ‘ability’ or ‘expertise.’ These concepts are better modeled on smother and more realistic landscapes, but keeping other parameters the same those are landscapes on which it is groups of the best performing that do better. Smoother landscapes seem to vindicate both the concept and the value of expertise. Change in other parameters, however, also vindicates diversity. With an increase in the pool of available heuristics, diverse groups again do better. Group dynamics makes a difference as well; simultaneous ‘tournament’ deliberation in a group in place of the ‘relay’ deliberation in Hong and Page’s original model further emphasizes an advantage for diversity. ‘Tournament’ 2 dynamics particularly shows the advantage of mixed groups that include both experts and non-experts. As a whole, our modeling results suggest that relative to problem characteristics and conceptual resources, the wisdom of crowds and the wisdom of the few each have a place. We regard ours as a step toward attempting to calibrate their relative virtues in different modelled contexts of intellectual exploration. (shrink)

OBJECTIVE: How outside factors affect physician decision making remains an open question of vital importance. We sought to investigate the importance of various influences on physician decision making when clinical guidelines differ from patient preference. -/- METHODS: An online survey asking 469 primary care providers (PCPs) across four practice sites whether they would order magnetic resonance imaging for a patient with uncomplicated back pain. Participants were randomized to one of four scenarios: a patient's preference for imaging (control), a patient's preference (...) plus a colleague's opinion against imaging (colleague), a patient's preference plus a professional society's recommendation against imaging (profession), or a patient's preference plus an accountable care organization's quality metric that measures physician use of imaging (ACO). Demographic information and the reasoning behind participants' decisions also were obtained. -/- RESULTS: A total of 168 PCPs completed the survey, yielding a 36% completion rate. A majority chose not to pursue imaging: control 68%, colleague 85%, profession 87%, and ACO 78%. Multivariate logistic regression revealed that participants were more likely not to order advanced imaging only when reminded of a professional society recommendation (P = 0.017). Regression also suggested that practice site exerted an effect on the primary outcome. Evidence-based medicine and clinical judgment were the most cited reasons for the decision. -/- CONCLUSIONS: Our results reinforce the potential to leverage professional societies to advance evidence-based medicine and reduce unnecessary testing. At the same time, practice site appeared to exert influence, suggesting that these recommendations must be part of local institutional culture to be effective. (shrink)

With the outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), global researchers were confronted with major challenges. The German National Pandemic Cohort Network (NAPKON) was launched in fall 2020 to effectively leverage resources and bundle research activities in the fight against the coronavirus disease 2019 (COVID-19) pandemic. We analyzed the setup phase of NAPKON as an example for multicenter studies in Germany, highlighting challenges and optimization potential in connecting 59 university and nonuniversity study sites. We examined the ethics application (...) process of 121 ethics submissions considering durations, annotations, and outcomes. Study site activation and recruitment processes were investigated and related to the incidence of SARS-CoV-2 infections. For all initial ethics applications, the median time to a positive ethics vote was less than two weeks and 30 of these study sites (65%) joined NAPKON within less than three weeks each. Electronic instead of postal ethics submission (9.5 days (Q1: 5.75, Q3: 17) vs. 14 days (Q1: 11, Q3: 26), p value = 0.01) and adoption of the primary ethics vote significantly accelerated the ethics application process. Each study center enrolled a median of 37 patients during the 14-month observation period, with large differences depending on the health sector. We found a positive correlation between recruitment performance and COVID-19 incidence as well as hospitalization incidence. Our analysis highlighted the challenges and opportunities of the federated system in Germany. Digital ethics application tools, adoption of a primary ethics vote and standardized formal requirements lead to harmonized and thus faster study initiation processes during a pandemic. (shrink)

