Structural biology has experienced several transformative technological advances in recent years. These include: development of extremely bright X-ray sources and the use of electrons to extend protein crystallography to ever decreasing crystal sizes; and an increase in the resolution attainable by cryo-electron microscopy. Here we discuss the use of these techniques in general terms and highlight their application for biological filament systems, an area that is severely underrepresented in atomic resolution structures. We assemble a model of a capped tropomyosin-actin minifilament (...) to demonstrate the utility of combining structures determined by different techniques. Finally, we survey the methods that attempt to transform high resolution structural biology into more physiological environments, such as the cell. Together these techniques promise a compelling decade for structural biology and, more importantly, they will provide exciting discoveries in understanding the designs and purposes of biological machines. Structural biology is undergoing technological advancements that now reveal the structures of previously inaccessible biological machines. Microfocus synchrotron beamlines, X-ray free electron lasers, and MicroED cope with vanishingly small crystals, while cryo-electron microscopy targets single molecules at near atomic resolution. These advances promise an exciting decade for structural biology. (shrink)

I address a question in moral metaphysics: How are conflicts between moral obligations possible? I begin by explaining why we cannot give a satisfactory answer to this question simply by positing that such conflicts are conflicts between rules, principles, or reasons. I then develop and defend the “Dispositional Account,” which posits that conflicts between moral obligations are conflicts between the manifestations of obligating dispositions (obligating powers, capacities, etc.), just as conflicts between physical forces are conflicts between the manifestations of (certain) (...) causal dispositions (causal powers, capacities, etc.). This account combines the so-called “moral forces” interpretation of prima facie obligations with a dispositional moral metaphysic according to which the metaphysical grounds of moral obligations are not rules or laws, but rather real, irreducibly dispositional properties (or powers) of moral agents and patients. My principal aims are to offer a theoretically attractive and suitably metaphysical account of conflicts of obligation, and to show that the dispositional moral metaphysic that grounds the Dispositional Account can explain and accommodate plausible normative views that rule- and law-based alternatives cannot, as well as to answer objections that have been pressed against other accounts of moral conflict (especially Ross’s) that appeal to moral dispositions or forces. This book gives a descriptive analysis of specific Madhyamika texts. It compares the ideology of Kumarajiva (a translator of the four Madhyamika treatises 400 A.D.) with the ideologies of the three Chinese contemporaries - HuiYuan, Seng-Jui and Seng-Chao. It envisages an intercultural transmission of religious and philosophical ideas from India to China.

Credences are beliefs about evidential probabilities. We give the view an assessment-sensitive formulation, show how it evades the standard objections, and give several arguments in support.

A science of metaphysics adhering to Immanuel Kant's critical demands as set forth in his "Critique of Pure Reason", and "Prolegomena to Any Future Metaphysic...." The work includes an Appendix that quotes Kant's most relevant remarks in this regard, along with his criterion for objective validity that, given the technical jargon, can be next to impossible to interpret even for those most familiar with Kant. The Appendix allows Kant to interpret himself, the point being that many secondary works enter into (...) conflict with Kant. This can be seen most notably with respect to the somewhat understandable notion that Kant was opposed to speculative metaphysics; but Kant's criterion for objective validity undermines this closed-to-speculative-metaphysics interpretation. As a science in the Kantian sense this work (in this latest edition there is a 2/3 page abstract diagram of the science that makes it more conceptually clear) offers an opportunity for those inclinec to shy away from speculative metaphysics to reexamine their interpretations, for as yet there is just as Kant states, no such thing as metaphysics, aside from such a science (Hegel notwithstanding). The objectives are as Kant mentions in the introduction to his CPR: God, Freedom, and Immortality; and that these problems can be dealt with rationally can be seen in this work; and without fear of violating Kant's reasonable and justifiable strictures, the main one being that where anything offered in the name of pure reason is concerned, it must be such that it helps us to make sense of our phenomenal world. And as to Kant's prerequisite test: "How are synthetic cognitions, a priori possible?" I refer only to my essay "Beyond Kant and Hegel (In Answer to the Question ...)" ... in "The Review of Metaphysics", March 2013. The premise advanced in the essay may be seen to be fully apparent in this science of metaphysics. (shrink)

Ludwig Wittgenstein is perhaps the greatest philosopher of the twentieth century, and certainly one of the most original in the entire Western tradition. Given the inaccessibility of his work, it is remarkable that he has inspired poems, paintings, films, musical compositions, titles of books -- and even novels. In his splendid biography, Ray Monk has made this very compelling human being come alive in a way that perfectly explains the fascination he has evoked. Wittgenstein's life was one of great moral (...) and spiritual depth. Although his work addresses problems of logic, language, mind, and knowledge, he often described it as having an ethical point, and once said that he "could not help seeing every problem from a religious point of view." In successfully bridging the gap assumed by many to exist between Wittgenstein's life and his work, Monk's biography helps to explain the interest in Wittgenstein's later philosophy on the part of Buddhist scholars. The purpose of this article is to discuss an external whistleblowing event that occurred after all internal whistleblowing through the hierarchy of the organization had failed. It is argued that an organization that does not support those that whistle blow because of violation of professional standards is indicative of a failure of organizational ethics. Several ways to build an ethics infrastructure that could reduce the need to resort to external whistleblowing are discussed. A relational ethics approach is presented as a (...) way to eliminate the negative consequences of whistleblowing by fostering an interdependent moral community to address ethical concerns. (shrink)

Examining one of the fundamental issues in cognitive psychology: How does our conscious experience come to be the way it is?

