Pancreatitis is a complex, progressively destructive inflammatory disorder. Alcohol was long thought to be the primary causative agent, but genetic contributions have been of interest since the discovery that rare PRSS1, CFTR and SPINK1 variants were associated with pancreatitis risk. We now report two associations at genome-wide significance identified and replicated at PRSS1-PRSS2 and X-linked CLDN2 through a two-stage genome-wide study. The PRSS1 variant likely affects disease susceptibility by altering expression of the primary trypsinogen gene. The CLDN2 risk allele is (...) associated with atypical localization of claudin-2 in pancreatic acinar cells. The homozygous CLDN2 genotype confers the greatest risk, and its alleles interact with alcohol consumption to amplify risk. How, if at all, is knowing how to do something defeasible? Some, the “intellectualists”, treat the defeasibility of knowledge-how as in some way derivative on the defeasibility of knowledge-that. According to a recent proposal by Carter and Navarro (Philos Phenomenol Res 3:662–685, 2017), knowledge-how defeat cannot be explained in terms of knowledge-that defeat; instead, knowledge-how defeat merits and entirely separate treatment. The thought behind “separatism” is easy to articulate. Assuming that knowledge of any kind is defeasible, since knowledge-that and knowledge-how (...) are fundamentally different beasts, the best accounts of their defeasibility must reflect the underlying differences between them. I reject a separatist treatment of the defeasibility of knowledge-that and knowledge-how. One can acknowledge certain important differences between knowledge-that and knowledge-how without thereby needing two theories of defeasibility. Ultimately, though, I’ll argue that the defeasibility of basic knowledge-how, unlike non-basic knowledge-how, is a much more difficult issue than has been acknowledged. (shrink)

Successful formulation and implementation of end-of-life care requires ongoing communication with the patient. When patients, for reasons of general medical or psychiatric illness, fail to verbally communicate, providers must be receptive to messages conveyed through alternate avenues of communication. We present the narrative of a man with schizophrenia who wished to forgo hemodialysis as a study in the ethical importance of attention to nonverbal communication. It is tempting to think (though many deny) that epistemic agents exercise a distinctive kind of control over their belief‐like attitudes. My aim here is to sketch a “bottom‐up” model of epistemic agency, one that draws on an analogous model of practical agency, according to which an agent's conditional beliefs are reasons‐responsive planning states that initiate and sustain mental behavior so as to render controlled.

The general conditions of epistemic defeat are naturally represented through the interplay of two distinct kinds of entailment, deductive and defeasible. Many of the current approaches to modeling defeasible reasoning seek to define defeasible entailment via model-theoretic notions like truth and satisfiability, which, I argue, fails to capture this fundamental distinction between truthpreserving and justification-preserving entailments. I present an alternative account of defeasible entailment and show how logic programming offers a paradigm in which the distinction can be captured, allowing for (...) the modeling of a larger range of types of defeat. This is possible through a natural extension of the declarative and procedural semantics of Horn clauses. (shrink)

The distinction between the modeling of information and the modeling of data in the creation of automated systems has historically been important because the development tools available to programmers have been wedded to machine oriented data types and processes. However, advances in software engineering, particularly the move toward data abstraction in software design, allow activities reasonably described as information modeling to be performed in the software creation process. An examination of the evolution of programming languages and development of general programming (...) paradigms, including object-oriented design and implementation, suggests that while data modeling will necessarily continue to be a programmer's concern, more and more of the programming process itself is coming to be characterized by information modeling activities. The Cas9 endonuclease is the central component of the Type II CRISPR/Cas system, a prokaryotic adaptive restriction system against invading nucleic acids, such as those originating from bacteriophages and plasmids. Recently, this RNA‐directed DNA endonuclease has been harnessed to target DNA sequences of interest. Here, we review the development of Cas9 as an important tool to not only edit the genomes of a number of different prokaryotic and eukaryotic species, but also as an efficient system for site‐specific transcriptional repression or (...) activation. Additionally, a specific Cas9 protein has been observed to target an RNA substrate, suggesting that Cas9 may have the ability to be programmed to target RNA as well. Cas proteins from other CRISPR/Cas subtypes may also be exploited in this regard. Thus, CRISPR/Cas systems represent an effective and versatile biotechnological tool, which will have significant impact on future advancements in genome engineering. (shrink)

In this paper I attempt to cast the current program verification debate within a more general perspective on the methodologies and goals of computer science. I show, first, how any method involved in demonstrating the correctness of a physically executing computer program, whether by testing or formal verification, involves reasoning that is defeasible in nature. Then, through a delineation of the senses in which programs can be run as tests, I show that the activities of testing and formal verification do (...) not necessarily share the same goals and thus do not always constitute alternatives. The testing of a program is not always intended to demonstrate a program's correctness. Testing may seek to accept or reject nonprograms including algorithms, specifications, and hypotheses regarding phenomena. The relationship between these kinds of testing and formal verification is couched in a more fundamental relationship between two views of computer science, one properly containing the other. Speakers differ from one another in philosophically problematic ways. Two speakers can vary not simply with respect to what they believe, but also in the ways they speak, the concepts they employ, and the standards they bring to bear. The fact of imperfect convergence gives rise to a wide range of philosophical puzzles, largely via a single generalization: If two speakers disagree with each other, then at least one of them says something false. The generalization is plausible, but mistaken. Counterexamples (...) are common, diverse, and thoroughly entrenched in ordinary talk, scientific discourse, and philosophical inquiry. I focus on a particular family of counterexamples, disagreements about the proper application of linguistic items, or what I call metalinguistic disagreements. Coming to grips with the widespread existence of metalinguistic disagreement requires a nuanced account of the ways in which information can be transmitted via an utterance, and it suggests a substantial rethinking of conventional philosophical wisdom about the nature of meaning. I begin with a philosophical case study in metalinguistic disagreement—disagreements about aesthetics. I go on to explore the more general linguistic properties of metalinguistic disagreements, focusing on their striking failure to license so-called metalinguistic negation. I conclude with a consideration of semantic methodology, arguing that, despite widely held assumptions to the contrary, metalinguistic disagreements can be adjudicated with reference to objective, non-linguistic features of the world. It is possible therefore to roundly reject a pernicious relativism about scientific discourse while simultaneously allowing for widespread, interpersonal variation in meaning. (shrink)