Pp. xii + 292.The Vivekacūḍāmaṇi or Crown Jewel of Discrimination has for centuries been celebrated as one of the most effective prakaraṇa grantha or independent pedagogical [End Page 616] treatises in the literature of Advaita, the nondualistic school of (...) Vedānta, owing to its straightforward dialogical style and lucid presentation of bedrock theories of the school. It has been ascribed, along with literally hundreds of other works, to the preeminent philosophical commentator in Advaita's illustrious history, Śaṅkara (ca. 650-700 C.E.). Modern lndological scholarship, however, has contested both of these grounds for its fame, calling into question its consistency with the theoretical framework laid down by Śaṅkara as well as its traditional attribution to the hand of the medieval Kerela master.In The Vivekacūḍāmaṇi of Śahkarācārya Bhagavatpāda: An Introduction and Translation, his recent translation of The Crown Jewel—the tenth made into English—John Grimes sets out to defend the integrity of tradition against modern criticism. Grimes, whose incisive scholarship has given us a number of important works on Advaita such as An Advaita Vedanta Perspective on Language and Problems and Perspectives in Religious Discourse: Advaita Vedanta Implications, as well as A Concise Dictionary of Indian Philosophy, applies his considerable Sanskrit and Advaita acumen to The Crown Jewel in order to achieve a twofold goal. In the Preface he vows to contribute a readable translation that is attentive to the religious sensibilities and goals of the Vivekac&umacrḍāmaṇi to guide the reader to ultimate wisdom and liberation but also to offer a rendition that speaks to contemporary audiences in an inclusive way, opening the possibility of knowledge of purified Selfhood to male and female readers alike (pp. vii-ix). At the same time, given the length to which he goes concerning this matter in his extensive Introduction, Grimes proves himself keen to show that, despite modern objections and reservations, "a strong case can be made that the Vivekacūḍāmaṇi is a genuine work of Śaṅkara's and that it differs in certain respects from his other works in that it addresses itself to a different audience and has a different emphasis and purpose" (p. 13). What is offered to the modern reader in this translation, then, is a defense of the traditional authorial and authoritative status of this popular Advaita treatise.Grimes' presentation falls into two parts. The first is a fifty-page Introduction that includes the life and teachings of Śaṅkara, a rundown on the basic doctrines of Advaita Vedānta, and reflections on the various approaches to the study of author and school. Much of this Introduction is occupied with the question of whether the historical Śaṅkara actually wrote the Vivekacūḍāmaṇi, in a discussion that engages the criteria for reliably attributing a work to him set down by Paul Hacker a half century ago. Here, Grimes immerses himself in a debate in which Hacker came down on the side of Śaṅkara having written the work, while Daniel lngalls and Sengaku Mayeda, following Hacker's own criteria, have rejected this conclusion. Grimes point by point traverses Hacker's criteria of authorship, particularly Hacker's analysis of how Śaṅkara defines basic terms such as avidyā, anirvacanīya, prāgabhāva, and other related concepts in works such his commentary on the Brahmasūtra compared to how these are defined in The Crown Jewel. The authors respond to a letter by Mitchell Berger in the March‐April 2024 issue of the Hastings Center Report concerning their essay “Securing the Trustworthiness of the FDA to Build Public Trust in Vaccines.”.

This thesis seeks to provide an overview and examination of the thought of the significant contemporary sociologist, Peter L. Berger. Berger is concerned with the issue of how meaning is constructed in modern, secular, bureaucratic society. Furthermore, this thesis seeks to outline, and trace the development of, Berger's thought. To achieve this the thesis examines Berger's use of the disciplines of the sociology of knowledge and religion, along with contemporary studies in religion and theology. Berger, (...) by linking the function of a theodicy with that of making meaning, allows for theodicies to be conceived of in the broader context of making meaning in contemporary society. As such, a contemporary theodicy needs to include (indeed, it needs to be inclusive, rather than exclusive) such factors as the relationship between self, others, the world, and the transcendent so as to provide some basis for an authentic and meaningful existence. There is a need for a more inclusive theodicy (other than the traditional individualistic type) which has hermeneutic concern for the 'whole' (wholeness of self, wholeness in relationships with others, wholeness with the world/environment, and wholeness with the transcendent). However, this 'wholeness' will not be provided by over-arching, public, structures or systems; it will need to be through chosen, private means which reflect the Post-Modernist situation where 'closure' on a grand scale is unobtainable (Marshall, 1992, pp. 192 -193). Berger's work provides the possibility for this legitimation of a theodicy (or theodicies) which will provide meaning in Post-Enlightenment society. The construction of meaning in contemporary society needs an ability to cope with complexity, it needs to be reasonable, as well as contemporary (to cope with the plurality in modern society), and it is on the way (that is, not given to closure). Therefore any contemporary theodicy, or system of meaning, must be able to be historically concerned (that is, conscious of its origins and open to the future), empirical (that is, open to scrutiny and review), inductive (that is, dealing with concrete reality, not abstract theory), and concerned with people's lived experience. Berger's signals of transcendence allow for the legitimation of this private, deinstitutionalized religion; that is, they legitimate a meaningful theodicy for contemporary humanity. This theodicy, which is able to accommodate the wider view current in modern society provided by the ecological movement, interaction between the various religious traditions, the feminist movement, the reality of multi-culturalism, and the resulting pluralism from the above factors, can provide some basis for a meaningful and authentic existence in contemporary society. The signals of transcendence are able to correlate people's lived experience (their 'natural reality') to a reality which is "in, with and under" that natural reality (Berger, 1992, p. 155).
