Genome maintenance requires various nucleoid-associated factors in prokaryotes. Among them, the SMC protein has been thought to play a static role in the organization and segregation of the chromosome during cell division. However, recent studies have shown that the bacterial SMC is required to align left and right arms of the emerging chromosome and that the protein dynamically travels from origin to Ter region. A rod form of the SMC complex mediates DNA bridging and has been recognized as a machinery (...) responsible for DNA loop extrusion, like eukaryotic condensin or cohesin complexes, which act as chromosome organizers. Attention is now turning to how the prototype of the complex is loaded on the entry site and translocated on chromosomal DNA, explaining its overall conformational changes at atomic levels. Here, we review and highlight recent findings concerning the prokaryotic SMC complex and discuss possible mechanisms from the viewpoint of protein architecture. Recent studies show dynamic movements of the prokaryotic SMC protein complex: initial loading, translocation on DNA for bacterial chromosome organization. Despite their diversity and ecological importance, many areas of the SAR—Stramenopila, Alveolata, and Rhizaria—clade are poorly understood as the majority of SAR species lack molecular data and only 5% of species are from well-sampled families. Here, we review and summarize the state of knowledge about the three major clades of SAR, describing the diversity within each clade and identifying synapomorphies when possible. We also assess the “dark area” of SAR: the morphologically described species that are missing molecular data. The majority (...) of molecular data for SAR lineages are characterized from marine samples and vertebrate hosts, highlighting the need for additional research effort in areas such as freshwater and terrestrial habitats and “non-vertebrate” hosts. We also describe the paucity of data on the biogeography of SAR species, and point to opportunities to illuminate diversity in this major eukaryotic clade. Preterm discharge effects: Relationship between the maternal experience and newborn's psychobiological regulation The purpose of this study is to investigate how the past experiences of mothers and their potentially traumatic events during pregnancy may have influenced the processes of psychobiological self-regulation and cognitive development in a child born preterm. Eighty children who had a gestational age of < 32 weeks were examined at the 9th month of the corrected age. The mothers and children were divided in two groups: multipara mothers (...) and their children and the primipara mothers and their children. These children were examined on the basis of sleep disorders, nutritional problems and mental development. Perinatal data from clinical observations and data from standardized developmental tests were used to describe the differences. The children of primipara mothers who had unexpected preterm delivery presented major incidences of sleep disorders while the children of primipara mothers, who programmed preterm delivery, presented a major incidences of nutrition problems. The predictability of preterm delivery has a major influence on the capacity of newborns for self-regulation in the case of primipara mothers. (shrink)

Enzyme engineering allows to explore sequence diversity in search for new properties. The scientific literature is populated with methods to create enzyme libraries for engineering purposes, however, choosing a suitable method for the creation of mutant libraries can be daunting, in particular for the novices. Here, we address both novices and experts: how can one enter the arena of enzyme library design and what guidelines can advanced users apply to select strategies best suited to their purpose? Section I is dedicated (...) to the novices and presents an overview of established and standard methods for library creation, as well as available commercial solutions. The expert will discover an up‐to‐date tool to freshen up their repertoire (Section I) and learn of the newest methods that are likely to become a mainstay (Section II). We focus primarily on in vitro methods, presenting the advantages of each method. Our ultimate aim is to offer a selection of methods/strategies that we believe to be most useful to the enzyme engineer, whether a first‐timer or a seasoned user. (shrink)

