In vertebrate and invertebrate nonmuscle myosins, light‐ and heavy‐chain phosphorylation regulate myosin assembly into filaments, and interaction with actin. Vertebrate non‐muscle myosins can exist in vitro in three main states, either ‘folded’ (assembly‐blocked) or ‘extended’ (assembly‐competent) monomers, and filaments. Light‐chain phosphorylation regulates the ‘dynamic equilibrium’ between these states. The ability of the myosin to undergo changes in conformation and state of assembly may be an important mechanism in regulating the organization of the cytoskeleton and cell motility.

Most donated organs in the United States come from brain dead donors, while a small percentage come from patients who die in “controlled,” or expected, circumstances, typically after the family or surrogate makes a decision to withdraw life support. The number of organs available for transplant could be substantially if donations were permitted in “uncontrolled” circumstances–that is, from people who die unexpectedly, often outside the hospital. According to projections from the Institute of Medicine, establishing programs permitting “uncontrolled donation after circulatory (...) determination of death,” or uDCDD, throughout the United States has the potential to provide 22,000 more donation opportunities annually. In contrast, U.S. controlled donation after circulatory determination of death, or cDCDD, cases have increased progressively over the past decade from 87 to 848 donors, but currently account for only 10.6 percent of all deceased donors. Following the IOM recommendations, several projects exploring the feasibility of uDCDD were funded by the federal government, including a grant from the Health Resources and Services Administration that supported a pilot project in New York City in which the authors of this article participated.A key feature of our protocol, and indeed of many uDCDD protocols, is the initiation of preservation methods such as chest compressions and extracorporeal membrane oxygenation shortly after death in order to perfuse and preserve the donor's organs. Critics of uDCDD argue that the means of determining death deviates from generally ascribed principles. They assert that reinstituting circulation in order to preserve organs has the effect of “undoing” the prior determination of death. The result is that cDCDD is widely accepted and practiced routinely even though it only marginally increases the number of organs available for transplantation, and uDCDD is widely considered unacceptable despite being ethically embraced and proven to significantly increase organ donation opportunities in other countries. This article explores the evolution of this counterintuitive state of affairs and calls for a policy that, in line with the IOM report, allows for both cDCDD and uDCDD protocols. (shrink)

Jonathan Haidt ( 2001 ) advances the 'Social Intuitionist' account of moral judgment , which he presents as an alternative to rationalist accounts of moral judgment , hitherto dominant in psychology. Here I consider Haidt's anti-rationalism and the debate that it has provoked in moral psychology , as well as some anti-rationalist philosophical claims that Haidt and others have grounded in the empirical work of Haidt and his collaborators. I will argue that although the case for anti-rationalism in moral psychology (...) based on the work of Haidt and his collaborators is plausible , a decisive case has yet to be made. It will require further experimental evidence before a decisive case could be made. My assessment of anti-rationalist philosophical arguments that are grounded in the empirical work of Haidt and his collaborators is much more negative than this. I will argue that this body of empirical work is a very unpromising basis for such arguments. (shrink)

This article sets out to describe how churches have responded and continue to respond to fast-changing urban environments in Pretoria Central and Mamelodi East, animating Henri Lefebvre’s sociological perspective of citadins or urban inhabitants. We make tentative interpretations and offer critical appreciation. Churches, which were historically separated from the city centre, now directly participate in claiming a right to the city. With necessary fluidity, churches express lived African urbanisms through informality, place-making, spatial innovation and everyday rituals. Though not (...) exhaustive, the article focuses on rituals shared among historic urban Christians and contemporary African urban Christians, namely prayer, listening to the Bible and worship. These rituals, when combined with the churches’ spatial innovation, to a certain degree contribute to place-making. (shrink)

