Since most of the functions in cells are mediated by multimeric protein complexes, the determination of protein–protein interactions is an important step in the study of cellular mechanisms. Traditionally, after screening for possible target interactors by means of a yeast two‐hybrid screen, several methods are used to validate the initial result before carrying out functional experiments. Nowadays, non‐invasive fluorescence‐based methods like Bioluminescence Resonance Energy Transfer (BRET) and Fluorescence Resonance Energy Transfer (FRET) are widely used in the study of protein–protein interactions (...) in living cells. In the present review, we address the individual strengths and weaknesses of both RET approaches, providing information on their possible future use in the study of G protein‐coupled receptor oligomerization. This is the first book-length study of Descartes's metaphysics to place it in its immediate historical context, the Late Scholastic philosophy of thinkers such as Suárez against which Descartes reacted. Dynamic Theodicy (DT) is a broad concept we bring up to designate some modern Philosophical Theology attempts to reconcile the necessary and perfect existence of God with the contingent characteristics of human life. In this paper we analyze such approaches and discuss how they have become incomprehensible because the metaphysical assumptions implicit in these explanations have lost their intrinsic relation to the natural human desire for salvation. In the first part we show Charles Hartshorne's DT-model, arising from the modal logic (...) of perfection, and the modern rational problems of this position in making infinite-necessary Being (God) and finite-contingent being (human) compatible. We note that at the heart of the contradictions in this DT account is a dialectical mode of thinking that makes it difficult to find a correct solution to this dichotomy, and to assume a human desire that could be considered related to lifelong goals. In the second part, supported by the proposal of Hans Urs von Balthasar's DT, we develop the concepts of bodily vulnerability, corporeal intentionality, and natural desire for salvation, which come from an Aristotelian-Thomistic thought. This theory is established in order to build an argument, following Alasdair MacIntyre’s ethical framework, on how to make possible the recovery of a metaphysical and anthropological desire that transcends natural aging and goes beyond death. We conclude that both human dependence and the virtues that arise naturally when human beings decide to seek the good of their transcendent condition, make it possible to recover the natural desire for salvation through divine and human love. In this chapter, we discuss a selection of current views of the neural correlates of consciousness (NCC). We focus on the different predictions they make, in particular with respect to the role of prefrontal cortex (PFC) during visual experiences, which is an area of critical interest and some source of contention. Our discussion of these views focuses on the level of functional anatomy, rather than at the neuronal circuitry level. We take this approach because we currently understand more about experimental (...) evidence at this coarse level and because these results are appropriate for arbitrating between current theoretical frameworks. We discuss the Two-Visual-Systems Hypothesis (Milner & Goodale 1995; 2006), Local Recurrency (Lamme 2010; Lamme 2006), Higher Order (Lau 2008; Lau & Rosenthal 2011) and Global Workspace theories (Baars 1997; Baars 2005; Dehaene & Naccache 2001; Dehaene 2014). Despite the apparent stark differences between conscious and unconscious perceptual processing, available evidence suggests that their neural substrates must be largely shared. This indicates that the difference in neural activity between conscious and unconscious perceptual processing is likely to be subtle and highly specialized. We argue that current experimental evidence about the involvement of specific activity in prefrontal cortex supports the higher order neural theory of consciousness. In consequence, imaging techniques that focus only on marked differences between conscious and unconscious level of activity are likely to be insensitive to the relevant neural activity patterns that underlie conscious experiences. Finally, it follows from the evidence we discuss that the functional advantages of conscious over unconscious perceptual processing may be more limited than commonly thought. (shrink)

Arguably the most foundational principle in perception research is that our experience of the world goes beyond the retinal image; we perceive the distal environment itself, not the proximal stimulation it causes. Shape may be the paradigm case of such “unconscious inference”: When a coin is rotated in depth, we infer the circular object it truly is, discarding the perspectival ellipse projected on our eyes. But is this really the fate of such perspectival shapes? Or does a tilted coin retain (...) an elliptical appearance even when we know it’s circular? This question has generated heated debate from Locke and Hume to the present; but whereas extant arguments rely primarily on introspection, this problem is also open to empirical test. If tilted coins bear a representational similarity to elliptical objects, then a circular coin should, when rotated, impair search for a distal ellipse. Here, nine experiments demonstrate that this is so, suggesting that perspectival shapes persist in the mind far longer than traditionally assumed. Subjects saw search arrays of three-dimensional “coins,” and simply had to locate a distally elliptical coin. Surprisingly, rotated circular coins slowed search for elliptical targets, even when subjects clearly knew the rotated coins were circular. This pattern arose with static and dynamic cues, couldn’t be explained by strategic responding or unfamiliarity, generalized across shape classes, and occurred even with sustained viewing. Finally, these effects extended beyond artificial displays to real-world objects viewed in naturalistic, full-cue conditions. We conclude that objects have a remarkably persistent dual character: their objective shape “out there,” and their perspectival shape “from here.”. (shrink)