Where much contemporary philosophy seeks to stave off the "threat" of nihilism by safeguarding the experience of meaning--characterized as the defining feature of human existence--from the Enlightenment logic of disenchantment, this book attempts to push nihilism to its ultimate conclusion by forging a link between revisionary naturalism in Anglo-American philosophy and anti-phenomenological realism in recent French philosophy. Contrary to an emerging "post-analytic" consensus which would bridge the analytic-continental divide by uniting Heidegger and Wittgenstein against the twin perils of scientism and (...) skepticism, this book short-circuits both traditions by plugging eliminative materialism directly into speculative realism. We provide an overview of a proposal for a new metaphysical framework within which theology and science might both find a home. Our proposal draws on the triadic semiotics and threefold system of metaphysical categories of C. S. Peirce. We summarize the key features of a semiotic model of the Trinity, based on observed parallels between Peirce's categories of Firstness, Secondness, and Thirdness and Christian thinking about, respectively, the Father, Son, and Holy Spirit. We test and extend the semiotic model (...) by exploring how Peirce's taxonomy of signs offers a new approach to theological reflection on the Incarnation. This leads to a novel way of framing scientific questions about human evolution in semiotic terms and to a new approach to theological anthropology. Finally, we use the semiotic model of the Trinity as the basis of a trinitarian approach to the theology of creation according to which the semiotic processes that are fundamental to life and to human behavior and cognition may be understood as “vestiges of the Trinity in creation.”. Virtues are acquirable, so if intellectual humility is a virtue, it’s acquirable. But there is something deeply problematic—perhaps even paradoxical—about aiming to be intellectually humble. Drawing on Edward Slingerland’s analysis of the paradoxical virtue of wu-wei in Trying Not To Try (New York: Crown, 2014), we argue for an anti-individualistic conception of the trait, concluding that one’s intellectual humility depends upon the intellectual humility of others. Slingerland defines wu-wei as the “dynamic, effortless, and unselfconscious state of mind of a person (...) who is optimally active and effective” (Trying Not to Try, 7). Someone who embodies wu-wei inspires implicit trust, so it is beneficial to appear wu-wei. This has led to an arms race between faking wu-wei on the one hand and detecting fakery on the other. Likewise, there are many benefits to being (or seeming to be) intellectually humble. But someone who makes conscious, strategic efforts to appear intellectually humble is ipso facto not intellectually humble. Following Slingerland’s lead, we argue that there are several strategies one might pursue to acquire genuine intellectual humility, and all of these involve commitment to shared social or epistemic values, combined with receptivity to feedback from others, who must in turn have and manifest relevant intellectual virtues. In other words, other people and shared values are partial bearers of a given individual’s intellectual humility. If this is on the right track, then acquiring intellectual humility demands epistemic anti-individualism. (shrink)

Evolutionary scepticism holds that the evolutionary account of the origins of the human cognitive apparatus has sceptical implications for at least some of our beliefs. A common target of evolutionary scepticism is moral realism. Scientific realism, on the other hand, is much less frequently targeted, though the idea that evolutionary theory should make us distrustful of science is by no means absent from the literature. This line of thought has received unduly little attention. I propose to remedy this by advancing (...) what I will call an evolutionary sceptical challenge to scientific realism. I argue that, given standard evolutionary theory, our possession of sound innate metaphysical intuitions would have taken an epistemically problematic ‘lucky accident’. This, as I will show, entails that scientific realism is a self-undermining position. I discuss objections to my argument’s two premises, including ones that appeal to the success of the sciences and to the possibility that sound innate metaphysical intuitions evolved as an evolutionary ‘by-product’. I then draw out an advantage of my argument over a similar one recently put forward by Graber and Golemon. I finish by submitting that scientific realism, given the soundness of my argument, is faced with a new ‘Darwinian Dilemma’, and briefly address the significance of this for the debate between realists and anti-realists in the philosophy of science. For the Intentionalist, utterance content is wholly determined by a speaker’s meaning-intentions; the sentence uttered serves merely to facilitate the audience’s recovering these intentions. We argue that Intentionalists ought to be Particularists, holding that the only “principles” of meaning recovery needed are those governing inferences to the best explanation; “principles” that are both defeasible and, in a sense to be elaborated, variable. We discuss some ways in which some theorists have erred in trying to tame the “wild west” of pragmatics (...) and context-sensitivity -- including recent work that makes essential appeal to the information structure of a discourse -- and in so doing, offer a general recipe for defending the Particularist picture of utterance content and its recovery that we favor. According to proponents of the face-value account, a beliefreport of the form ‘S believes that p’ is true just in case the agentbelieves a proposition referred to by the that-clause. As againstthis familiar view, I argue that there are cases of true beliefreports of the relevant form in which there is no proposition that thethat-clause, or the speaker using the that-clause, can plausibly betaken as referring to. Moreover, I argue that given the distinctiveway in which the face-value theory of belief-reports (...) fails, there ispressure to give up the metaphysical thesis that belief is apropositional attitude. I conclude by suggesting that we allownon-propositional entities to be amongst the relata of thebelief-relation, and make some speculative remarks concerning whatsuch entities might be like. Relying on inference to the best explanation requires one to hold the intuition that the world is ‘intelligible’, that is, such that states of affairs at least generally have explanations for their obtaining. I argue that metaphysical naturalists are rationally required to withhold this intuition, unless they cease to be naturalists. This is because all plausible naturalistic aetiologies of the intuition entail that the intuition and the state of affairs which it represents are not causally connected in an epistemically appropriate (...) way. Given that one ought to rely on IBE, naturalists are forced to pick the latter and change their world-view. Traditional theists, in contrast, do not face this predicament. This, I argue, is strong grounds for preferring traditional theism to naturalism. (shrink)