By challenging the state's prerogative to distinguish between insiders and outsiders, citizens and non-citizens, political movements by and in support of migrants and refugees are forcing questions about what criteria, if any, can and should be used to determine who can claim membership in the political community. To illustrate the complexity of this politics this article analyzes the major demand that underscores every campaign undertaken by non-status refugees and migrants in Canada: a program that would allow them to "regularize" their (...) status. Notably, these campaigns are being directed at both the state and city levels of governance. Together, these are two sites in which claims and counter-claims about community, belonging, and citizenship are being made by, for, and against non-status immigrants. In each case, migrant political agency is asserted in places meant to deny, limit, or repress it. The article argues that the significance of these sites is that they allow for non-status refugees and migrants themselves to act as mediators or translators between the city and nation, between polis and cosmopolis. This paper rereads David Hume's Dialogues Concerning Natural Religion as dramatising a distinctive, naturalistic account of toleration. I have two purposes in mind: first, to complete and ground Hume's fragmentary explicit discussion of toleration; second, to unearth a potentially attractive alternative to more recent, Rawlsian approaches to toleration. To make my case, I connect Dialogues and the problem of toleration to the wider themes of naturalism, scepticism and their relation in Hume's thought, before developing a new interpretation of Dialogues part (...) 12 as political drama. Finally, I develop the Humean theory of toleration I have discovered by comparison between Rawls's and Hume's strategies for justification of a tolerant political regime. (shrink)

This essay seeks to question a certain imbalance in many existing accounts of Plato's dialogues. This imbalance involves a tendency to place too much emphasis upon a dualism between matter and spirit, soul and body. Although the author by no means denies the presence of such dualistic elements, she wishes to qualify them with reference to those aspects of Plato's dialogues which appear to place a stress upon the importance of multiplicity, myth, ritual, society, history, mimesis and time. Such instances (...) of mediation, it will be argued, are just as central to an understanding of Plato's philosophy as instances where the body and/or instances of multiplicity appear to be deprioritised in favour of the soul and the unity of the intelligible realm. These issues will be explored with particular reference to Plato's examination of the nature of justice in the city and its relationship with the philosopher‐guardians exercise of phronesis. What is the relationship between dikaiosyne and phronesis? In order to answer this question, several further questions will be raised: What, for Plato, is a city? What is philosophy? And why, in the Laws, does the Athenian describe the city as “the true tragedy”? (shrink)

Critics allege that there are structural inconsistencies between the city and the soul, and that even if they were somehow structurally analogous, they are nevertheless dif- ferent. Why, then, would one expect that justice in one would be enlightening for the discovery of justice in the other? This paper examines (...) the passages in which the anal- ogy is developed and finds that Socrates’ claims regarding similarities between the city and the soul are not as far-fetched as critics assert. Criticisms of the analogy are shown to be misunderstandings arising from an unwillingness to accept the political implications of Socrates’ plan. Ultimately, the analogy rests on important structural similarities between the city and the soul, and is a powerful tool towards an under- standing of the individual, of the city and of justice. This is a book about the theory of the city or commonwealth, what would come to be called the state, in early modern natural law discourse. Annabel Brett takes a fresh approach by looking at this political entity from the perspective of its boundaries and those who crossed them. She begins with a classic debate from the Spanish sixteenth century over the political treatment of mendicants, showing how cosmopolitan ideals of porous boundaries could simultaneously justify the freedoms of itinerant (...) beggars and the activities of European colonists in the Indies. She goes on to examine the boundaries of the state in multiple senses, including the fundamental barrier between human beings and animals and the limits of the state in the face of the natural lives of its subjects, as well as territorial frontiers. Drawing on a wide range of authors, Brett reveals how early modern political space was constructed from a complex dynamic of inclusion and exclusion. Throughout, she shows that early modern debates about political boundaries displayed unheralded creativity and virtuosity but were nevertheless vulnerable to innumerable paradoxes, contradictions, and loose ends.Changes of State is a major work of intellectual history that resonates with modern debates about globalization and the transformation of the nation-state.
