Drug design methods have made significant new advances over the last ten years, mainly in the areas of molecular modelling. In more recent times important developments in theory have led to a different type of modelling becoming possible, the so‐called de novo or automated design algorithms. In this new method the programs perform much of the chemist's thinking, in finding appropriately sized chemical groups to fit into a target site. However this is a combinatoric problem which has no general analytical (...) solution; it is ripe for optimization. Other advances, such as combinatorial chemical synthesis and screening, will dramatically influence the search for new lead structures for target sites, which at present are poorly understood. Already these methods are being applied to peptide libraries. Peptides do not make good drug compounds because of their poor bioavailability; further, their flexibility reduces their affinity. In some cases peptide backbones can be removed and replaced with rigid non‐peptide scaffolds. (shrink)

Philip James Jones, a Fellow of the British Academy, was one of the most distinguished, complex, and challenging of medieval historians. His works on the Italian city-states of the thirteenth to fifteenth centuries and on Italy's agrarian history are monuments built to last, benchmarks that defined the field for a generation. Jones was elected a Fellow of the British Academy in 1984 and was awarded the Serena Medal for Italian studies in 1988. He won a major open scholarship in (...) Modern History at Wadham College, University of Oxford. Jones took a First in Modern History in 1945 and was appointed to a research studentship at Magdalen College. Young people have an enormous stake in the present and future state of Earth. Almost half of the human population is under the age of 25. If young people’s resources of energy, time, and knowledge are misdirected towards violence, terrorism, socially-isolating technologies, and unsustainable consumption, civilization risks destabilization. In general, it would seem that a genuinely " empirical" approach would seek to engage the actual truth claims of religious com· munities on their own terms-even when those claims conflict with historicist suppositions. Second, in so far as Dean thinks there (...) is a uniquely American reli· gious experience or style of thought, privileging that style seems a dangerous move indeed. Dean is not unaware of the dangers. He specifically denies that he is engaging in any form of " American exceptionalism " (p. 25) ; it is simply that an " appropriately American historicism, centering on American culture, would have a greater chance of suggesting practices that work for and are true for America " (p. 24). But I am skeptical that such disclaimers address the real problem. Near the very end of the book, Dean suggests that a modi· fied naturalism might lead to " a notion of God that was more genuinely American than anything earlier," culminating in the recognition that " American thought about God is really the thought of a national com· munity" (p. 144). Descriptively that may well be the case; but then the Christian theologian must seek norms elsewhere. History Making History does not show us how historicist insigh~ might be incorporated into ecumenical, trinitarian theology, but that does not mean the attempt will prove unfruitful. Sources of Catholic Teaching is a compendium of recent "official" statements by Roman Catholic ecclesiastical bodies on issues concern· ing medical ethics. Arranged alphabetically according to topics, this col- 546 BOOK REVIEWS lection of statements from the papacy, the Roman Congregations, and the conferences of Catholic bishops could prove to he a very helpful reference tool. It contains a brief introduction which seeks to explain the role of these statements in the Catholic moral theology and Catholic medical ethics, a table of contents of the statements included, and then the statements themselves. In most instances it does not contain the entire document or statement hut only excerpts therefrom. There are some weaknesses in this work. First, these statements are not self-explanatory; they are in need of interpretation and explanation in many instances. But the authors present only the bald text and do not offer any explanatory comments. They apparently presume that these statements are pellucid to the reader, hut this is an unwarranted pre· sumption. For example, clarifying comments should have been added to the statement from the Declaration on Euthanasia because the allocution of Pius XII about giving lethal doses of analgesia is not entirely clear. A second difficulty with this work is the brief and somewhat insubstantial introduction on conscience and the role these statements should play in the formation of conscience. This sort of work invites a substantial and extended commentary on the nature of moral judgments, the relationship of conscience to the natural law, and the authority of these statements. The authors note that not all of the official statements issued by various organs of the Church are of equal authority, hut they do not try to identify the specific authoritative status of the various statements they include in their work. The reader is thus left to his or her own devices to determine, for example, the relative weight of statements of the administrative hoard of the NCCB versus statements of state episcopal conferences. A still further problem is that some of the statements are of virtually no significant authority for Catholics. For example, the statement of the administrative hoard of the NCCB on AIDS is of questionable... (shrink)

