Within the highly organized nuclear structure, specific nuclear domains (ND10) are defined by accumulations of proteins that can be interferon-upregulated, implicating ND10 as sites of a nuclear defense mechanism.Compatible with such a mechanism is the deposition of herpesvirus, adenovirus, and papovavirus genomes at the periphery of ND10. However, these DNA viruses begin their transcription at ND10 and consequently initiate replication at these sites, suggesting that viruses have evolved ways to circumvent this potential cellular defense and exploit it. Other ND10-associated proteins (...) belong to ubiquitin-related pathways. These findings, together with the accumulation of various overexpressed cellular and viral proteins, suggest that ND10 function as nuclear dumps or as nuclear depots. Consistent with the recruitment or deposition of various proteins and viral genomes adjacent to ND10, ND10 themselves may only be protein accumulations at specific but as yet undefined nuclear deposition sites. The concept of specific nuclear deposition sites may explain the juxtaposition of various nuclear bodies and allows testable predictions about a potential supramolecular regulatory mechanism whereby proteins are selectively segregated or released by global changes induced in nuclear functions such as viral infections, stress, or hormonal induction. A certain generalization of the mathematical formalism of quantum mechanics beyond operator algebras is considered. The approach is based on the concept of conditional probability and the interpretation of the Lüders-von Neumann quantum measurement as a probability conditionalization rule. A major result shows that the operator algebras must be replaced by order-unit spaces with some specific properties in the generalized approach, and it is analyzed under which conditions these order-unit spaces become Jordan algebras. Max Weber claims that human behavior can be explained equally well as „natural“ phenomena. This claim has not gained widespread empirical realization in social research so that the question is still of eminent actuality in what way research can profit from Weber’s thought. The article therefore discusses the development of Weber’s theory of causality. It introduces his juridical view of causality which Weber develops in his methodological writings up to his theory of meaningful and causal adequacy. In this development, Weber (...) stresses more and more the need to understand human behavior from a participant’s point of view. The article re-describes this change in the language of causality in order to show that explaining and understanding are not rival forms of knowledge. In order to prove that this train of thought has fundamental consequences for the interpretation of data in social research, the article discusses research results on meritocracy and society. Weber’s theory of meaningful and causal adequacy could be applied in research if it is conceptualized as combining two sorts of causal attributions. To Weber, meaningful causal attributions are at the basis of practical behavior from a participant’s point of view which must be understood whereas causal attributions devoid of meaning (sinnfremd) characterize an observer’s and scientific point of view which reveals additional causal influences on human behavior. Evidence-based recommendations for lifestyles to promote healthy cognitive aging root in reductionistic studies of mostly physical measurable factors with large effect sizes. In contrast, most people consider factors like autonomy, purpose, social participation and engagement, etc. as central to a high quality of life in old age. Evidence for a direct causal impact of these factors on healthy cognitive aging is still limited, albeit not absent. Ultimately, however, individual lifestyle is a complex composite of variables relating to both body and (...) mind as well as to receiving input and generating output. The physical interventions are tied to the more subjective and mind-related aspects of lifestyle and wellbeing in the idea of the “embodied mind,” which states that the mind is shaped by and requires the body. The causality is reciprocal and the process is dynamic, critically requiring movement: the “embodied mind” is a “embodied mind in motion.” Hiking, playing musical instruments, dancing and yoga are examples of body–mind activities that assign depth, purpose, meaning, social embedding, etc. to long-term beneficial physical “activities” and increase quality of life not only as delayed gratification. The present motivational power of embodied activities allows benefiting from the side-effects of late-life resilience. The concept offers an access point for unraveling the mechanistic complexity of lifestyle-based prevention, including their neurobiological foundations. (shrink)