Studying the neural correlates of conscious awareness depends on a reliable comparison between activations associated with awareness and unawareness. One particularly difficult confound to remove is task performance capacity, i.e. the difference in performance between the conditions of interest. While ideally task performance capacity should be matched across different conditions, this is difficult to achieve experimentally. However, differences in performance could theoretically be corrected for mathematically. One such proposal is found in a recent paper by Lamy, Salti and Bar-Haim [Lamy (...) D, Salti M, Bar-Haim Y. Neural correlates of subjective awareness and unconscious processing: an ERP study. J Cognitive Neurosci 2009,21:1435-46], who put forward a corrective method for an electroencephalography experiment. We argue that their analysis is essentially grounded in a version of High Threshold Theory, which has been shown to be inferior in general to Signal Detection Theory. We show through a series of computer simulations that their correction method only partially removes the influence of perfor- mance capacity, which can yield misleading results. We present a mathematical correction method based on Signal Detection Theory that is theoretically capable of removing performance capacity confounds. We discuss the limitations of mathemati- cally correcting for performance capacity confounds in imaging studies and its impact for theories about consciousness. (shrink)

Der Einblick in die Geschichte der Entgegensetzung von Gutem und Schlechtem vermittelt nicht nur die einzelnen Stationen der Auseinandersetzung mit diesem Problem, sondern erörtert es auch unter sachlichen Gesichtspunkten. Wenngleich jede Entgegensetzung einen den entgegenzustellenden Gliedern gemeinsamen Bezugspunkt fordert, lassen sich Gutes und Schlechtes weder einer gemeinsamen Gattung unterordnen, noch als Gattungen in irgendeinem Sinne begreifen. Trotzdem sind Gutes und Schlechtes einander entgegengesetzt. Dies zeigt, daß die Entgegensetzung nicht nur ,,eine Sache" des Denkens ist, sondern auch von der Seinsstruktur beider (...) Glieder abhängt. Diese Entgegensetzung ist zudem als eine Wesenszusammengehörigkeit zu deuten, die mit der Entgegensetzung von Sein und Nichts wesenhaft verknüpft ist. (shrink)