This paper argues that the language of rights cannot express distinctively ecofeminist insights into the treatment of nonhuman animals and the environment. An alternative is proposed in the form of a politicized ecological ethic of care which can express ecofeminist insights. The paper concludes with consideration of an ecofeminist moral issue: how we choose to understand ourselves morally in relation to what we are willing to count as food. "Contextual moral vegetarianism" represents a response to a politicized ecological ethic of (...) care. (shrink)

Conscious states—state that there is something it is like to be in—seem both rich or full of detail and ineffable or hard to fully describe or recall. The problem of ineffability, in particular, is a longstanding issue in philosophy that partly motivates the explanatory gap: the belief that consciousness cannot be reduced to underlying physical processes. Here, we provide an information theoretic dynamical systems perspective on the richness and ineffability of consciousness. In our framework, the richness of conscious experience corresponds (...) to the amount of information in a conscious state and ineffability corresponds to the amount of information lost at different stages of processing. We describe how attractor dynamics in working memory would induce impoverished recollections of our original experiences, how the discrete symbolic nature of language is insufficient for describing the rich and high-dimensional structure of experiences, and how similarity in the cognitive function of two individuals relates to improved communicability of their experiences to each other. While our model may not settle all questions relating to the explanatory gap, it makes progress toward a fully physicalist explanation of the richness and ineffability of conscious experience—two important aspects that seem to be part of what makes qualitative character so puzzling. Philosophy has often been criticized for privileging the abstract; this volume attempts to remedy that situation. Focusing on one of the most concrete of human concerns, food, the editors argue for the existence of a philosophy of food.

In this paper I draw on Thaddeus Metz’s pioneering work in African ethics, and particularly his account of the concept described by the terms ubuntu, botho, hunhu or utu, to sketch an African normative understanding of the act of rebellion against the authority of the state. Most commonly articulated in the phrase “a person is a person through other persons”, ubuntu is interpreted by Metz as a unique communitarian moral principle which can be described in its essence as the claim (...) that “actions are right, or confer ubuntu on a person, insofar as they prize communal relationships, ones in which people identify with each other, or share a way of life, and exhibit solidarity toward one another, or care about each other’s quality of life”. On the face of it, this principle appears at odds with rebellions against state authority. Following Metz, I argue, however, that a deeper grasp of this principle does, in fact, provide a justification for instances of civilian rebellion against state authority, under appropriate circumstances. Predictive processing accounts of neural function view the brain as a kind of prediction machine that forms models of its environment in order to anticipate the upcoming stream of sensory stimulation. These models are then continuously updated in light of incoming error signals. Predictive processing has offered a powerful new perspective on cognition, action, and perception. In this chapter we apply the insights from predictive processing to the study of emotions. The upshot is a picture of emotion as inseparable from (...) perception and cognition, and a key feature of the embodied mind. (shrink)

A core project for deep ecologists is the reformulation of the concept of self. In searching for a more inclusive understanding of self, deep ecologists often look to Buddhist philosophy, and to the Japanese Buddhist philosopher Dōgen in particular, for inspiration. I argue that, while Dōgen does share a nondualist, nonanthropocentric framework with deep ecology, his phenomenology of the self is fundamentally at odds with the expanded Self found in the deep ecology literature. I suggest, though I do not fully (...) argue for it, that Dōgen’s account of the self is more sympathetic to one version of ecofeminism than to deep ecology. (shrink)

In this work, the prominent political philosopher Philip Pettit embarks on a massive undertaking to offers major new accounts of the foundations of the state and the nature of justice. In doing so Pettit builds a new theory of what the state is and what it ought to be, addresses the normative question of how justice serves as a measure of the success of a state, and the way it should operate in relation to its citizens and other people.