In the six decades since the publication of Julian Huxley's Evolution: The Modern Synthesis, spectacular empirical advances in the biological sciences have been accompanied by equally significant developments within the core theoretical framework of the discipline. As a result, evolutionary theory today includes concepts and even entire new fields that were not part of the foundational structure of the Modern Synthesis. In this volume, sixteen leading evolutionary biologists and philosophers of science survey the conceptual changes that have emerged since Huxley's (...) landmark publication, not only in such traditional domains of evolutionary biology as quantitative genetics and paleontology but also in such new fields of research as genomics and EvoDevo. Most of the contributors to Evolution—The Extended Synthesis accept many of the tenets of the classical framework but want to relax some of its assumptions and introduce significant conceptual augmentations of the basic Modern Synthesis structure—just as the architects of the Modern Synthesis themselves expanded and modulated previous versions of Darwinism. This continuing revision of a theoretical edifice the foundations of which were laid in the middle of the nineteenth century—the reexamination of old ideas, proposals of new ones, and the synthesis of the most suitable—shows us how science works, and how scientists have painstakingly built a solid set of explanations for what Darwin called the "grandeur" of life. The Leuphana Semester at Leuphana University Lüneburg, together with the module “Science bears responsibility” demonstrate how innovative methods of teaching and learning can be combined with the topic of sustainable development and how new forms of university teaching can be introduced. With regard to module content, it has become apparent that, due to the complexity of the field of sustainability, a single discipline alone is unable to provide analyses and solutions. If teaching in higher education is to adequately deal with (...) this complexity, then it is necessary to develop inter- and transdisciplinary approaches that go beyond a purely specialist orientation. Ludwig Wittgenstein refers to Heinrich Hertz as one of his major influences. However, up to now it was not clear from where he acquired his knowledge of mechanics and in which way it contributed to his philosophy. The aim of this book is to answer these questions. It shows that Wittgenstein came in contact with Hertz’s physical world view during his studies in Berlin. It had an effect on his view of the natural sciences in the Tractatus, and Hertz's conception (...) of mass-particles led to its central notion of a simple object. It was especially Wittgenstein’s teacher in Berlin, Joseph Petzoldt, who played an important intermediary role. The manuscript of his 1907 course of lectures "Grundbegriffe der Mechanik" is published here for the first time. It is complemented by reprints of related texts on the topic by Neumann, Lange and Petzoldt. (shrink)

In the debate regarding the different possibilities for gene therapy, it is presupposed that the manipulations are limited to the nuclear genome (nDNA). Given recent advances in genetics, mitochondrial genome (mtDNA) and diseases must be considered as well. In this paper, we propose a three dimensional framework for the ethical debate of gene therapy where we add the genomic type (nDNA vs. mtDNA) as a third dimension to be considered beside the paradigmatic dimensions of target cell (somatic vs. germ-line) and (...) purpose (therapeutic vs. enhancement). Somatic gene therapy can be viewed today as generally accepted, and we review t he contemporary arguments surrounding it on the basis of bioethical-pragmatic, socio-political and deontological classifications. Many of the supposed ethical questions of somatic gene therapy today are not new; they are well-known issues of research ethics. We also critically summarize the different international perspectives and the German ethical discussion regarding manipulations of germ-line cells. (shrink)

The well-known proposal to consider the Lüders-von Neumann measurement as a non-classical extension of probability conditionalization is further developed. The major results include some new concepts like the different grades of compatibility, the objective conditional probabilities which are independent of the underlying state and stem from a certain purely algebraic relation between the events, and an axiomatic approach to quantum mechanics. The main axioms are certain postulates concerning the conditional probabilities and own intrinsic probabilistic interpretations from the very beginning. A (...) Jordan product is derived for the observables, and the consideration of composite systems leads to operator algebras on the Hilbert space over the complex numbers, which is the standard model of quantum mechanics. The paper gives an expository overview of the results presented in a series of recent papers by the author. For the first time, the complete approach is presented as a whole in a single paper. Moreover, since the mathematical proofs are already available in the original papers, the present paper can dispense with the mathematical details and maximum generality, thus addressing a wider audience of physicists, philosophers or quantum computer scientists. Consequently, this book will be ideal for a wide-range of upper level undergraduate and postgraduate courses. The author interprets consciousness as a property that can be possessed by many creatures lacking a language faculty and comprises all of the following: awareness of the surrounding world; awareness of the self as an entity; and awareness of such things as thoughts and feelings. He argues that a biological approach can achieve both the necessary conceptual clarifications and a joint explanation of these divisions of awareness in terms of just two accurately defined concepts of 'internal representation' and two empirically supported assumptions about the functional architecture of a specific set of brain processes. Despite this striking simplicity, his model covers these divisions of awareness both as objective faculties of the brain and as subjective experience. A demarcation between kant's general metaphysics (transcendental principles) and his special metaphysics is attempted, through a discussion of kant's three accounts of lawlikeness, 'transcendental', 'empirical' and 'metaphysical'. the distinctions are defended via a number of 'indicators' in kant's writings, and the 'looseness of fit' between the different types of lawlikeness is discussed.

The aim of the paper is to analyse whether and to what extent the network concept has become the Leitbild of an emerging new economy. The analysis is based on a company survey conducted in eight European territories. There is empirical evidence that only a minority of companies have applied the network concept as a dominant restructuring model, while various types of Fordism are still influencing companies' view on efficient techno-organisational forms. The regional analysis demonstrates that there is not only (...) one path into an informational economy. Some territories still stick to low-tech Fordism, while others apply a technology-based flexi-Fordism, in which ICT functions as a key mechanism to make Fordist structures more flexible. Only in a minority of territories is an informational network economy emerging in which ICT has the function of enabling and supporting communication and co-operation within decentralised organisational forms.