El artículo analiza las condiciones de validez del testimonio en ciencias sociales. Sus potencialidades centrales son el constituir un campo de exploración e investigación de acercamiento a la subjetividad, especialmente a la de los sectores populares. Ello ha permitido incorporar sus discursos y representaciones en las ciencias sociales. Sin embargo, dicha incorporación se ha realizado bajo una perspectiva empirista, según la cual el testimonio es una representación inmediata, directa de los sujetos, lo que oculta la mediación del testimonio a través (...) del cientista social, particularmente a través de la entrevista. Por ende, la idea del testimonio como una ¿voz de los sin voz¿ oculta el diálogo entre investigador-sujetos, que podría ser potenciado en un tipo de investigación hermenéutica en la que el investigador ya no ostentaría el monopolio de la interpretación. IF THE HISTORY OF PHILOSOPHY studies ideas from the past, as is generally accepted, then historians of philosophy face a serious problem concerning their object of study for two reasons. In the first place, like all history, the history of philosophy is concerned with the past and we can never have direct empirical access to the past unless that past is close to us and we have taken part in it. In order to know the past in which we have (...) not participated we must rely on the testimony of those who had direct access to it and left records of what they witnessed. In the second place, the problem arises because the specific object that the history of philosophy studies is ideas and ideas are not things, events, or facts for which we can have direct empirical evidence even if we are contemporaneous with them. The most we can have is indirect empirical evidence. We do not perceive ideas; what we perceive are certain phenomena that suggest to us certain ideas. If I ask you, for example, "Do you approve of what the President did?" and you frown in return, I conclude that you do not. But it is altogether possible that you do in fact approve of the President's action, although you wish me to think that you do not and thus mislead me by making the frown. My conclusion that you do not, then, can be taken only as an interpretation of what you are thinking based on certain empirical evidence that is only indirectly related to what you think. Thus the study of the history of philosophy is very difficult, more difficult than the study of the type of history that relies on events for which there can be direct empirical evidence; for not only is direct access to the past impossible for historians of philosophy from the present, but even if they had it they would not have direct access to the ideas which are supposed to be the object of their study. HISPANIC PHILOSOPHY. The notion of Hispanic philosophy is a useful one for trying to understand certain historical phenomena related to the philosophy developed in the Iberian peninsula, the Iberian colonies in the New World, and the countries that those colonies eventually came to form. It is useful for two reasons. First, it focuses attention on the close relations among the philosophers in these geographical areas; and second, other historical denominations and categorizations do not do justice to such relations. This becomes (...) clear when one examines the standard general categorizations according to which the philosophical thought of the mentioned geographical areas is divided and studied: Spanish philosophy, Portuguese philosophy, Catalan philosophy, Latin American philosophy, Spanish-American philosophy, and Ibero-American philosophy. Esta última, en lo que concierne a los Cuadernos de Heidegger, era dura y recubierta por un hule negro. The aim of this paper is to present a revision of international versions of the Utrecht Work Engagement Scale and to describe the psychometric properties of a Portuguese version of the UWES-9 developed simultaneously for Brazil and Portugal, the validity evidence related with the internal structure, namely, Dimensionality, measurement invariance between Brazil and Portugal, and Reliability of the scores. This is the first UWES version developed simultaneously for both countries, and it is an important instrument for understanding employees' work engagement (...) in the organizations, allowing human resources departments to better use workforces, especially when they are migrants. A total of 524 Brazilian workers and 522 Portuguese workers participated in the study. Confirmatory Factor Analysis, group comparisons, and Reliability estimates were used. The use of workers who were primarily professionals or administrative support, according to ISCO-08, reinforced the need to collect data on other professional occupations. Confirmatory factor analysis showed acceptable fit for the UWES-9 original three-factor solution, and a second-order factor structure has been proposed that presented an acceptable fit. Full-scale invariance was obtained between the Portuguese and Brazilian samples, both for the original three-factor first-order and second-order models. Data revealed that Portuguese and Brazilian workers didn't show statistically significant differences in the work engagement dimensions. This version allows for direct comparisons of means and, consequently, for performance of comparative and cross-cultural studies between these two countries. (shrink)

La opera flotante de John Barth tiene varias puertas de acceso para el análisis. Una de ellas nos lleva a indagar sobre el tema que circunda la obra: el suicidio. Para ello, en el presente trabajo vamos a contrastar las posibles características apocalípticas del personaje principal, Todd Andrews, con determinados postulados de la filosofía existencialista de Jean Paul Sartre. La propuesta estará centrada en revisar puntos de divergencia y de afinidad en ambas cosmovisiones.

Rescue workers have a stressful and risky occupation where being engaged is crucial to face physical and emotional risks in order to help other persons. This study aims to estimate work engagement levels of rescue workers (namely comparing nurses, firefighters, and police officers) and to assess the validity evidence related to the internal structure of the Portuguese versions of the UWES-17 and UWES-9, namely, dimensionality, measurement invariance between occupational groups, and reliability of the scores. To evaluate the dimensionality, we compared (...) the fit of the three-factor model with the fit of a second-order model. A Portuguese version of the instrument was applied to a convenience sample of 3,887 rescue workers (50% nurses, 39% firefighters, and 11% police officers). Work engagement levels were moderate to high, with firefighters being the highest and nurses being the lowest engaged. Psychometric properties were evaluated in the three-factor original structure revealing acceptable fit to the data in the UWES-17, although the UWES-9 had better psychometric properties. Given the observed statistically significant correlations between the three original factors, we proposed a 2nd hierarchal structure that we named work engagement. The UWES-9 first-order model obtained full uniqueness measurement invariance, and the second-order model obtained partial (metric) second-order invariance. (shrink)

We present some results concerning elimination of quantifiers and elementary equivalence for Boolean products of real closed valuation rings and fields. We also study rings of continuous functions and rings of definable functions over real closed valuation rings under this point of view.