Se propone revisar la elaboración del concepto de piñen examinando la relación entre lenguaje y colonialidad, para, de ese modo, subrayar el potencial político y reivindicativo que comporta en la novela del mismo nombre, Piñen (2019). Dicha hipótesis articulará el ejercicio de lectura aquí propuesto, atendiendo principalmente las estrategias narrativas empleadas –servirse de un polilingüismo que rompe estructuras gramaticales– para descentrar el lenguaje de su determinismo colonial. Así, interpretamos dicha (...) estrategia escritural en consonancia con las potencias micropolíticas y descolonizadoras del arte descritas por Suely Rolnik y, en consecuencia, apostando por la posibilidad de diagramar un devenir minoritario que dispute los imaginarios del inconsciente colonial-capital. Over the last decade, research in semantics and pragmatics has started to increasingly incorporate new experimental methods from cognitive psychology. That this empirical stance on utterance interpretation has now reached maturity is revealed by two unmistakable symptoms: an increased reflection on the contextual methods used to elicit experimental data, and a continuous expansion of the linguistic phenomena and themes being investigated through these methods. The articles gathered in this volume testify to this very recent evolution of the field: a first (...) set of papers questions the interpretation or interpretability of data collected in non-naturalistic settings, while a second set reflects the extension of experimental methods to new topics. This article has as its theme the relationship between "power", "reason" and "religion" in O alienista, one of the central texts of Machado de Assis, written in 1881 and initially published in eleven chapters, between October 1881 and March 1882. It is intended to identify and analyze the power relations between the State ("Throne") and the Catholic Church ("Altar"); as well as to highlight the relations between "reason" (science) and its "absence" (madness); without neglecting the Machadian denunciations and criticisms of (...) scientism prevailing in the nineteenth century. To do so, at first, it is presented, even if summarily, the Author and the Tale; in a second moment, shedding lights on its "space", "time" and "action" its plot, necessary assumption for the last moment, where the "Power", "Reason" and "Religion" will be analyzed. This research is justified by the relevance of the author, the place that the alienist occupies in the set of his works, as well as the social relevance of his ironic criticisms. In this analysis, a bibliographic method of critical-analytical reading of the Tale was used, using also the interpretations of some scholars, listed in the References. (shrink)

For families all over the world, going through a pandemic has presented a number of challenges. In particular, social distancing measures involving the closure of schools and day care centers, as well as increasing work hours at home, made parents face very demanding situations. However, we know little about whether parents’ burnout levels are influenced by the age of their children. This study sought to determine whether levels of parental burnout are higher in families with at least one child under (...) the age of four than in families with older children. The second goal was to explore whether having children under 4 years of age moderates the relationship between parental cooperation and PB. A cross-sectional study was conducted with a sample of 651 participants since May 18th until August 27th, 2020. The main results showed that child age is a predictor of PB. Besides, having a child aged 0–4 years old moderates the relationship between parental cooperation and PB. Finally, it was found that in cases where there was at least one child under 4 years of age in the family, with one of the partners who worked remotely, the respondent’s PB rose by 7.9 points. The implications of these results with respect to the consideration of children’s ages in the different parental scenarios were discussed. (shrink)

Soccer players inescapably live under stress during the sportive career, and many real-life aspects of soccer situations operate in the ongoing performance. This study’s main objective was to elaborate the List of Stressors in Professional Indoor and Field Soccer, a self-report instrument designed to measure the impact of 77 soccer situations upon the sport performance. Participants were 138 indoor and field soccer players from the Brazilian Premier League. Each situation was evaluated on a 7-point scale, ranging from the most negative (...) to the most positive. Data were analyzed according to the players’ perception of the items: distress or eustress and its intensity, and after that, situations perceived as plus −1 and +1 were compared by time in which they were experienced and distributed among five categories established by the literature: Expectations about the Performance, Personal Factors, Competition Aspects, Training Demands, and Relationship with Significant People. Narratives of athletes’ experiences were also used to discuss the results. An Exploratory Structural Equation Modeling using Bi-factorial was employed to assess the factor structure. For the total participants, 49 situations were perceived as distress and 28 as eustress. Using the criteria established a priori, the distribution was among the five categories in the remaining 32 situations. Differences in perception between less and more experienced players were found in 11 situations. The results revealed that Brazilian professional soccer players experience various stressful situations. These events are important representations of environmental demands and could predict the performance as they are perceived as eustress or distress. Some of these stressful situations are inherent in sport and others adjacent to the sports system or environment. Coach pressure to win and conflicts with teammates are examples of stressors in-sport, family problems and disputes with press or fans are examples of stressors external to the team, also called peripheral opponents, and showed the relative social influence of significant others in soccer performance. We can conclude that the knowledge of the direction of a given stress situation has important practical implications in preparing athletes and helping them face the performance stressors that are part of soccer daily life. What place should be given to ruins in the phenomenology of memory sketched by Ricœur in Memory, History, Forgetting? The contemplation of ruins raises the same problem as that which occupies the whole of Ricœur’s work: the representation of the past and the relations of memory and imagination in the formation of recollection. Can ruins give rise to a rearrangement of the relations of memory and imagination and produce an “imagining remembering”?