This paper explores the role of moral uncertainty in explaining the morally disruptive character of new technologies. We argue that existing accounts of technomoral change do not fully explain its disruptiveness. This explanatory gap can be bridged by examining the epistemic dimensions of technomoral change, focusing on moral uncertainty and inquiry. To develop this account, we examine three historical cases: the introduction of the early pregnancy test, the contraception pill, and brain death. The resulting account highlights what we call “differential (...) disruption” and provides a resource for fields such as technology assessment, ethics of technology, and responsible innovation. Over twenty years of research have examined the cognitive consequences of positive affect states, and suggested that positive affect leads to a broadening of cognition (see review by Fredrickson, 2001). However, this research has primarily examined positive affect that is low in approach motivational intensity (e.g., contentment). More recently, we have systematically examined positive affect that varies in approach motivational intensity, and found that positive affect high in approach motivation (e.g., desire) narrows cognition, whereas positive affect low in approach motivation (...) broadens cognition (e.g., Gable & Harmon-Jones, Citation2008a; Harmon-Jones & Gable, 2009). In this article we will review past models and present a motivational dimension model of affect that expands understanding of how affective states influence attentional and cognitive breadth. We then review research that has varied the motivational intensity of positive and negative affect and found that affect of low motivational intensity broadens cognitive processes, whereas affect of high motivational intensity narrows cognitive processes. Predictive processing has begun to offer new insights into the nature of conscious experience—but the link is not straightforward. A wide variety of systems may be described as predictive machines, raising the question: what differentiates those for which it makes sense to talk about conscious experience? One possible answer lies in the involvement of a higher-order form of prediction error, termed expected free energy. In this paper we explore under what conditions the minimization of this new quantity might underpin conscious (...) experience. Our suggestion is that the minimisation of Expected Free Energy is not in itself sufficient for the occurrence of conscious experience. Instead, it is relevant only insofar as it helps deliver what Ward et al. ( 2011 ) have previously described as a _sense of our own poise over an action space_. Perceptual experience, we will argue, is nothing other than the process that puts current actions in contact with goals and intentions, enabling some creatures to know the space of options that their current situation makes available. This proposal fits with recent work suggesting a deep link between conscious contents and contents computed at an ‘intermediate’ level of processing, apt for controlling action. (shrink)

Violence can be physical and psychological. It can characterize personal actions, forms of group activity, and abiding social and political policy. This book includes all of these aspects within its focus on institutional forms of violence. Institution is also a broad category, ranging from formal arrangements such as the military, the criminal code, the death penalty and prison system, to more amorphous but systemic situations indicated by parenting, poverty, sexism, work, and racism. Violence is as complex as the human beings (...) who resort to it; its institutional forms pervade our relational lives. We are all participants in it as victims and perpetrators. The chapters in this book were written in the hope that violence can be explicated, even if not fully understood, and that such clarification can help us in devising less violent forms of living, even if it does not lead to its total abolition. This paper offers a theological critique of the future of ‘nature’ as suggested by New Biology, including recent developments in genetic engineering. It explores the biblical basis for grounding a theology of creation in the wisdom motif. The relationship between wisdom and creation in the Old Testament is discussed. The link between wisdom, Christ and the Holy Spirit is suggestive of wisdom's involvement in re‐creation as well as initial creation. An argument is put forward for a Trinitarian basis for wisdom. (...) The relationship between wisdom and apocalyptic literature gives a clue as to how wisdom might contribute to theological reflection on the future. Wisdom as metaphor is used to construct a new future of science. By reformulating the future of creation in the light of wisdom a future of science comes into view that meets the postmodern requirement for adaptability and diversity, but without forgetting the idea of distinction between humanity and the natural world. The long tradition of wisdom brings both a rootedness in historical perspectives and dynamic flexibility that serves to inform the relationship between God and the natural world. A measure of stability is a requirement in shaping perspectives for the future, particularly if the ambivalence and anxiety associated with new explorations in science are to be met. (shrink)