I argue that if intentions are what Grice, and most contemporary action theorists, take them to be, they are inessential for acts of speaker meaning. More specifically, my primary aim is to show that the consensus view of speaker meaning is in deep tension with certain plausible, and widely accepted, cognitive constraints on rational intention pertaining to an agent’s assessment of her prospects of achieving her goal. My secondary aim is to offer an initial case for thinking that the best (...) way to resolve this tension is to give up the claim that meaning is fundamentally a matter of intention. In pursuing this strategy, we will see some of the potential benefits of Bratman’s (1987) work on intentions, plans, and practical reasoning for the theory of meaning, including some interesting consequences of his rejection of the so-called ‘Simple View’ on which we intentionally do only those things that we specifically intend to do. Moreover, our discussion will help to illustrate how some familiar tools from decision theory might find new use in discussions of meaning and communication. (shrink)

According to the most popular versions of the flicker defense, Frankfurt-style cases fail to undermine the Principle of Alternative Possibilities (PAP) because agents in these cases are (directly) morally responsible not for making the decisions they make but for making these decisions on their own, which is something they could have avoided doing. Frankfurt defenders have primarily focused on trying to show that the alternative possibility of refraining from making the relevant decisions on their own is not a robust alternative, (...) while generally granting that this alternative cannot easily be eliminated from successful cases of this sort. In a recent issue of this journal, Stockdale (2022) attempts to sidestep the debate concerning robustness and develops a novel kind of Frankfurt-style case in which agents are unable to avoid making the relevant decisions on their own. The fundamental problem with Stockdale’s argument is that it hinges on an implausible conception of acting on one’s own. I help clarify the pertinent sense of what it means to do a thing on one’s own in this context and show that these new cases are unable to overcome the targeted versions of the flicker defense of PAP. One of the standard approaches to the metaphysics of personal identity has some counter-intuitive ethical consequences when combined with maximising consequentialism and a plausible doctrine about aggregation of consequences. This metaphysical doctrine is the so-called ‘multiple occupancy’ approach to puzzles about fission and fusion. It gives rise to a new version of the ‘utility monster’ problem, particularly difficult problems about infinite utility, and a new version of a Parfit-style ‘repugnant conclusion’. While the article focuses on maximising consequentialism for simplicity, the (...) problems demonstrated apply more widely to a range of ethical views, especially flavours of consequentialism. This article demonstrates how these problems arise, and discusses a number of options available in the light of these problems for a consequentialist tempted by a multiple occupancy metaphysics. Your belief that Obama is a Democrat would not be the belief that it is if it did not represent Obama, nor would the pain in your ankle be the state that it is if, say, it felt like an itch. Accordingly, it is tempting to hold that phenomenal and representational properties are essential to the mental states that have them. But, as several theorists have forcefully argued (including Kripke (1980) and Burge (1979, 1982)) this attractive idea is seemingly in (...) tension with another equally attractive thesis, namely, the token-identity thesis; the thesis according to which every mental state token is identical with some or other token physical state. In this paper, we show that these seemingly incontrovertible essentialist intuitions are in fact compatible with “token physicalism” regarding the mental. Given a suitably plentitudinous ontology of objects, we argue that there are physical things with which our token mental states can be identified. This is preferable to existing views that give up the essentiality claims or simply reject the token-identity thesis. (shrink)

One approach to validating simulation models is to formally compare model outputs with independent data. We consider such model validation from the point of view of Frequentist statistics. A range of estimates and tests of goodness of fit have been advanced. We review these approaches, and demonstrate that some of the tests suffer from difficulties in interpretation because they rely on the null hypothesisHypothesis that the model is similar to the observationsObservations. This reliance creates two unpleasant possibilities, namely, a model (...) could be spuriously validated when dataData are too few, or inappropriately rejected when data are too many. Finally, these tests do not allow a principled declaration of what a reasonable level of difference would be considering the purposes to which the model will be put. We consider equivalence tests, and demonstrate that they do not suffer from the previously identified shortcomings. We provide two case studies to illustrate the claims of the chapter. (shrink)