Henri Bergson, Vladimir Jankélévitch, Emmanuel Levinas ont en commun d'avoir marqué la philosophie européenne du XXe siècle. En plus de leurs oeuvres respectives, ils ont tous les trois entretenu des liens manifestes à la fois dans leurs textes et dans leurs biographies. De ce trio, on ne peut pas dire que Bergson, philosophe pourtant déjà installé dans le paysage intellectuel français, prenne la place privilégiée du maître. Ni Jankélévitch, ni Levinas n'ont été, à proprement parler, ses élèves, cependant une relation (...) étroite s'est instaurée entre eux, marquée non pas par une posture de déférence, mais par des préoccupations conceptuelles partagées ainsi qu'un travail engagé sur des questions communes. Les pensées de Jankélévitch et Levinas sont liées par un même franc retrait vis-à-vis de la majorité des courants philosophiques dominants à leur époque, notamment la vogue existentialiste. N'adhérant pas à cette tendance, Jankélévitch et Levinas se tournent vers Bergson pour saisir l'inactualité des questions posées par sa pensée ; plutôt que la restitution de ses thèses, leurs interactions reposent sur un véritable dialogue dans lequel chacun apporte une variation nouvelle. Ces trois philosophes seront finalement réunis par le partage de questions fondamentales, qui résistent à la compréhension et qui s'ouvrent au souci de l'expérience proprement humaine : métaphysique, morale, temps. Bergson, Jankélévitch et Levinas, trois philosophes français marqués par une même inquiétude face à la dégradation de l'humain ; trois moments qui forment une ligne de pensée discrète mais irradiant la philosophie contemporaine, révélée après-coup par les suites qu'elle a autorisées. Leading philosophical models of curiosity represent it as a desiderative attitude whose content is a question, and which is satisfied by knowledge of the answer to that question. I argue that these models do not capture the distinctive character of a form of curiosity that I call 'erotic curiosity'. Erotic curiosity addresses itself not to a question but to an object whose significance for the inquirer is affective as well as epistemic. This form of curiosity is best understood by analogy (...) to erotic love as theorized by Plato in the Symposium. (shrink)

It is generally believed that one argument advanced by Aristotle in favor of the political authority of the multitude is that large groups can make better decisions by pooling their knowledge than individuals or small groups can make alone. This is supported by two analogies, one apparently involving a “potluck dinner” and the other aesthetic judgment. This article suggests that that interpretation of Aristotle’s argument is implausible given the historical context and several features of the text. It argues that Aristotle’s (...) support for the rule of the multitude rested not on its superior knowledge but rather on his belief that the virtue of individuals can be aggregated and even amplified when they act collectively. This significantly alters our understanding of Aristotle’s political thought and presents a powerful alternative to the epistemic defenses of mass political activity popular today. (shrink)