Heuristics can be regarded as justifying the actions and beliefs of problem-solving agents. I use an analysis of heuristics to argue that a symbiotic relationship exists between traditional epistemology and contemporary artificial intelligence. On one hand, the study of models of problem-solving agents usingquantitative heuristics, for example computer programs, can reveal insight into the understanding of human patterns of epistemic justification by evaluating these models'' performance against human problem-solving. On the other hand,qualitative heuristics embody the justifying ability of defeasible rules, (...) the understanding of which is provided by traditional epistemology. (shrink)

This article critically examines recent work on free will and moral responsibility by Randolph Clarke, Robert Kane, and Timothy O’Connor in an attempt to clarify issues about control and luck that are central to the debate between libertarians (agent causationists and others) and their critics. It is argued that luck poses an as yet unresolved problem for libertarians.

Previous studies conducted in the United States indicate that people associate numbers with gender, such that odd numbers are more likely to be considered male and even numbers considered female. It has been argued that this number gendering phenomenon is acquired through social learning and conditioning, and that male-odd/female-even associations reflect a general, cross-cultural human consensus on gender roles relating to agency and communion. However, the incidence and pattern of number gendering in cultures outside the United States remains to be (...) established. Against this background, the purpose of this study was to determine whether people from a culture and country very different from the United States also associate numbers with gender, and, if they do, whether the pattern of these associations is the male-odd/female-even associations previously observed. To investigate this issue, we adopted the Implicit Association Test used frequently in previous research, where associations between numbers and gender were examined using male and female Arabic participants native to, and resident in, the United Arab Emirates. The findings indicated that the association of numbers with gender does occur in Arabic culture. But while Arabic females associated odd numbers with male faces and even numbers with female faces, Arabic males showed the reversed pattern of gender associations, associating even numbers with male faces and odd numbers with female faces. These findings support the view that number gendering is indeed a cross-cultural phenomenon and show that the phenomenon occurs across very different countries and cultures. But the findings also suggest that the pattern with which numbers are associated with gender is not universal and, instead, reflects culture-specific views on gender roles which may change across cultures and gender. Further implications for understanding the association of numbers with gender across human societies are discussed. (shrink)

The socially responsible investment industry (SRI) is slowly changing from a screening, avoidance paradigm to a comprehensive paradigm that seeks to affect corporate behavior. Credible rating systems are a key component of this sea change. Reliable and recognizable social and environmental metrics are critical to this progress. The Total Social Impact (TSI) rating approach is a new social metric scheme based on a comprehensive rating of stakeholder issues. Philoso- phers traditionally focus on their disagreements with one another. My reply is squarely within that tradition, as are the articles to which I am replying.

The author argues that philosophers' attempts to use philosophy of language to solve problems of jurisprudence have often failed- the most dramatic failure being that of Jeremy Bentham. H.L.A.Hart made some related mistakes in his creative use of philosophy of language, yet his focus on language still yields some very significant insights for jurisprudence: the context principle (that the correct application of linguistic expressions typically depends on context in ways that are important for jurisprudence), the diversity principle (that grounds of (...) correct application of legal language may be diverse), and the importance of vagueness. New targets for brain tumor therapies may be identified by mutations that cause hereditary microcephaly. Brain growth depends on the repeated proliferation of stem and progenitor cells. Microcephaly syndromes result from mutations that specifically impair the ability of brain progenitor or stem cells to proliferate, by inducing either premature differentiation or apoptosis. Brain tumors that derive from brain progenitor or stem cells may share many of the specific requirements of their cells of origin. These tumors may therefore be susceptible to (...) disruptions of the protein products of genes that are mutated in microcephaly. The potential for the products of microcephaly genes to be therapeutic targets in brain tumors are highlighted hereby reviewing research on EG5, KIF14, ASPM, CDK6, and ATR. Treatments that disrupt these proteins may open new avenues for brain tumor therapy that have increased efficacy and decreased toxicity. -/- . (shrink)

What can a philosophical analysis of the concept of interpretation contribute to legal theory? In his recent book,Interpretation and Legal Theory, Andrei Marmor proposes a complex and ambitious analysis as groundwork for his positivist assault on “interpretive” theories of law and of language. I argue (i) that the crucial element in Marmor's analysis of interpretation is his treatment of Ludwig Wittgenstein's remarks on following rules, and (ii) that a less ambitious analysis of interpretation than Marmor's can take better advantage of (...) those insights about rules. I explore some implications of such an analysis for the role of interpretation in legal reasoning. (shrink)