Internationally, there is considerable inconsistency in the recognition and regulation of children's genetic connections outside the family. In the context of gamete and embryo donation, challenges for regulation seem endless. In this paper, I review some of the paths that have been taken to manage children' being closely genetically related to people outside their families. I do so against the background of recognising the importance of children's interests as moral status holders. I look at recent qualitative research involving donor-conceived people (...) and borrow their own words to make sense of a purported interest to know (of) their close genetic ties. I also review ways in which gamete donation may have facilitated new kinds of kinship, which are at the same time genetic and chosen. In short, in this paper, I explore what meaning there could be in genetic connections that is not about parenthood. Further, I argue that the focus on parenthood in previous work in this area may be detrimental to appreciating some of the goods that can be derived from close genetic connections. (shrink)

Self-referential sentences have troubled our understanding of language for centuries. The most famous self-referential sentence is probably the Liar, a sentence that says of itself that it is false. The Liar Paradox has encouraged many philosophers to establish theories of truth that manage to give a proper account of the truth predicate in a formal language. Kripke’s Fixed Point Theory from 1975 is one famous example of such a formal theory of truth that aims at giving a plausible notion of (...) truth by allowing truth value gaps. However, not only the concept of truth gives rise to paradoxes. A syntactical treatment of epistemic notions like belief and knowledge leads to contradictions that very much resemble the Liar Paradox. Therefore, it seems to be fruitful to apply the established theories of truth to epistemic concepts. In this paper, I will present one such attempt of solving the epistemic paradoxes: I adapt Kripke’s Fixed Point Theory and interpret truth, knowledge and belief within the framework of a partial logic. Thereby I find not only the fixed point of truth but also the fixed points of knowledge and belief. In this fixed point, the predicates of truth, belief and knowledge find their definite interpretation and the paradoxes are avoided. (shrink)

The sources, extent and margins of parental obligations in taking decisions regarding their children’s medical care are subjects of ongoing debates. Balancing children’s immediate welfare with keeping their future open is a delicate task. In this paper, we briefly present two examples of situations in which parents may be confronted with the choice of whether to authorise or demand non-therapeutic interventions on their children for the purpose of fertility preservation. The first example is that of children facing cancer treatment, and (...) the second of children with Klinefelter syndrome. We argue that, whereas decisions of whether to preserve fertility may be prima facie within the limits of parental discretion, the right to an open future does not straightforwardly put parents under an obligation to take actions that would detect or relieve future infertility in their children—and indeed in some cases taking such actions is problematic. This article analyzes the autofigurative strategies of La travesía by Luisa Valenzuela in terms of the narratives of the self, the Argentine dictatorship and feminism. A correspondence is found between the disavowal of the voice, the structure of the autobiographical account and the portrait of the experience of the violence of the military regime and of gender. Initially, the novel is situated at the height of the narratives of the self that, with literary resources that are separated from the most (...) conservative autobiographical writings, denounce the impossibility of understanding the subject as univocal, while founding different approaches to the retelling of the experience. This location allows exploring the formal experiments that Valenzuela carries out around the autobiographical exercise and that highlight the crisis of the construction of memory and the exposure of contemporary subjectivity. In addition, it is shown how such strategies serve as a vehicle for reflection on feminism and dictatorship that permeate all of her work. For this, Valenzuela's notion of feminine writing is used, which puts the multiplicity of perspectives before the univocity of the autobiographical voice, the ambiguous before the precise, and the collective before the authoritarian. La travesía is, then, read as part of the works that come to the narration of the experience itself in a framework of the retelling of collective memory, with narrative games that point out the deictic futility of the self and the difficulty of recounting past time, without, therefore, abandoning the call for its reconstruction as a guarantee of non-repetition. It is widely believed that we ought not to criticize others for wrongs that we ourselves have committed. The author draws out and challenges some of the background assumptions about the practice of criticism that underlie our attraction to this claim, such as the tendency to think of criticism either as a social sanction or as a didactic intervention. The author goes on to offer a taxonomy of cases in which the moral legitimacy of criticism is challenged on the grounds (...) that the critic him- or herself engages in the behavior that he or she criticizes in others. The author argues that, in each type of case, the would-be critics should not constrain their participation in moral discourse on the grounds that they are not themselves innocent of the wrongdoing they criticize in others.
